University Health Network C. ACADEMIC HISTORY 1. Research Endeavors: a) Molecular regulation of vascular smooth muscle cell proliferation and development: Our

studies focus on the role of the c-myb proto-oncogene. We have demonstrated how its encoded product, the c-Myb transcription factor, facilitates G1 to S phase cell cycle transitions in vascular smooth muscle cells (SMC), in part, by down-regulating the transcription of specific plasma membrane Ca2+ ATPase (PMCA1 and PMCA4). This, in turn, enables critical elevations in cell Ca2+ concentrations that are required for cell cycle progression. We have cloned the PMCA1 promoter and have demonstrated its specific structural and functional interactions with c-Myb. We have also recently cloned the mouse PMCA4 gene. We are identifying its promoter, and aim to study the structural basis for its functional regulation during cell cycle. We also study the role of other Ca2+ regulatory genes (SERCA, IP3R) in cell cycle progression and are interested in defining putative Ca2+-responsive cell cycle regulators. In this regard, we have become particularly interested in candidate mediators such as the calcineurin/NFAT pathway, Ca2+/calmodulin-dependent kinases and cdk2/cyclinE. Finally, we have developed an in vitro SMC differentiation schema for mouse embryonic stem cells and are characterizing the transcriptional profiles of differentiating embryoid bodies at specific temporal windows. We aim to employ these techniques for studies on the role of c-myb and calcium regulatory genes during SMC

differentiation. The above studies form the molecular basis of our in vivo experimentation (see below) and have been the specific objectives of my CIHR grants on c-myb.

b) Mouse models of cardiovascular gene regulation and pathobiology: We have developed

transgenic mice possessing arterial SMC-restricted expression of a tetracycline-responsive transcriptional activator (tTA) and have used this system to conditionally express a variety of tTA-dependent transgenes in vivo (Myb-Engrailed: a dominant negative Myb; PMCA1; and PMCA4). We have characterized the function of the tTA system in a wire-mediated denudation model of carotid artery injury and use this model to gauge the physiological importance of transgenes of interest. We have bred our conditional transactivation system into LDL receptor knockout mice (LDLr-/-) in order to study the effects of our transgenes in cholesterol-mediated atherogenesis. The Myb-Engrailed over-expressing animal is dramatically protected from the neointima formation that follows arterial injury in control animals. PMCA4 over-expressing animals are also protected in such a model, but show an additional phenotype that has become the major focus of our HSFO-funded research. Namely, PMCA4 over-expression in arterial SMC results in a unique model of increased blood pressure, which is associated with enhanced myogenic tone and increased sensitivity to endogenous vasoconstrictors. We believe this is due to a heightened pharmaco-mechanical coupling of SMC contraction caused by compensatory increases in the storage of intracellular Ca2+, and decreased activity of neuronal nitric oxide synthse (nNOS) which has been shown to be both structurally and functionally coupled with PMCA4. These two mechanisms are not mutually exclusive and both are being actively pursued. We have also engaged in a number of collaborative projects employing our mouse models of injury and/or conditional expression systems for other transgenes of interest. These include Elafin, an elastase inhibitor and Chymase, an endogenous matrix and zymogen protease (collaborations with Marlene Rabinovitch, Stanford University), and iNOS and Endothelin (collaborations with Duncan Stewart, University of Toronto). The latter two genes have also been expressed in a conditional cardiac-restricted pattern. The cardiac iNOS over-expressing mouse represents a unique model of sudden cardiac death due to a particular propensity of the mouse AV conduction system for iNOS-derived peroxynitrite and/or superoxide. The cardiac endothelin over-expressing animal is a fascinating model of immune cardiomyopathy resulting in heart failure. Both the iNOS and Endothelin projects are part of HSFO and CIHR grant applications from Duncan Stewart and I, and these models will be a major focus of our collaborative research for the next 3 years. Dr Peter Backx (as PI, University of Toronto) and I (as co-PI), have recently been awarded a 5-year CIHR grant for studying the cardiac and vascular function of the ACE2 knockout animal. My role in this project will be to focus on the vascular phenotype of this intriguing mouse. Finally, other collaborations in which we began by characterizing the cardiovascular phenotypes of other genetically engineered mice have revealed unique models of diastolic cardiac dysfunction and cardiac hypertrophy (GLP-1R knockout, in collaboration with Dan Drucker, University of Toronto) and vasomotor dysfunction leading to arteriovenous malformations (Endoglin knockout, in collaboration with Michelle Letarte, University of Toronto). These exciting investigations have been spearheaded by our laboratory and may be developed with our collaborators as distinct submissions for peer-reviewed operating grant support.

c) Cardiac applications of radionuclide imaging: In collaboration with Dr. Robert Mark Iwanochko (University of Toronto), I study the role of 99mTc and/or 201Thal myocardial perfusion imaging with Single Photon Emission Computed Tomography (SPECT) in the diagnosis and prognosis of coronary artery disease. In collaboration with Ren-Ke Li and Richard Weisel (University of Toronto), we also employ SPECT to gauge the viability and function of autologous cardiac cell transplants in a pig model of myocardial infarction.

d) Acute cardiac care: At the Toronto General Hospital, I was principal investigator for the

CURE trial (multicentre randomized placebo-controlled clinical trial of Clopidogrel in unstable angina), and for the Canadian Heart Research Centre’s Registry of Acute Coronary Syndromes. We also study the role of acute physiological scoring systems in the prognosis and management of patients requiring cardiac intensive care.
