Documentation Guidelines

1.Application Form for master’s Degree

(1) Please sign and seal. (Foreigners may use their signatures instead of a seal.)

(2) The Academic advisor’s signature and seal are required.

(3) Please write candidate’s names of examiners.

The Chief examiner must select a professor from the GSMS.

An academic advisor, an introductory advisor, coauthors of Thesis, and advisors who belong to the same

department of applicant are not permitted to be examiners.

2.Master's Thesis

(1) Language should be Japanese or English.

(2) If the title of the Thesis is Japanese, an English translation should be provided along with the Japanese;

if English, Japanese should be provided with the English.

(3) The paper should be A4 size, vertical, horizontal, and the margin should be 25mm on both sides.

(4) Make the font size about 10.5 to 12 points.

(5) A table of contents should be prepared according to the order below. Figures and tables should be

appropriately inserted.

5) Results

6) Discussion (including bibliography)

(6) The paper shall be printed color double-sided,

The submission in permanent binding is not required.

3.Abstract of Master’s Thesis

(1) Please keep it simple and easy to understand in 2 pages of A4 size

(2) As the sample below shows, it is recommended in principle that the abstract to be divided into 5 parts;

objective, method, results, discussion, and conclusion.

(3) Please do not use of abbreviations as much as possible. In case of using abbreviations, please write the

words in full followed by the abbreviation in parentheses, when you use an abbreviation for the first time.

Example: Duchenne muscular dystrophy (DMD). However, this does not apply to abbreviations widely

recognized in medical science and biology (for instance: DNA, ATP).

(4) The titles recorded in the Master’s Thesis and the abstract should be corresponded every single word and

phrases, including either capital or small letter as well as italic letter.

4.Resume

(1) Prepare the margins: top 25 mm, bottom 15 mm, left 25 mm, right 15 mm.

(2) The size of the text should be 10 point.

(3) Please sign and seal. (International Students may use their signatures instead of a seal.)

(4) The resume should be no more than one page.

(5) Kana over name to indicate pronunciation.

(6) International Students shall enter their nationality.

(7) Please state your educational background starting from graduation of bachelor’s degree of the University.

[Purpose]HIV-1(ヒト免疫不全ウイルス)特異的な細胞傷害性 T 細胞(CTL)エピトープを同定することは

、AIDS(後天性ヒト免疫不全症候群)の病態やワクチン開発など様々な研究のために必須である。これまで

に HIV-1 clade B 感染者において比較的多数の clade B 特異的 CTL エピトープが同定されているのと対照

的に、それ以外の non-clade B 特異的 CTL エピトープは報告が少なく、clade E においては全くない。そこ

で、アジア人に高頻度にみられるヒトリンパ球抗原(HLA)である HLA-A*1101 に拘束される clade B 特異

的 CTL エピトープの同定を行い、さらにこれを用いて東南アジアにおいて主なサブタイプである clade E に 特異的な CTL エピトープの同定を試み、clade E ウイルスに対する CTL の認識を解析した。 [Methods](1)はじめに、リバースイムノジェネティクス法によって、日本人の HIV-1 clade B 感染者で認識

されている 4 種の HLA-A*1101 拘束性 clade B 特異的 CTL エピトープを同定した。(2)これらと既知の 4種の HLA-A*1101 拘束性 clade B CTL エピトープを用いて、clade E の CTL エピトープを同定することを試

みた。まず、タイ人の clade E 感染者の末梢血単核球を、clade B CTL エピトープ部位に相当する clade Eペプチドで刺激することによって特異的 CTL を誘導できるか調べた。(3)さらに、誘導された clade E CTLが clade B ペプチドを認識するか、clade B CTL が clade E ペプチドを認識するか調べることによって、CTLエピトープが clade 間で共通(cross-clade)に認識されるか、あるいは clade 特異的に認識されるか解析した

。 [Results]Pol 由来の 3 種の CTL エピトープは、タイ人の clade E 感染者でも認識されている

cross-clade CTL エピトープであった。また、3 種の CTL エピトープ部位に相当する clade B、clade E ペプチ

ドは、それぞれが日本人の clade B 感染者、タイ人の clade E 感染者で認識されている clade 特異的 CTLエピトープであった。HIV-1 clade E 感染 T 細胞におけるこれらの CTL エピトープの抗原提示能を明らかに

するために、それぞれ 3 種の cross-clade CTL および clade E 特異的 CTL が clade E ウイルスに感染し

た CD4+ CXCR4+細胞を認識するか観察した。その結果、これらの CTL エピトープを認識する CTL は clade E ウイルスに感染した標的細胞を特異的に傷害したことから(図1)、6 種の CTL エピトープは clade E ウイ

ルス感染細胞の細胞内でプロセッシングされ抗原提示されていることが示された。

Expression of HIV-1 p24 and HLA-A*1101 antigens in HIV-1 clade E infected cells. 721.221-CD4-A*1101 cells were infected with HIV-1 clade E clone p93JP-NH1. Uninfected cells and infected cells were analyzed for HLA-A*1101 expression on target cells by staining with a p24-specific mAb and anti-HLA-A11 mAb. 95% of cells were p24-positive and approximately 30% of p24-positive cells showed downregulation of HLA-A*1101. B. Cytolysis of HIV-1 clade E infected cells by HIV-1 specific CTL. Infected 721.221-CD4-A*1101 cells (closed bar) and uninfected 721.221-CD4-A*1101 cells pulsed with (shaded bar) or without (open bar) the corresponding peptide were used as target cells. Cytolytic activities of three cross-clade epitope-specific CTL clones (Pol424-432-34, Pol894-903-14 and Pol675-683-5E-3) and three clade E epitope-specific CTLs for these target cells were examined. The Gag 263-272-specific, HLA-A*2402-restricted CTL clone Gag 263-272-3 was used as a negative control. The specific lysis of this clone for C1R-A*2402 cells pulsed with (dotted bar) and without (open bar) the corresponding epitope peptide was also examined. [日本語説明でも可] [Discussion]本研究によって、すでに明らかにされている HIV-1 CTL エピトープを用いて、他の clade のエ

ピトープを同定する事が可能である事が示された。この事は、clade E, C のように未だほとんど明らかにされ

ていない CTL エピトープ同定をより効率的におこなえる事を示している。しかしながら、本研究では、変異

性の高い Env 領域に対しての CTL エピトープの同定は不可能であった。今後、Env 領域内のエピトープを

用いて、変異性の高い領域での CTL エピトープの同定がどの程度可能かを明らかにする必要がある。 [Conclusion]本研究において、HIV-1 clade B 特異的 CTL エピトープを利用したこの解析法は、clade E 特

異的 CTL エピトープを決定するためにきわめて有効であることが示され、この既知の clade B CTL エピトー

プを利用する方法を用いて、ほとんど解明されていない clade E CTL エピトープの同定が可能となった。
