TITLE: Applying the Principles of the Tesla Engine to Design and Construct a Prototype of a Bladeless Wind Turbine

Traditionalwindturbinesareextremelyinefficientandcauseenvironmentalproblemsbecausetheykillbirdsandoftengeneratenoisethatdisturbsnearbyresidents.In1913,NikolaTeslainventedtheTeslaenginethatachievedanastonishing60%efficiency.Withmoderndayimprovementsinmaterialsandtechnologyhigherefficienciescanbeobtained.ThisprojectexplorestheideaofusingaTeslaenginetoharnesswindenergyandsolvethekeyenvironmentalproblems.Aprototypeofthebladelesswindturbinewasbuilt.Acowlingmadefromanumbrellawilldirectthewindthroughscreenednozzlesthatwilldirectittotheedge of a series of disks. The disks are comprised of thin, plastic CD-ROM disks. The wind will drag on the disks following theprincipleofviscosityandtheadhesionofthesurfacelayerofthegas.Thiswilladdforcetothedisks.Inbetweenthedisks,magnetsareemployedtoholdthediskstogetherandtoalsoprovideamagneticfieldthatpassesthroughcoppercoilsand generates electricity. The bladeless wind turbine spun at speeds of 1221 rpm at wind speeds of 25 mph. This produced approximately 32 Volts DC and 2 amps DC. The power generated was 64 Watts. The 32 Volts DC EMF that was produced is belowthetheoreticalvalueof131VoltsDCthatwascalculatedusingFaraday’sLaw.Thereforetheefficiencyoftheprototypeisonly25%.Thisismuchhigherthanthe15%efficiencyofmodernwindturbines.Usingmoreefficientmaterials(strongermagnets,lessfrictionbetweendisksandshaft,additionallayersofmagneticwirecoils,etc.)willwidenthisgap.BasedontheprototypeoftheTeslaBladelessWindTurbinethatwasbuiltandtheteststhatwererun,itisclearthatthedeviceproducesenvironmentallyfriendly,clean,reliable,scalable,low-costrenewableenergy.

TITLE: DevelopmentofaCompositeClayPolymerSpongeforRemediationofOilContaminatedWater.

TherecentExxonValdezandGulfoilspillposedamajorenvironmentalimpacttothefragilemarineandcoastalecosystems.Thisprojectexplorestheideaofcreatinganoleophilic(oilabsorbent),lightweightspongeusingacompositeofclaywithanactivepolymerbinder.Thespongewouldbemoldedandflattenedintolargesheetsandthesheetswouldrolloutandfloatonthetop of the oil spill. The main ingredient for the clay polymer sponge is Fuller’s earth clay which was used centuries ago to soak upoilfromsheepinthewoolmakingindustry.BecauseofthefinegranularityofFuller’sEarth,itisverybrittle.Therefore,aPVCpolymerbinder(LiquidSculpey)wasaddedtothemixture.Themixturewillbemoldedintoshapeandtheproductwillbebakedtodriveoffthewaterandtheresultingmaterialwillbeaporous,low-densitycompositeclaypolymersponge.HighresolutionScanningElectronMicroscopeshowsthattheporousclaypolymerisquitesimilartothatofasyntheticsponge.The porous nature of the material is important in increasing the surface area of the clay polymer sponge sheet and thereby the contact area with the oil. The clay polymer sponge sheet did an exceptional job in absorbing oil. The rate of oil absorption was between 4.3 ml per minute for 5W30 grade of oil and 5.0 ml per minute for 10W40 grade of oil. It absorbed 9.3 times its weight inoilbeforereachingasaturationlevel.Theingredientsoftheclaypolymerspongearenontoxic.ThePVCpolymerusedasthebindingagent,translucentLiquidSculpey,isnon-toxicandconformstoASTMD4236.Fuller’sEarthisasafe,non-toxic,naturallyoccurringclaythatisoftenusedtodeodorizedomesticdogsandcats.

Benthicinvertebrates,vitalcomponentsofthecommunitiesofwhichtheybelong,areimportantindicatorsofenvironmentaldisturbanceinaregion.Inthemarshstudied,threesoilsampleswereobtainedateachsamplingsiteandinvertebratesinthesampleswerelater identifiedbygenusand, ifpossible,species.Theorganismsfoundareknowntoberelativelypollutant-tolerant, indicating a potentially disturbed system. Additionally, due to the intrinsic link between benthic communities and waterquality,waterqualitydatawasobtainedat twosamplingsitesandassessedfordissolvedoxygen,nitrate,phosphate,biologicaloxygendemand,pH,andcoliformbacterialevels.Thefindingsindicatedrelativelynormalconcentrationsateachsite,althoughmorequantitativedataisneededtobetterexaminethewaterqualityanditsbiologicalimpact.Asawhole,thisprojectwillallowforabetterunderstandingoftheenvironmentalconditionsbothinthisecosystemand,onaglobalscale,inother ecological communities.

TITLE: ImpactofTurbineDesignonRenewableEnergyStorageSystemEfficiency ThepresentconcernsofusingfossilfuelsandtheresultingCO2ismakingnationsinvestigaterenewablewaystoproduceandstoreenergy.Thisprojectinvestigatestheimpactofturbinedesignontheefficiencyoftworenewableenergystoragesystems,pumped water and compressed air storage. Two methods of renewable storage that were tested were compressed air storage where energy is used to compress air in storage tank and is released when it is needed, to spin a turbine to produce energy. The pumpedwaterstoragesystemraiseswatertoahighelevationreservoirandwhenenergyisneededthewaterisreleasedtoturna turbine to create energy. In this experiment a turbine design, different than last year, was used in both the pumped water and compressed air storage systems. The pumped water turbine was changed from an open waterwheel design to a closed design. Thecloseddesignhastheadvantageofcollectingalltheenergyfromthewaterbecausenoneislostthroughsplashing.Withthis change the system output energy became 292.63 times greater. The compressed air system was changed by making the diametersmallerandthebladesthickertocatchmaximumairflow.Thesechangesmadethenewdesign58.88timesmoreefficientthantheolddesign.Thisprojectdeterminedthatwhentheturbinedesignischanged,bothstoragesystemswereabletostoreenergymoreefficiently.Continuedrefinementofturbinedesignscouldleadtocapturingahigherpercentageofenergycontained in the storage system.

TITLE: Autonomous Kite Wind Generator to Capture Troposphere Winds

Thequestforcreatingarevolutionarypowergeneratorwiththepotentialtocompetewithfossilfuelsstartswiththewind.Modernwind turbines are extremely efficient, but have insurmountable limitations.The future ofwind energy lies in thetropospherewhichmodernturbinescannotreach.Themostefficientpartofawindturbineisthetipofthebladewhich,insomecases,produces75%ofthepower.Moreover,ifthetower,hub,andinnerbladeareremoved,thetipoftheblademayresemblea kite. Draw a tether to the ground, and the tip of the blade becomes a kite. Mount smaller turbines onto the kite, and power isgenerated.Itwillnotonlyfacethewind,butflyautonomouslyinacircularpath.Inthisproject,akitepowerapparatus,asdescribed,wasbuiltandtested.Toensureanorbitalflightpath,aroboticsystemwascreatedtomaneuveritinacircularmotion. Two different power generators were explored: a traditional turbine and an experimental generator called a windbelt. Thiscost-effectivegeneratorisanylonribbonutilizingaeroelasticflutter.Thewindinducesribbonvibrationandproduceselectricity. The windbelt and turbine were attached to an aerodynamic sport kite under the control of the robotic system. Flying autonomouslyatninetyfeet,thekiteflewinacircularpathmimickingthebladetip.Intheexperiment,anidenticalgroundbased setup was compared to the kite’s output. Experimentation supported the mathematical model in that a 55 inch wingspan kite power generator in 10 miles per hour can produce greater than 100 watts of electricity at a fraction of the expense of modern wind turbines.

Lyme disease is a serious health concern in the U.S. with more than 30,000 reported cases each year. It is caused by the bite ofablackleggedtickinfectedbytheB.burgdorferibacteria.ImeasuredtheprevalenceofB.burgdorferiinfectiontobeashighas93%usingdirectimmunofluorescencemicroscopy.Presently,theonlymethodtocontroltickpopulationsisthroughenvironmentallyharmfulinsecticides.Theseinsecticideskillnotonlyticks,butothernon-targetspeciesincludingmammals.Inordertodeterminethefeasibilityofbiologicalcontrolusingentomopathogenicnematodes,Itestedtheeffectivenessoftwodifferentspeciesontheirabilitytokillticksovera7-dayperiod.Neitherspecieswereeffectiveinkillingnormaladultticks.However,bothwereeffectiveinkillingengorgedadultfemaleswithanLD50of63/cm2.Thenematodeswereapproximatelyhalfaseffectiveagainsttickscomparedtoapositivecontrolofmealworms(LD5025/cm2).Sinceonlyengorgedfemaletickscanlayeggs,entomopathogenicnematodescanpotentiallybeusedonalargescaletodecreasetickpopulationsandpreventLymediseaseinanenvironmentally-soundmanner.

In2007GovernorJodiRelladvisedConnecticuttoofferschoolsassistanceinpreventingthespreadofthepotentiallydangerousbacteria,Methicillin-Resistant StaphylococcusAureus (MRSA).There has been increased attention onMRSA due to theresultingriskofseriousinfection.ThepurposeofthisexperimentwastoinvestigatetheoccurrenceofStaphylococcusAureus,specificallyMRSA,inNewMilfordHighSchool.Thirtysamplesweretakenfromcommonlytouchedobjectsintheschoolandfromthesharedgymequipmentincludinghandballs,goggles,fieldhockeysticks,andmats.ThesampleswerethenincubatedatNewMilfordHospitalonagarbloodplatestoinvestigategrowth.Asneeded,thefollowingtestswerecompletedtoproperlyidentifythegrowthontheagar:agramstainconfirmedgrampositivebacteria,acatalasetestconfirmedStaphylococcus,acoagulasetestconfirmedStaphylococcusAureus,andanOxcillinSensitivityTestconfirmedMRSA.Theresultsconcluded:outofthirtysamples,sixwerefoundtobeStaphylococcusAureus,twoofwhichtestedpositiveforMRSA.Furtherinvestigationwouldbenecessary,howeverthisexperimentationprovesthatMRSAdoesexistinNewMilfordHighSchool,thereforetheimportanceofsanitationcanbeemphasized.

Oneof themainproblems incancervaccination is the strongsuppressionofvaccine-induced immune responses in tumorenvironments,especiallyinpatients65yearsandolder,duetoTcellunresponsiveness.ThegoalofthisstudywastodevelopandtestapotentiallyeffectivebreastcancervaccinetoeradicatemetastasiswithoutinducingTcellresponses.Thedevelopedvaccine isacombinationofCaspase-3,anapoptoticenzyme,and thebacteriumListeriamonocytogenes–an intracellularpathogenthatcandelivervaccineantigens.Listeria-Caspase-3killstumorcellsthroughdifferentpathways:(1)tumorcellkillbyCaspase-3inducedapoptosis,(2)tumorcellkillthroughListeria-inducedReactive-Oxygen-Species,and(3)tumorcellkillby natural-killer cells. Usingatwo-stepprocedure,wild-typeCaspase-3wasclonedintotheListeriabacterium,andthentestedforits infectivity,kill capabilities, and ability to induce apoptosis in MCF-7, a human breast cancer tumor cell-line. The kill experiments demonstrated thatListeria-Caspase-3 killsMCF-7 cells at a similar rate asListeria alone, 97.2% and 97.9% respectively,however,theinfectionexperimentsgiveevidencethatListeria-Caspase-3infected133%moreMCF-7cellsthanListeriaalone.Theapoptosisassay,however,wasinconclusive.Althoughinitialtestingshowspromisingresults,furthertestingisneededtounderstand the capabilities of Listeria-Caspase-3.

Themyeloiddifferentiationprimaryresponsefactor(88)(MyD88)isacriticaladaptorproteininthesignalingoftheinnateimmunesystemthroughToll-likereceptors(TLRs).EvidencefromourlaboratoryindicatedthatexpressionofDNAdamagerepairgeneswasinducedbyTLRstimulation.ToevaluateMyD88’sroleintheDNAdamageresponse,murineembryonicfibroblasts(MEFs)derivedfromMyD88wild-typeandMyD88-deficient(knockout)mouseembryoswerestudied.MyD88knockoutMEFshadlessgrowthwhencomparedtothatofwild-typeMEFs.Knockoutcellsweremoresensitivetoionizingandultravioletradiationthanwild-typecellsasdefinedbycellsurvivalsevendaysfollowingirradiation.Knockoutcellsalsoexhibiteddelayedreentryintomitosisfollowingirradiation.TheseresultssuggestanunexpectedandnovelroleforMyD88inthe cellular response and repair of DNA damage. Thoughhydrogenhasbeenwidelyacceptedasafeasibleenergycarrierforthefuture,itsproductionviaplatinum-basedwaterelectrolysisremainscostlyandinefficient.Whilemostresearchforplatinumalternativeshasfocusedonhydrogenasemimics,thisreportinvestigatestheprotonreductionmechanismofanickelelectrocatalyst,abbreviatedLNiX.Icomputedandanalyzedthe resultsofab-initiocalculationsat theDFT/B3LYPlevelof theory tocharacterize the identityof thisnovelcatalyst. Inaddition,Idevelopedanunderstandingofintermolecularinteractions,andproposedaplausiblereactionmechanismandkineticprofile after considering alternative reactionpathways.Thoroughanalysisof several configurations forLNiX revealed thewater-ligated(LNiH2O+2)andbromine-ligated(LNiBr+1)complexestobethemoststablespecies.Comparisonofreductionpotentialsdevelopedstrongagreementbetweentheoryandexperiment,andfromthisthewater-ligatedcomplexwasdeterminedto be the active catalytic species.A reactionmechanismwas proposed, and though the overall reaction is spontaneous, asmallkineticbarrierwasfoundtoexistbeforethefirstprotonationstep.Thefindingsadvancenotonlythepotentialforthisnewclassofcatalystsasamoreaffordablealternativetohydrogenproduction,butthetimeandcost-efficientcomputationalmethodologies implemented can be extended to study similar organometallic complexes

BioelectrochemicallyAssistedMicrobialReactor(BEAMRs)promisetoemployorganicmaterialsinwastewatertoproducehydrogenathighconversionefficiencies.Inpastliterature,thenon-spontaneousBEAMRreactionhasbeensupplementedbyexternalartificialpowersources.MicrobialFuelCells(MFCs)may,instead,drivesuchBEAMRsbymanipulatingbacteriainthesamewastewatertoconvertbiodegradablepollutantsintoelectricity,therebyeliminatingoutsideinputs.Thisinvestigationfirst determined optimal conditions for anMFC to enable practical applications. Samples ofwastewaterwere taken fromthreestagesatalocalwastewatertreatmentplant:influent,biologicalreactor,andthickenerunderflow.TheeffectivenessofwastewatertreatmentwasestablishedthroughthecolorimetricdeterminationofChemicalOxygenDemand(COD).Pyrexcellsservedasthebaseforsingle-chamberaircathodeMFCs,whiletheelectricitygeneratedwasmeasuredwithadataacquisitionsystem.TheMFCoutput andcost efficiencywas thusmaximizedbyvaryingelectrodematerials, catalyst loadings,PTFEdiffusionlayers,inoculantpre-treatment,carbonsupportmaterials,andbacterialsource(wastewater,C.saccharolyticus).SEMandEDSdeterminedthecompositionandbacteriapresentonsuchelectrodes.Amaximumvoltageof0.65Vwasultimatelygenerated,amountingtoa67%increaseinproductionovercellsconstructedaccordingtopreviousliterature.CODremovalefficiencyincreasedasafunctionofvoltageandindicatedthat94.9±3%oforganicmaterialinthewastewaterwasremoved.ThesuccessfullyoptimizedMFCeffectivelypoweredaBEAMRinafullyautonomoussystemrequiringonlywastewaterasfuel,producing25%hydrogengas,asmeasuredthroughthermalconductivity-basedGC.

TITLE: Anticancer Property of Major Goat Milk Fatty Acids and Delineating their Mechanisms of Action

Colorectal cancer, breast cancer and skin cancer are commonly reported cancer types in the US. Although radiations and chemotherapeutic drugs are routinely used to treat cancer, they produce harmful side effects in patients. Additionally, resistance tochemotherapeuticdrugshasbeennoticedincancers.Thus,thereisaneedforeffectiveandsafemoleculesforpreventingandtreatingcancers.Naturalproductsfromavarietyofsourceshaveservedasabasisforthedevelopmentofnoveldrugs,therebycontributingtohumanhealth.Themedicinalvalueofgoatmilkhasbeenrecognizedforcenturies,andisattributedtothreefattyacids,namelycapric,caprylicandcaproicacids.Thisresearchinvestigatedtheanticancerpropertyofthesefattyacidsonhumancolorectal,skin,mammaryglandcancercells.Thecancercellsweretreatedwithvariousconcentrationsoffattyacidsfor48hoursandcellviabilitymonitoredbyMTTassay.Additionally,thepotentialmechanismsbehindtheanticancerpropertyofthreefattyacidswasinvestigatedbysubjectingmRNAfromcancercellstoreal-timequantitativePCR(RT-qPCR)for determining the expression of cell cycle and apoptosis (cell death) genes. Capric, caprylic and caproic acids reducedcellviabilityofthreecancertypesby70to90%(P<0.05)comparedtocontrols.RT-qPCRdataindicatedthatthesenaturalmoleculesproducedanticancer effectbydown-regulatingcell cyclegenes, andup-regulatinggenes involved in apoptosis.Capric,caprylic,andcaproicacidsexertedsignificantinhibitoryeffectoncancercells,andfutureresearchwillvalidatetheiranticancereffectinanappropriateinvivomodel.
