Judul : Korelasi Luas Area Wharton’s jelly dengan Luaran Berat Lahir
Bayi pada Kehamilan Cukup Bulan
Tempat : Laboratorium Sentral RSUP Dr. Kariadi
1. Potong tali pusat sepanjang 2 cm dari ujung akhir
pemotongan, lalu masukkan kedalam container
yang telah berisi Neutral Buffer Formalin dengan
perbandingan 1:10. Beri Identitas dan bawa ke
laboratorium Patologi Anatomi
2. Jaringan mentah dipotong menjadi 3 bagian (pilih
bentuk potongan yang baik) lalu masukkan
kedalam kaset. Lalu, masukkan kaset kedalam
Neutral Buffer Formalin.
3. Pemasakan jaringan selama 17,5 jam :
Kaset dimasukkan kedalam keranjang, kemudian
masukkan kedalam alat pemasakan jaringan
Microm STP 120, yang terdiri atas beberapa
tabung dengan lama masing-masing waktu
perendaman :
- Formalin 10% 1 selama 2 jam
- Formalin 10% 2 selama 2 jam
- Alkohol 70% selama 1 jam
- Alkohol 80% selama 1 jam
- Alkohol 96% selama 2 jam
- Etanol selama 2 jam
- Xylol selama 1 jam
- Parafin 1 selama 2 jam
- Parafin 2 selama 2 jam
- Parafin 3 selama 2 jam

4. Jaringan yang telah melalui proses pemasakan
kemudian dibuat menjadi blok jaringan
menggunakan alat histocenter. Kemudian blok
parafin didinginkan kedalam freezer.
5. Setelah didinginkan, blok jaringan kemudian
dpotong menggunakan mikrotom. Hasil
pemotongan dimasukkan kedalam air hangat lalu
ditempelkan pada kaca objek. Beri identitas
6. Preparat kemudian dipanaskan dalam oven 70%
selama 10 menit untuk melelehkan sisa parafin.
Kemudian dinginkan dan tata di rak.
7. Proses pewarnaan dengan alat Veristein dengan
total waktu 2,5 jam :
- Xylol 1, 2, dan 3 masing-masing 10 menit
- Etanol selama 3 menit
- Alkohol 96% selama 3 menit
- Alkohol 80% selama 3 menit
- Alkohol 70% selama 3 menit
- Alkohol 50% selama 3 menit
- Bilas dengan air mengalir
- Hematoksilin selama 15 menit
- HCl selama 1 menit
- Bluing selama 10 menit
- Air mengalir
