BackgroundGFPisafluorescentproteinisolatedfromseveralorganismsincludingthepacificjellyfishAequoriaVictoria.GFPconvertstwospecificwavelengthsofbluelightintogreenfluorescentlightbyenergytransfer.Proteinsthatfluorescecontaincomponentscalledfluorophores.InGFPthefluorophoreoriginatesfromaninternaltripeptidesequence(Ser‐Tyr‐Gly)whichispost‐translationallymodifiedtothestructureshownbelow,a4‐(p‐hydroxybenzylidene)‐imidazolidin‐5‐one.ItisworthnotingthattheformationofGFP’sfluorophoreisanautocatalyticprocessthatrequiresnocofactororenzymaticreaction.ThisfluorophoreishighlystableevenathightemperaturesandvariouspHconditions.ThegeneforGFPhasbeenisolatedandhasbecomeausefultoolformakingfusionproteinsinwhichGFPislinkedtootherproteinsandfunctionsasafluorescentproteintag.GFPtaggingcanbeusedinawiderangeofapplications:asatracerforcelllineage,asareporterofgeneexpression,orasareporterofprotein‐proteininteractions.GettingstartedwithStarORFStarORFisasix‐frametranslationsoftwaretoolforexploringgenesequences.• TogettoStarORF,pleasenavigatetohttp://web.mit.edu/star/ORF.• ClickontheStartbuttontolaunchtheapplication.• ClickTrustwhenapromptappearsaskingyouifyoutrustthecertificate.• IntheInputbox,pastethesequenceprovidedbelow.
