FREESLATE CORE MODULE 3 (CM3)

Increasing chemical research and development (R&D) productivity by delivering high throughput research solutions has been a focus for Freeslate since 2010. Freeslate scientists have years of hands-on experience developing best-in-class automated research systems designed to dramatically impact customers’ R&D productivity. The CM3 is a configurable laboratory automation system for preparing, processing, and testing complex chemical samples and mixtures. This third-generation platform comes with the powerful LEA software suite and enables scientists to leverage a single instrument to run an array of applications, including:

• Small molecule solid form screening

• Reaction screening and process optimization

LAB EXECUTION & ANALYSIS (LEA) SOFTWARE

All CM3s are run by the powerful LEA software suite*. LEA provides a complete laboratory infor-matics environment to support high throughput, parallel, and automated experiment design, execu-tion, analysis, and reporting. The software suite supports automating, integrating, and linking laboratory hardware and capturing instrument data in a centrally accessible and shareable information store. LEA software captures experiment design informa-tion, real-time run informa-tion, and analytical results together in one place for easy reporting and fast analytical decisions.

6. Variable Vial and Plate Gripper – Sample handling arm capable of gripping, moving, and rotating vials from 1 mL to 125 mL and microtiter plates. The range of motion supports physical integration with a variety of analytical and processing equipment.

7. Flexible Solid Dispensing – Solid dispensing using either storage vial (SV) powder dispensing for small quantities or classic Powdernium™ powder dispensing* for larger quantities. Accurate and precise dispensing is supported from 500 µg to 5 g.

8. Controlled Heating/Cooling and Mixing – Three independently controlled temperature zones for heating up to 180 °C and cooling down to -20 °C. Element includes magnetic stirring up to 750 rpm.

Flexible Liquid Dispensing – Disposable single-channel dispenser for viscous liquids available along with versatile six-channel dispenser and heated one- and four-channel dispensers, allowing access to plates with commonly used vial spacing. Heated four-channel dispenser includes a single extendable tip, allowing four samples to be dosed at once or one at a time with the tip extended.

e. Rapid pH Measurement – Available with single or four-channel pH measurement probe using the latest solid-state pH probes with 1-13 pH range and a resolution of 0.1 pH units.

1. Viscometer* – On-deck viscometer for accurate measurement of Newtonian and non-Newtonian samples from 1 cP to 100 cP.

2. High Pressure and Temperature Reactors* – Eight reactors that support high pressure reactions and high temperatures. Additions and sampling can occur at pressure during the reaction.

3. Visual Inspection Station – Non-invasive color inspection, turbidity measurement, and particle count in a sample using an integrated camera.

4. Carousel – Integrated carousel for flexible storage of samples, tips, etc.

5. Integrated Balance – Analytical balance that allows for dispensing into plate-based arrays or individual vials on the balance. The balance includes an integrated digital camera for visual sample inspection.

CM3 PRODUCTS

Biologics Formulation SystemCreating stable biologics formulations poses a significant challenge. Through collaborations with pharmaceutical/biotechnology R&D organizations, Freeslate has developed a unique automated system to address the challenges in biologics formulations. Built on the CM3 platform, the system automates formulation preparation, sample stressing, sample preparation for analysis, and analytical tests such as pH and viscosity measurement and particle detection. The goal of this innovative approach is to quickly develop more robust formulation candidates while using less material and keeping personnel levels unchanged.

Preformulation SystemThe Freeslate CM3 enables scientists to conduct preformulation studies on a single product in a high-throughput manner. The platform is configured with heated liquid handling, powder dosing, and independently controlled heating, stirring, and cooling zones that seamlessly support Freeslate’s class–leading heated filter block and universal crystallization substrate. With the CM3 Preformulation System, the number of crystallization and solubility studies completed yearly may increase by up to a factor of 50. For polymorph screens, finding a suitable crystalline form is accelerated while reducing the risk of late-stage failure due to emergence of new metastable forms. For salt screens, a broader examination of reaction conditions and solvent systems is achieved, effectively optimizing the crystallization process.

Reaction Screening SystemFreeslate’s automated and configurable CM3 system enables scientists to perform process research and development (PRD) in a high-throughput manner. Because the CM3 comprises multiple independently controlled heated and cooled zones, heated liquid handling, powder dosing, automated capping and decapping, and magnetic stirring, it is now possible to screen hundreds of reaction conditions daily

under a variety of reaction composition variables and conditions to quickly optimize process parameters while using minimal amounts of materials. With greater throughput, scientists can explore reaction parameters outside conventional experimentation, reducing risk and allowing for non-intuitive discoveries.

Solid Dispensing SystemThe Freeslate automated and configurable CM3 robotic system enables scientists to perform solid dosing in a high-throughput manner. The CM3 Solid Dispensing System allows reliable and consistent solid dosing dispensing of a broad range of solids with varying properties. Both the classic Powdernium™ powder and SV powder dispensers are used for accurate, precise dosing of varying quantities of solid materials into an on-deck balance with integrated camera.

Metal Impregnation System (MIPS)True acceleration of heterogeneous catalyst development cannot be fully realized without the ability to prepare catalysts rapidly. The Freeslate CM3 MIPS has unmatched capabilities in this area. The CM3 integrates unit operations of solid support dispensing, precursor solution preparation, solution pH measurement, and incipient wetness or other impregnation techniques. MIPS allows for the synthesis of hundreds of supported catalysts per week, producing high-quality materials for screening in either high-throughput reactors or larger scale individual reactors. This allows for rapid discovery and optimization of new heterogeneous catalysts.
