The Custom TaqMan ® Array Card is a 384-well microfluidic card that enables you to perform 384 simultaneous real- time PCR reactions without the need for liquid-handling robots or multi-channel pipettors to load samples. This highly flexible card enables 1 to 8 samples to be run in parallel against 12 to 384 TaqMan ® Gene Expression Assay targets (including a manufacturing control). The assays are preloaded into each well of the card and are ready for use immediately upon delivery. Choose from over 50,000 inventoried TaqMan ® Gene Expression Assays designed for human, mouse, and rat genes to be loaded onto a Custom TaqMan ® Array Card (Figure 1). For added flexibility, you can contact your local Life Technologies sales representative to order cards loaded with Predesigned made-to- order or Custom TaqMan ® Gene Expression Assays. The Custom TaqMan ® Array Card is designed for use on the state-of-the-art QuantStudio 12K Flex, or ViiA 7 Real-Time PCR Systems when equipped with TaqMan ® Array Card blocks. Ultimate microarray validation tool TaqMan ® Array Cards are exactly the right tool for validating the tens or hundreds of hits generated from microarrays because they can be customized to include up to 384 of those hits in one easy-to-use card. Using individual TaqMan ® Assays, or even SYBR ® dye–based assays, to look at 12, 48, or 96 targets can quickly become unmanageable and expensive. TaqMan ® Array Cards enable researchers to accomplish the validation necessary to arrive at the right answer easily and affordably. Create the perfect cardYou simply select TaqMan® Gene Expression Assays and the optimal Custom TaqMan® Array Card format for your experiment, and we will deliver your TaqMan® Array Card preloaded with your selected assays. Choose 12 to 384 target assays from our collection of inventoried TaqMan® Gene Expression Assays covering human, mouse, and rat genes. Ordering is easy with the online Custom TaqMan® Array Card configuration tool, which helps you find and select genes and assays. Custom TaqMan® Array Cards are available in 10 different formats covering 12, 16, 24, 32, 48, 64, 96 (2 choices), 192, and 384 assays.

Designing a Custom TaqMan® Array CardCustomizing a TaqMan® Array card through the Life Technologies website at lifetechnologies.com/arraycards is simple. Choose your array format and method for choosing TaqMan® Gene Expression Assays, select a quantity, and place your order—it is as easy as that. Online selection of TaqMan® Assays is simple: search by using common keywords like Entrez gene symbols, RefSeq accession numbers, or UniGene IDs (Figure 2A). You can narrow your search results using the Target Class filters relevant to your research area (Figure 2B). For example, if you want to find TaqMan® Assays for caspase genes in the area of apoptosis, select the “Apoptosis” target class filter and enter “caspase” in the Keyword Search box to return results for caspase genes (Figure 2C).

Additionally, TaqMan® Gene Panels, which are predefined gene panels of TaqMan® Gene Expression Assays corresponding to a biological pathway or target class, can be used to simplify configuration of a Custom TaqMan® Array Card. The TaqMan® Gene Panels are accessible through the online Custom TaqMan® Array Card configurator.

Predesigned gene signature cards are also availableIf you do not have the time to compose a list of assays, use one of our predesigned TaqMan® Array Gene Signature Cards. TaqMan® Array Gene Signature Cards contain select TaqMan® Gene Expression Assays for focused gene sets for many important target classes and pathways. Genes in each of the arrays have been culled from pathway analysis tools, published review papers, and collaborator and customer input. Select from a variety of TaqMan® Array Gene Signature Cards covering areas such as GPCRs, immune response, or protein kinases at our website at lifetechnologies.com/signaturecards. You can also use our Endogenous Control Arrays to assess which housekeeping genes are best for your specific study.

Simple and straightforward loadingOnce you receive your TaqMan® Array Card, using it is fast and simple. The TaqMan® Array Card has eight sample-loading ports, each connecting to 48 reaction wells. Simply combine 30–1000 ng of cDNA sample with TaqMan® Universal Master Mix II (for each port), pipette sample mix into the TaqMan® Array Card, centrifuge briefly to fill the wells,

and seal the card. Within minutes, your 384-well TaqMan® Array Card is ready to run on the QuantStudio™ 12K Flex, ViiA™ 7, or 7900HT Real-Time PCR System (Figure 3). Imagine how much time and costs you can save by not having to set up individual reactions, as well as reducing the the risk of introducing variability. Accurate, quantitative gene expression results are available when thermal cycling is complete. The reduced number of pipetting steps streamlines the reaction setup process and eliminates the need for liquid-handling robotics.

Applied Biosystems® real-time PCR instrumentsTaqMan® Array Cards function seamlessly with the Applied Biosystems® QuantStudio™ 12K Flex, ViiA™ 7, and 7900HT Real-Time PCR Systems. The new QuantStudio™ 12K Flex Real-Time PCR System offers unprecedented throughput, flexibility, and scalability and is also capable of running TaqMan® OpenArray® Plates. The ViiA™ 7 Real-Time PCR System also features interchangeable blocks as well as 21 filter combinations. The 7900HT Fast Real-Time PCR System has been used to run TaqMan® Array Cards to generate
