� A S S AY P R I N C I P L ECholesterol Dehydrogenase (CDH) catalyses the following reaction:

Cholesterol + NAD+ Cholest-4-en-3-one + NADH + H+

The appearance of NADH is measured at 340nm by spectrophotometry.

Isoelectric Point: 4.5

Kmvalues: Cholesterol; 1.5 x 10-4 MNAD+; 2.3 x 10-4 M

Optimum pH (Fig. 1): pH 10.0

Optimum Temperature (Fig. 2): 30°C

pH Stability (Fig. 3): 6.5 to 7.5 (37°C for 15 minutes)

Thermal Stability (Fig. 4): Stable at 35°C and below (pH 7.0 for 5 minutes)

Inhibitor: Ag+

Activator: Triton X-100
