Assays forboth, the activation of G-protein coupled receptors(GPCRs or 7TM receptors) inducing inositol phospholipid degradation and the activity of voltage gated Ca++ ion channels – the latter causing a rather slow influx – can easily be performed with the use of the Centro´s reagentinjectors.

ATP determinationA detection limit of less than 1.5 attomoles of ATPper well makes the Centro XS3 one of the best suitedmicroplate instruments for the determination ofcell viability, e.g. in tumour chemosensitivityassays, cell proliferation, antibiotic susceptibilitytesting or bacterial detection.

QC luminescence test kitWith the QC luminescence test kit the performanceof your luminometer can be checked. This is theideal quality control (QC) methodas performance of both injectionand detection systems can bemonitored. The test kit follows afast flash–type kinetics. The test kitcontains a „label” and two types of starterreagents.

Injector Cleaning SolutionFor proper maintenance of the injection system the Cleanit solutionsare recommended for regularcleaning. Daily cleaning ensures thataccuracy and precision as well aslong life cycle of the injectors will bemaintained.

Temperature controlThe Centro can be equipped with plate heating forapplications where the assay must be measured atelevated and constant temperatures as common incell based assays.

The sandwich-type heating plate design reducesevaporation effects and ensures homo-geneous temperature distribution.

Robot accessSpecially designed for, but not restricted to, use inHTS departments of drug discovery companies, the Centro as well as the Centro XS3 robot accessmodule allows for easy integration of the unit intoany type of lab automation system.

Software and hardware easily interface into existing robot, stacker or liquid handling systems. A barcode reader may be used for positive plateidentification.

Reagent dispensing3 independently controlled injectors with variablevolume give entire freedom in the selection ofassay type and assay sequence.

The volume is adjustable from 10 to 100 µL– perfectly fitting the demands for reagent addition into 96 and 384 well plates.

In Centro dispensing can even be performedduring a kinetic measurement, e.g. to watch theeffects of added agonists / antagonists.

Applications

Reporter Gene AssaysIn basic research of generegulation as well as in drug discovery the use ofluciferases, ß-galactosidases,ß-glucuronidases and secreted alkaline phosphatases have become a standard tool offering the highest sensitivity.

Especially the dual luminescence type assays,e.g. Dual-Luciferase® Reporter Assay, have become a favourite means as they provide aninternal control for transfection efficiency orgeneral expression level.

Aequorin based Calcium monitoring

The monitoring of the changes in intracellular Ca++

levels by the means of aequorin in a flash-typeluminescent reaction is highly efficient. Assays forboth, the activation of G-protein coupled receptors(GPCRs or 7TM receptors) inducing inositol phospholipid degradation and the activity of voltage gated Ca++ ion channels – the latter causing a rather slow influx – can easily be performed with the use of the Centro´s reagentinjectors.

ATP determinationA detection limit of less than 1.5 attomoles of ATPper well makes the Centro XS3 one of the best suitedmicroplate instruments for the determination ofcell viability, e.g. in tumour chemosensitivityassays, cell proliferation, antibiotic susceptibilitytesting or bacterial detection.
