add the buffer when you are done3. Connect cables and run the gel at 125 volts

for 15 minutes. Note the starting time.4. While you wait, make observations and lab

notes. (Note any dye spills or over flow. Did your pipettor tip puncture the gel? Can you see any thing happen in the box?)

5. While you wait, start writing the answers to the questions in complete sentences.

Conclusions—Write these on the back of your DNA lab!1. Which dye is the smallest? Largest? Which dyes

are positive? Negative?2. What dyes make up the unknown (X sample)?3. Compare the relative size of one of the positively

charged dyes, to one of the negatively charged dyes. Identify dyes in this comparison by number and color. Use the side (+ or -) the dye move towards, and measure the distance from the center for this analysis.

Protein Synthesis

To get from a DNA gene to a protein, the cell has to perform protein synthesis.

It requires 3 types of RNA: mRNA rRNA tRNA

It happens in 2 parts: Transcription Translation

Translation

Translation is the process of making an amino acid chain from the instructions coded in the mRNA. Occurs in cytoplasm.

1. mRNA binds to the ribosome & the ribosome begins reading at the START codon AUG

3. Each tRNA that enters the ribosome adds an amino acid to the growing chain. A peptide (protein) bond is formed between each amino acid in the chain.

There are 20 amino acids Each tRNA can only carry ONE type

of amino acid

3. When a STOP codon (UAA, UAG, or UGA) on mRNA enters the ribosome, the large & small rRNA subunits fall apart and translation ends
