Faculty Senate Awards Committee OHSU Dear Awards Committee, It is my pleasure to nominate Ganesh Cherala for the OHSU Faculty Senate research award representing the college of pharmacy. Dr. Cherala has served as an excellent example of research for the college of pharmacy since his appointment in 2007. In addition to conducting his own research work, he has worked tirelessly to involve undergraduates in his lab, inspiring many to go on to graduate degrees and research projects of their own. Some of Dr. Cherala’s notable accomplishments and contributions include: 1. K12 award: Through the Office of Research on Women’s Health, Dr. Cherala was awarded his K12 grant in 2012. It is an offshoot of his work with Building Interdisciplinary Research in Women’s Health (BIRCWH) which was partly established by the National Institute of Child Health & Human Development (NICHD). Dr. Cherala has worked to assemble an interdisciplinary team of mentors to guide him through the execution of this award which focuses on the question of women’s reproductive health and oral contraceptive failure.

2. Intracollegiate Collaboration: I am pleased to detail a bit of the collaboration that Dr. Cherala has initiated. In collaboration with Dr. Craig Williams at the college of pharmacy, Dr. Cherala undertook a research project funded by the OHSU Medical research Foundation to elucidate non-genetic mechanisms that could explain inter-individual variability observed with clopidogrel (Plavix). Dr. Cherala collaborated with Dr. Matthew Ito to develop a sterol assay to investigate absorption of dietary sterols and the effects of lipid lowering therapy on those sterols. In collaboration with Dr. Myrna Munar, Dr. Cherala studied the effects inter-individual variations on the levels of tacrolimus in different patients. Tacrolimus is an important medication which helps organ transplant patients to avoid organ rejection. Along with Dr. Ali Olyeai, Dr. Cherala studied the variability of cefuroxime in pediatric cardiac surgery subjects. Cefuroxime is an important antibiotic which helps to avoid infectious complications after surgery. Dr. Cherala also has many collaborations outside of the COP at OSU. Along with Dr. Robert Tanguay, Department of Environmental and Molecular Toxicology, Dr. Cherala developed a high throughput assay in Zebra fish to serve as a research tool for drug discovery projects. 3. Research Environment: Dr. Cherala has done a wonderful job involving junior research personnel. He has established a focus on graduate students which has been particularly tough since most of the infrastructure support for lab-based work is located in Corvallis. Dr. Cherala has been successful attracting private scholarship funding for his graduate students from multiple sources, including the

P a g e | 2 Murdock Foundation. The first student from the Murdock program (2010-2011) is currently enrolled in OSU’s graduate program. To compliment these efforts, Dr. Cherala has been extremely productive in getting his work into the public realm. He is on a strong upward trajectory with corresponding author publications with one in 2011, three in 2012, and four in 2013. He has co-authored 21 papers since joining the college in 2007. Importantly, Dr. Cherala has achieved his research success with the college of pharmacy despite several early roadblocks which were beyond his control. Since there were no successful laboratory-based research faculty in Portland, Dr. Cherala faced a nearly 2-year delay in having permanent laboratory space identified for him. On a personal note, Dr. Cherala is one of the most pleasant co-workers that I have ever had the opportunity to work with. His love of his work is infectious and he never seems to have a bad day at the office. He has been a pleasure to work with from day one. 2014 Faculty Senate Awards Summary

I joined the Oregon State University /Oregon Health & Science University College of Pharmacy (CoP) to establish myself as a clinical translational science researcher and educator. My graduate training in drug metabolism, pharmacokinetics, and bioanalysis followed by postdoctoral training in clinical pharmacokinetics prepared me to undertake bench-to-bedside research. Since joining the CoP, I have developed an integrated research program that investigates perinatal programming of clinical pharmacology with a focus on women’s health.

The focus of my research program is to understand and identify sources of inter-individual variability in pharmacokinetics. The overall objective is to identify and develop easily accessible and economical tools to individualize pharmacotherapy. Ongoing research is focused on body weight as a source of inter-individual variability with a particular interest in birth weight, and its interaction with adult body weight, especially obesity. My research program includes both traditional human clinical pharmacokinetic studies as well as in vitro and animal laboratory work. I have been able to successfully establish a strong research program that has resulted in 21 peer-reviewed publications, 59 poster/podium presentations, and 15 successful extra and intramural grant applications so far.

PI: Daniel Dorsa (Career Development Awardee: Ganesh Cherala) 01/2013-12/2014 NIH Office of women’s health research [K12- Building Interdisciplinary Research Career in Women’s Health] [2K12HD043488-11] $220,000 Prenatal growth programs oral contraceptive metabolism and effectiveness The guiding hypothesis is that the in utero growth restriction will permanently program drug metabolizing enzymes and transporters. This in turn might affect pharmacokinetics. In this proposal, we set out to examine alterations in pharmacokinetics with oral contraceptives as a prototype drug. PI: Ganesh Cherala 09/2013-08/2014 OHSU Oregon Clinical Translational Research Institute $16,000 [NIH NCRR 1 UL1 RR024120] Obesity and Oral Contraception: Obfuscation by Bioanalytical Method Goal of this project is to develop a highly specific and sensitive LCMS/MS assay for oral contraceptive agents. This assay will strengthen ‘innovative’ section of future grant application(s) besides providing more reliable data for pharmacokinetic data analysis. (c) Completed PI: Ganesh Cherala 06/2008-06/2009 13Therapeutics, Inc. $26,895 Assay development and stability studies of new drug molecules PI: Ganesh Cherala 12/2008-12/2009 Heart Drug Research Inc. $2,000 Secondary analysis of Plavix data We have determined plasma levels of clopidogrel and its metabolites and used as a measure of clopidogrel compliance among relevant patient population. These findings led a publication followed by a research grant from a foundation. This research gift has resulted in peer-reviewed publications (Original research -PMID: 20062927; Review article- PMID: 19958858). PI: Ganesh Cherala Spring 2009 Oregon State University Research Equipment Reserve Grant $15,000 Real-time PCR upgrade Real time PCR machine was upgraded using these internal funds. This equipment grant greatly facilitated significant amount of research. A peer-reviewed publication [PMID: 23507230] is a result of this funding. PI: Ganesh Cherala 01/2009-06/2010 Cubist Pharmaceuticals, Inc. $15,497 Evaluation of renal and non-renal metabolism of daptomycin in neutropenic rats In prior human studies carried out by our research team [PMID: 19015332], we suspected metabolism of daptomycin to be altered as a function of neutropenia in cancer patients. This particular project examined the above hypothesis. PI: Amira Al-Uzri (Co-I: Ganesh Cherala) 06/2009-12/2010 Doernbecher foundation $15,000 The center for Pediatric Pharmacology and Therapeutics at DCH. We were successful in attracting funds to form a pediatric research center. As a pilot study, we have examined ethnicity as a source of inter-individual variability in Tacrolimus pharmacokinetics, and developed a relatively non-invasive drug monitoring technique for pediatric population. This project has led to two publications [PMID: 22172831 and 23548676], and an invention disclosure. PI: Alison Edelman (Co-I: Ganesh Cherala) 09/2009-08/2011 NIH (R01HD061582] $460,240 Improving contraceptive effectiveness in obese women

Collaborated with clinician researcher to investigate (a) the mechanisms of oral contraceptive failure in obese women, and (b) intervention strategies to make contraceptive regimens effective for obese women. We have successfully completed the grant, and disseminated findings in various forums including peer-reviewed journals. Furthermore, this grant has provided valuable pilot data for the successful K12 grant I received last year. PI: Ganesh Cherala 06/2010-05/2011 Medical Research Foundation of Oregon $40,000 Use of TNF-α, in addition to CYP2C19 genotype, to predict pharmacokinetics and pharmacodynamics of clopidogrel. Inter-individual variability in clopidogrel is a major hurdle in the pharmacotherapy of stroke. We have examined non-genetic factors to understand the pharmacokinetics and pharmacodynamics of clopidogrel in adult women. This work has identified a novel mechanism that could further explain the large inter-individual variability associated with clopidogrel. The study findings are disseminated in the form of peer-reviewed article [PMID: 23116430], and presentations at various national meetings. PI: Joshi Alumkal 06/2010-05/2011 Kuni Foundation $100,000 The effects of sulforaphane dose escalation in men with recurrent prostate cancer The pharmacokinetics and pharmacodynamics of sulforaphane were determined in men with recurrent prostate cancer. My contributions include PK study design, analysis, and assistance with manuscript development. Besides two poster/podium presentations on this work, a manuscript is currently under preparation. PI: Ganesh Cherala 01/2011-06/2011 GE Healthcare $5,951 Development of Dried Blood Spot technique for CYP phenotyping In addition to NIH sub-award, funds were secured from industry to develop dried blood spot technique. PI: Jeffrey Jensen 10/2012-12/2012 OHSU (NIH subcontract- HHSN275200403378I) $5,000 A Phase I Study to Evaluate Pharmacokinetic and Pharmacodynamic Profile of a Single Injection of Levonorgestrel Butanoate (LB) for Female Contraception In this project, novel contraceptive agent was clinically tested for PK and PD profile. Curriculum Development

(1) PHAR 770 Advanced Pharmacokinetics. [cross listed for graduate studies] I created and frequently update all of the lectures I provide to the students in my area of expertise. This is a continually evolving process designed to improve the student’s knowledge of pharmaceutical care in the area of preclinical and clinical pharmacokinetics. I have introduced three new topics to this course: (a) bioanalysis and PK study design, (b) inter individual variability, and (c) pharmacogenomics. These three topics address “personalized medicine,” an up and coming area in the clinical practice. I believe that these new topics will prepare professional Pharm.D. students to be at the forefront of delivering individualized/personalized medicine.

(2) PHAR 762 Pharm CSI Elective. [cross listed for graduate studies] I have been extensively involved in the development of this new elective course. We have used real world examples to combine pharmacokinetics with forensic sciences using celebrity cases as examples. We have limited student enrollment to a maximum of 12 to provide the best experience to enrolled students. We have developed didactic lectures, journal club readings, and homework problems to solve each of the celebrity death cases. The students learn about various pharmacokinetics tools available to solve such high profile cases.

(3) PHAR 505/705 Reading & Comprehension (Variability in PK) I have developed a reading & comprehension course catering to students interested in understanding inter-individual variability in PK processes. The course involves a combination of journal article readings, presentations, and wet-lab experiments to get a comprehensive understanding of the concepts. The laboratory component is geared toward awareness of common techniques and tools for undertaking research study.
