Introduction
The 971-FP Performance Check Kit (AX2055-8) contains eight Test Capsules, each preloaded with a three component mixture carefully chosen to routinely monitor the performance of your 971-FP Flash Purification System. In addition, the Performance Check Kit includes a pack of eight SuperFlash SF10-4 g Si 35 columns (AX1370-8).
The 971-FP Test Capsule 48-Pack (AX2055-48) contains six separately sealed packets of eight Test Capsules, each preloaded with a three component mixture, 48 capsules in total. The capsules are intended for use with SuperFlash SF10-4 g Si 35 columns which can be purchased separately in packs of eight (AX1370-8) or 96 (AX1370-96).
Using the preloaded Test Capsules and matching SuperFlash columns after installation and then as a periodic system check helps build a clear picture of your instrument’s performance over time. The necessary method details to run the 971-FP Performance Check Kit are provided overleaf.
Before You Begin
Ensure you have sufficient solvent and you primed your 971-FP Flash Purification System with Heptane (A) and Ethyl Acetate (B). Remember you should start by priming line B then switch to priming line A because it is primed with the least polar solvent.
Method Setup
Stage 1: Select “New” to create a new run:
Stage 2: Select the Column Type “SF10-4 g”, and ensure that the correct solvents are selected:
Stage 3: Select the following detection settings:
Stage 4: Create the following gradient:
0 – 100% B, 0 – 8 mins; 100 – 0% B, 8 – 9 mins; 0% B, 9 – 10 mins.
Stage 5: Load a SuperFlash SF10-4 g, Si 35 column onto the column station and press P to confirm:
Stage 6: Press Run:
Stage 7: Check the details are correct in the Run Summary and then press Continue to begin equilibrating the column:
Stage 8: Once the column has been equilibrated, the run will pause. Fit a Test Capsule onto the top of the SuperFlash column:
Stage 9: Resume the run:
Stage 10: Run complete after 10 minutes:
Stage 11: Performance check of the system
If the system is working correctly then a chromatogram showing three separate peaks, as illustrated above, should be produced.
The system performance check is now complete. Enter comments and Finalize:
Note: The chromatogram may not exactly match the example above. Slight variations in peak height may occur due to laboratory conditions, exposure to light, etc.
If the chromatogram does not show three clearly resolved peaks then the system is NOT performance checked. It is recommended that the process is repeated using a new Test Capsule and SuperFlash column.
