    Subject Operating Procedure for Polarimeter.

    1.0 Purpose:

    To ensure the proper operation of Polarimeter.

    2.0 Scope :

    The SOP is applicable for the operation of Polarimeter

    4.0 Definition:

    SOP: Standard Operating Operating Procedure.

    5.0 Procedure:

    1. Reading the Scale:

    The scale seen in the telescope is divided at 1o intervals from 1 to 3600 adjacent to 0 and

    below the main scale is the international sugar scale that may be used directly in the

    evaluation of sugar solutions.

    2. Use of Micro meter Drum:

    1. The micrometer drum is employed to subdivide the angular and I.S.S. scale and itself

    carries two scales, angular on the left subdivided of intervals of .05 and I.S.S. on the

    right subdivided at intervals of 0.1.

    2. The index marker is at the top of the aperture for both scales. It is noted that movement

    of the drum from 0 to 1.0 cause the line seen in eyepiece to traverse exactly one scale

    division respectively for the I.S.S. and angular scales.

    3. Using the knurled wheel of the micrometer drum rotate it to bring the index line

    coincident with a scale line. The true reading is then the scale line concerned plus the

    drum reading indicated on the appropriate scale.

    3. Sample Tubes & Filling:

    1) To obtain accurate results, great care must be exercised in filling the tube and

    ensuring that the end windows are not strained thus causing erroneous results.

    2) Remove both end caps of the tube and detach the glass window which must be

    carefully cleaned. The ends of the tube must now be cleaned and dried.3) Place one window over the end of the tube and replace the caps.

    4) Carefully screw the cap down until the first resistance appears. Do not screw any

    further. Invert the tube and ensuring that the open end is dry, pour in the sample

    carefully until a meniscus appears bulging above the tubes end.

    5) There should be no air bubble, if there is any air bubble that is supplied in bubble

    trap.

    4. Adjusting to Zero:

    1. With no sample in the through set the field to a precise match observed in

    telescope. Set the micrometer drum to zero and observed the scale in

    telescope.

    2. Grasp the knurled washer and with the other hand slacken the knurled

    nut. The drum is now released from the screw.

    3. Turn the screw by mean of knurled washer until the index line coincides

    with 0 on the scale. Ensure that the drum still reads zero, if not bring it tothis position by moving it through the rectangular aperture in the sleeve.

    4. Check the balance and that the scale and drum read zero.

    5. Operation Mode:

    1. Prior to test the operator ascertains that the instrument with empty

    observation tube inserted is at zero position. The field of view show three

    comparison fields of equally gray shade, separated from each other by

    two fine vertical dividing line and the scales read zero.

    2. Thereafter the solution poured into the observation tube and place on the

    through.

    3. Now the operator observes three field of different shade.

    4. The analyzer with triple shadow effect and the graduated circle scale now

    be turned by mean of the knob until the three field of comparison showagain equally gray shade.

    5. The reference graticule on the scale plate now indicate the angle of

    rotation both for dextro rotation and laevo rotation.
