Pharmacodynamic

Salbutamol is a short-acting, selective beta2-adrenergic receptor agonist used in the treatment of asthma and COPD.

Salbutamol is formulated as a racemic mixture of the R- and S-isomers.

Mechanism of Action

Salbutamol is a beta(2)-adrenergic agonist and thus it stimulates beta(2)-adrenergic receptors.

Salbutamol increases cAMP production by activating adenylate cyclase.

Increased intracellular cyclic AMP increases the activity of cAMP-dependent protein kinase A, which inhibits the phosphorylation of myosin and lowers intracellular calcium concentrations.

Mechanism of Action

A lowered intracellular calcium concentration leads to a smooth muscle relaxation and bronchodilation.

Increased cyclic AMP concentrations are also inhibits the release of bronchoconstrictor mediators such as histamine, leukotreine from the mast cells in the airway.

Pharmacokinetics

Pharmacokinetics Bioavailability :oral bioavailability 50%Half life : half life is 4 hours (Taken from rang & dell page 168)

Onset of Action:oral: 30 minutes, Inhalation : 5-15 minutes

Duration of action :

8 to 12 hours(page 361 R&D)

Metabolism : It is subject to first pass metabolism in the liver

Pharmacokinetics

Exretion : Readily excreated in the urine as metabolite and unchanged drug Some excreate in faeces.
