1) Introduce la estructura de tu work plan PROJECT embraces a forma2ve research approach organised in four synergis.c work packages that focus on coordina2ng and managing the project (WP1), science development (WP2 and WP3) and science communica2on (WP4) tasks (Fig. X). WP1 aims to ensure effec2ve coordina2on and implementa2on of the work-‐plan and efficient resource u2liza2on. Task 1.1. Management. Administra2on and financial management of the Ac2on; prepara2on of reports; communica2on with EC; and visibility ac2vi2es (including PROJECT webpage) Task 1.2. Coordina2on and evalua2on (C&E). Three major C&E mee2ngs are scheduled (months 1, 10, 20), involving researchers from various disciplines from the host and partner ins2tu2ons. The kick-‐off will coordinate foreseen tasks. 1) La estructura de organización y management del proyecto FPS will act as host insEtuEon and will support the candidate through Communica2on, Legal, Intellectual Property and Transfer Technology Departments and Interna2onal Research Projects Office belonging to the Network of Research Management Founda2ons of the Andalusian Public Health System. The host ins2tu2on will support the candidate in the management of the project, in the integra2on of knowledge and results arising from research. According to this, the procedure to manage this grant will be the same followed with other research grants hold by Bionand scien2sts. While Bionand develops the research project and training, FPS ensures the fulfillment of the financial and scien2fic commitments, and the implementa2on of the project following the Marie Curie Grant Agreement clauses and specific provisions.

Following alternaEve soluEons are proposed to overcome these issues: Aim 1: Developing magne1c nanopar1cle with specific DNA sequences and detec1on by PCR requires a very heat-‐stable nanopar1cle plaOorm. The use of a clinically approved NP (Feraheme) coated with carboxymethyl-‐dextran have proven this thermal stability requirement and will be used in the design of the next genera2on NPs. NPs will be used in isothermal PCR with the proper adjustments and in collabora2on with Biohelix company. Although sensibility of PCR is amongst the highest in molecular biology field, sensibility of the MRSw can be further increased with higher size NPs (above 50nm). Synthesis and thermal analysis of big NPs will be executed and tested in both, regular and isothermal PCR enviroments

The proposed project is based on the experience of the applicant and supervisor who work in these topics and believes, awer bibliography revision and a deep discussion, that the project will be a success. However, there are risks that can make endanger reaching project objecEves. Regarding the previous experience, bibliography and study of the project, these risks that can appear during the research are presented here : • Polymer employed for the nanotube surface are not working or are not in stock: The micro/nanomotors that will be synthesized for the AFB assay contain a surface layer made of a carboxi polymer (carboxi-‐EDOT) which is commercial. It is relevant for the following modifica2on steps. If this polymer is not available, the applicant have the knowledge for development this superficial layer of motors with other similar polymers as carboxi-‐pyrrol and changing the electropolymeriza2on condi2ons (Voltages and saline solu2ons). These new motors will have a surface with carboxi-‐groups which is required for modifica2on steps.

We do not foresee technical problems in the basic prepara2on of therapeu2c nanostructures (TNs), in vitro or in vivo experiments, due to the background of the applicant and the excellent exper2se of the host and partner organiza2ons in their respec2ve areas. The criEcal issue of this project is the efficiency of nanomedicines to reach macrophages in vivo and exert a significant funcEonal acEvity. For this reason we have designed at least two different “targeEng” strategies and two “therapeuEc” strategies that have been thoughxully chosen on the basis of the available literature and our own experience. The re-‐educa2on of TAM loading the nanomedicines with ac2vators of TLR7 receptors will be priori2zed. However, an alternaEve approach (WP1.2. ), by means of TNs loaded with chemotherapeuEc drugs, has been considered and incorporated in the proposal. This alterna2ve strategy will be pursued if the TLR7-‐TNs are not effec2ve in vivo, or if their ac2vity is not sufficiently long-‐las2ng. This alterna2ve approach is based on a dis2nct ra2onale, as TNs loaded with chemotherapeu2c drugs are expected to kill, and not to re-‐educate macrophages. Key milestones, intended for the exclusion and selec2on/iden2fica2on of TNs, will be strictly obeyed at each step of the project, thus guaranteeing the appropriate progress and success of the proposal.

The project will be developed at BIONAND (www.bionand.es), which is the Andalusian Centre for Nanomedicine and Biotechnology. This is a mixed centre with stakes held by the Regional Ministry of Innova2on, Science and Enterprise, the Regional Ministry of Health and the University of Malaga with the purpose to promote and encourage the development of an andalusian center for excellence research in Nanomedicine. Bionand was created in 2011 with the help of European Regional Development Fund (ERDF, 11.6Millions €) and Spanish Ministry of Economy and Compe22vity (grants IMBS10-‐1C-‐247, PCT-‐420000-‐2010-‐003 and INP-‐2011-‐23-‐420000-‐ACT2, 11.7Millions €), as a mulEdisciplinary research space, where different professionals from the healthcare, university and business areas interact. Focused in the development of new nanomaterials and its biomedical applicaEons, the center facilitates the genera2on of new diagnosis systems for preven2on and treatment of diseases.

The XXXX is one of the 6 research unit laboratories of the European Ins2tute for Marine Studies, which is one of the most important marine science research centres in France. As will be explained in part B5, the XXXX is strongly pluridisciplinary and units scien2sts from several ins2tu2ons. This will ensure that the fellow gets mul2ple opportuni2es for presen2ng her work and interac2ng with researcher at these neighbouring ins2tu2ons. In addi2on, the mulEdisciplinary approach of the XXXX is essen2al for research on the highly complex interac2ons between components of the marine environment. The unit’s interdisciplinary policy is built on a sound basis of skills in the relevant scien2fic disciplines and an integra2on of this approach in daily ac2vi2es.

With direct relevance to this proposal, the main infrastructures needed consist in computers, specialized sowware and diverse kind of data (e.g. acous2c, oceanographic and biological) available at the host ins2tu2ons. New data will be included in the LEMAR databases awer the planned cruises. The host ins2tu2on will also provide the postdoc with an office space, full access to the network of the ins2tute, as well as to library facili2es such as Internet subscrip2ons to top scien2fic journals. Within the project there are no needs from research in laboratory. Therefore, the infrastructures of the ins2tute, and in par2cular the host group, comprise the equipment and the competence that perfectly respond to the needs of the project. During secondements, the fellow will be hosted at IRD and SPC of Nouméa, and at the XXX of Toulouse and she will benefit from the respec2ve center facili2es (see sec2on 6), including office space, compu2ng and technical resources.

The Max Planck Society has a world-‐leading reputa2on as a science & technology research organisa2on. In 2006, the Times Higher Educa2on Supplement rankings of non-‐university research ins2tu2ons placed the Max Planck Society as number 1 in the world for science research, and number 3 in technology research. The MPI for Intelligent Systems belongs to the Max Planck Society, a German research ins2tu2on strong in basic research. The Max Planck Ins2tute for Intelligent Systems approaches intelligent systems from the viewpoint of learning in systems with percep2on-‐ac2on loops while these systems interact with natural, complex environments. The ins1tute focuses on establishing the scien1fic founda1ons of percep1on, ac1on and learning through interdisciplinary and collabora1ve research across scales and systems.

3.3 Appropriateness of the ins.tu.onal environment
The MPI for Intelligent Systems and the ETH Zurich have launched a research network in the research field of learning systems. Thus, the scien2sts are conduc2ng joint research projects and will hold joint conferences and workshops. They will also have easier access to each other’s research faciliEes. In addi2on, the applicant will join a large, well resourced, mul2disciplinary team led by Dr. XXXX (see hdp://www.com.com). The group comprises some people (PhD student and post-‐docs) from different part of whole world, and includes materials chemists, analy2cal scien2sts, physics scien2sts and mechatronic engineers, as well as admin support, and is well equipped in terms of general lab instrumenta2on, materials, and rapid prototyping facili2es. The team is located in the MPI for Intelligence systems and has full access to specialist facili2es for microfabrica2on, clean rooms, lithographic techniques, surface analysis and visualisa2on. These facili2es offers by MPI and the research mul2disciplinary team which is exper2se in the topic presented here, make them the most properly place for the development of the proposed project.

Outgoing: The Massachuseds General Hospital, where CMIR is located, maintains an Office for Interna2onal Scholars and Students to assist and advise scien2sts like Dr. Alcántara in immigra2on, visa, health insurance and related maders. Upon establishment of the start date, Dr. Alcántara will be given a list of contacts and/or possible accommoda2ons, and will be helped to understand and sign the rela2ve contract. As a rule, the foreign fellow is always supported by a na2ve speaker in all the ini2al interac2on with the USA ins2tu2ons. CMIR has an induc2on week for all new researchers where they are introduced to and receive important and appropriate training in safety (COSHH, GLP, fire, handling skills, disposal prac2ces) and core skills (eg. Record keeping, data handling, experimental/factorial design, IT, databases, analy2cal facili2es eg NMR, Mass Spectroscopy etc).

CMIR encourages researchers to take, at appropriate 2mes during their tui2on, a comprehensive range of courses to develop general skills & awareness e.g., Scien2fic & Report Wri2ng, IT Skills, Ethics & Science, Science & the Media, Library skills, Time management, Teaching Prac2ce, Careers, Research Management, Good supervisory prac2ce. At the beginning of the project, in addi2on to standard departmental induc2on, the researcher will be orientated with respect to the teamworking of the research group, ini2ated in background reading and all laboratory procedures clearly explained. Prac2cal work will start as soon as possible to promote his interac2on with other department and group members. Dr. Alcántara does not have children at the moment; however, CMIR has an internal childcare programme in which the applicant could be enrolled, if required.

Return: Dr. Alcántara will be integrated in the Biofunc2onal Nanopar2cles Group (Dr. Jesus Mar{nez). He will be provided with lab space, students (Ph.D and undergraduate) and infrastructure to achieve with success his project. In parallel to this stage Dr. Alcántara will be encourage and supported to create a spin-‐off enterprise to apply his managing and scien2fic knowledge into the market place. He will be provided with lab space and money enough to start the ac2vity and will be monitored by INA. INA have a specific administra2on helpdesk to facilitate Dr. Alcántara asking for European funding programs (scien2fic or enterprise form). Depending on his results and orienta2on, at the end of this fellowship, INA or University of Zaragoza will accept Dr. Alcantara´s candidature to be part of INA scien2fic staff.

Managing dryland ecosystems addressing their poten2al state transi2ons requires a predic2ve understanding of the interdependent ecological and hydrological factors and mechanisms that govern how these systems func2on, par2cularly under changing condi2ons and pressures. Addressing this need requires a unique mix of competences and experience that PROJECT has integrated. The fellow, Dr. XXX, brings a decade of research experience in experimental ecology. The Supervisor, Dr. YYY, is an interna2onally recognized and regarded scien2st and modeller with highly complementary research interests and skills who has made available the excep2onal facili2es and pooled exper2se represented by the host ins2tu2on, XX-‐CNRS. Her highly produc2ve track record achieved in part through innova2on and highly effec2ve mentoring and project management underscore the appropriateness of the proposed collabora2on.

Moreover, it is clear that Dr. XXX brings to ZZZ-‐CNRS the capacity to conduct forma2ve modelling research 2ghtly coupled to empirical field research and the network necessary through UA (Secondment) to ensure both 2mely research results and successful knowledge transfer. This is the right combina2on to take on such an important and yet challenging ini2a2ve such as PROJECT. The host ins2tu2on will recruit Dr. XXX conform to the principles of the “Code of conduct for recruitment of researchers”, appoint Dr. YYY as her Supervisor during the whole dura2on of the ac2on, and take complete responsibility for execu2ng the proposed ac2on. Both, the Beneficiary and the Partner Organisa2on have a strong track record in suppor2ng Marie Curie Ac2ons, and commit themselves to support the career development and training of the fellow and provide her with the assistance, infrastructure, equipment and products needed for implemen2ng PROJECT as detailed in the proposal.
