1. GETTING STARTED 

1.  Turn on the items (1‐6) 3 hours before usage (for stabiliza9on of the temperature chamber, and water temperature).  

2.  Double‐click on the Biosta9on IM icon on computer screen.

3.1 NEW TIME‐LAPSE SETTINGS 

3.1.1 For 1 Phase Contrast and 2 Fluorescence Channels Experiment 

Step 1 (Refer to Figure 1) : Click on New Time-lapse Setting to start new Time-Lapse Experiment.

Step 2 (Refer to Figure 2): Click on Manual tab and set the adjustments for: i)  Light Intensity ii)  Exposure Time iii)  Gain iv)  Resolution

Step 3 (Refer to Figure 3): Click on the excitation lights to select the light channels: a) Phase Contrast (Ph) b) Fluorescence I c) Fluorescence II

Step 4 (Refer to Figure 4): Select the magnifications required: a) 10X b) 20X c) 40X

Step 5 (Refer to Figure 5): i) Click the “Points” tab when defining multi-points. ii) Once the desired points have been chosen, click on the “Triangle” tab to register point. Repeat the procedures for another points.

Step 6 (Refer to Figure 6): i) Click on “Time” tab for time configuration. ii) Click the “New” button to enter the acquisition cycle and total time. iii) Click Add.

Step 7(Refer to Figure 7): Ensure temperature is stable.

Step 9(Refer to Figure 9): When the dialogue box for assigned time-lapse settings appears, check through the settings and click “Start Time Lapse” Save the settings in .nex file and time-lapse is in process.

Step 8(Refer to Figure 8): Click on the “Start Time Lapse” Button to start experiment.

6. SAVING TIME‐LAPSE RESULT FILE 

1)  Click on the “Save” Button to display the Save Image dialog box. 2)  Select “Export Images and Data (Multi-points)” and save the images into (.TIFF/

JPEG) for images snapshots or save the images into .AVI for video.

IMAGES ACQUIRED CAN BE VIEWED IN 2 MODES: A) CHANNELS MODE OR B) POINTS MODE

Click Export Images and Data (Multi-points)

Select all points, check channels and click Next.

Assign saving folder, file format (either .TIFF or .AVI) and filename.

Proceed to save the files.
