Secondary electrons (SE)
From sample surface. An inelastic scattering. Energy of the SE are typically < 50 eV (compared with 10-30 kV of the
PE beam) SE yield () = [Number of SE go into the detector] / [Number of PE] SE yields mainly depends on the PE beam energy
Interaction volume
The interaction volume depends on beam energy (Eo) 10-30 keV (normal SEM) 100-300 keV (TEM)
The greater the Z, the bigger the interaction volume
Back scattering electrons (BE) BE = the PE which penetrate into sample surface and undergo several
scattering before travel back through the surface BE coefficient () = [No. of BE that go into the detector]/[No. of PE] mainly depends on the Atomic No (Z)
Comparison between BE and SE yields
SE: smaller interaction volume very close to the surface provide a better resolution more suitable for a surface
topography examination
BE mode is recommended to be
used for a compositional contrast image.
Image contrast
There are 2 main types of contrast:
Compositional contrast (or atomic number contrast)
Topographical contrast
Both types of contrast occurs simultaneously. This means that even pure element can also produce contrast if the surface is not smooth and the detector can detect SE.
Image contrast from BE
1. Different atomic number (Z) provide a compositional contrast
High Z element = High = Appear bright
Lower Z element = Low = Darker area
2. Trajectory effect provide a topological contrast Trajectory effect = effect of travel direction of BE on BE
yield
Topological contrast = The contrast attributed to the difference in surface shape

Neither SE nor BE detectors are 100% efficient (not all electrons are detected)
Depends on an orientation of the sample to the detector

About the LaB6 electron gun
Normal operating T ~ 1800 Kelvin, which provide a current density
of 40 A/cm2 (compare with 3 A/cm2 from tungsten obtained at 2700 K).
LaB6 is reactive (tends to be oxidized) and thus require a better vacuum (~10 -5 Torr).
LaB6 is naturally available in fine grain form and need to be fabricated into rod or block form. The fabrication process is difficult and thus the LaB6 emitter is expensive.
About the field emission gun
A very sharp tip (radius < 0.1 m) tungsten needle wire
The electric field at the tip is very strong (> 107 V/cm) due to the sharp point effect.
Electron are pulled out from the tip by the strong electric field (without requiring any thermal energy).
Ultra-high vaccum is needed to avoid ion bombardment to the tip from the residual gas.
Electron probe diameter < 1 nm is possible
Brightness
The current density is related to brightness of the electron beam.
The greater the brightness, the more electron can be focused.
To increase the brightness; Use a higher temperature (at the expense of a shorter
filament life) Use a higher accelerating voltage (but risk of sample damage
and lower SE yield) Using a higher brightness electron gun (the material with a
lower work function).

BE detector
The specimen was prepared by staining
SE detector
BE (too energetic to be deflected by a 200V potential) will also produce a signal in the ET detector.
BE detector (wide angle scintillator)
This detector has a similar concept to the ET detector. However, there are some differences including the following points
The detector stay on top of the specimen
Scintillator surface is larger
The positive bias voltage and the Faraday cage are not necessary.

Characteristic X-ray

Family of X-ray lines
For carbon (Z= 6), there are 2 electrons in the K shell 4 electrons in L shell
Only K x-ray are produced (0.277 keV)
For Sodium (Na, Z = 11) 2 electrons in the K shell
8 electrons in L shell
1 electron in M shell
K (1.041 keV) and K(1.071 keV) x-rays are produced
L X-rays of Na are also possible but those cannot be detected due to their low energy.
K might be splitted into K1 and K2 This is because the L shell is further divided into sub-shells
X-rays detector
The detector crystal is sensitive to visible light, it is necessary to block the visible light using opaque window.
The most common window is Be. The material will absorb low x-ray energy generated from light elements (Z < 11, Na).
A window made from plastic can be used for x-ray from lighter element (Z ~ 3)
The x-ray window needed to be maintained at low temperature to reduce noise (from thermionic emission of carriers from the crystal)

Why Si(Li)?
Si is a kind of semiconductor
X-ray from the specimen interact with Si detector crystal, producing: Photoelectron in the conduction band
Hole (in the valance band)
On average for a Si crystal, 3.8 to 3.9 eV are dissipated per hole created. Thus, X-rays with energy >1 keV make many holes

Moseley's Law describes the relationship between atomic number and energy of a spectral line
For a certain element (fixed Z), energy of K-line > L line > M line

Overlap
Moseley’s law says that each element has a unique set of characteristic lines.
But the limited energy resolution of the Si(Li) detector is in many case unable to distinguish between peaks which have very similar energies.
Recognizing and dealing with overlaps is important for qualitative analysis
Guideline for reading X-ray spectra
1. แปลผลยอน จากขวาไปซาย (Working [interpretation] backward).
2. เรมจากการดวา พคแรกนาจะเปน K ของธาตใด (Start with K line).
3. เพอใหแนใจ ลองพจารณาหาพคเลกๆ ทางขวา ซงนาจะเปน k ของธาตเดยวกน (Keep looking for a tiny k line of the same element).
4. จากนน อาจจะ ลองพจารณาหา X-ray line อนๆ ของธาตเดยวกน ทางดานซายมอถดลงไป (Find and mark all other family members (L lines, M lines) of the same element).
5. เมอครบแลว เรมพจาณาความเปนไปไดทจะเจอ k ของธาตชนดอนๆ (Identify next (second high energy) peak for other element).
K-lines
Highest energy (most right hand side).
K and k come together as a pair with a ratio of 10:1 (statistically).
Symmetry shape
For low Z elements (e.g. < Ca), K and k lines may not be resolved.
L-lines
Often occur in groups. Peaks are not well resolved.
Un-symmetry shape. (L/L) ~ 3/1.
M-lines, N-lines and so on .. are very complex and poor in resolution. Avoid use if possible.
Ambiguities
A line at 4.94 keV could be the K line of Ti or K line of V.
In cases like this look for other family lines (big Ti K at 4.51 or tiny V K at 5.43) which are separated.

Estimation of section thickness
The approximate thickness of sections can be estimated by observing their interference colors as they float on the liquid in through.
Peachey (1958) measured the thickness of methacrylate sections with an ellipsometer and established the following scale:

Some comments for the staining with OsO4
1. OsO4 is available either as crystals or readily dissolve in water (~ 2% aqueous solution)
2. When staining a block of material in an aqueous solution (by immersion), the reaction is slow, often take days to weeks, depending on type of the polymers.
3. OsO4 can be sublimed. The high vapor pressure of OsO4 make vapor staining of a specimen possible
4. However, toxicity and high vapor pressure of the agent makes it very dangerous. Care must be taken to handle this material in hood with good ventilation
2 steps staining of poly(meth)acrylate by using hydrazine/OsO4

• “Hydrazine cleaves the ester groups of polyacrylate to produce hydrazide which subsequently reduce OsO4 to a lower oxidation state”.
