Histones and Modified HistonesHistones are basic nuclear proteins that are responsible for the nucleosome structure of the chromosomal fiber in eukaryotes. Nucleosomes consist of approximately 146 bp of DNA wrapped around a histone octamer composed of pairs of each of the four core histones (H2A, H2B, H3, and H4). The chromatin fiber is further compacted through the interaction of a linker histone, H1, with the DNA between the nucleosomes to form higher order chromatin structures.

DNA Methyltransferase Enzymes (DNMTs)DNA methylation is a major epigenetic modification in the genome of higher eukaryotes. The effects of DNA methylation on cells include transcriptional repression by methylation of promoter regions, formation of compact chromatin structures, X chromosome inactivation and imprinting control. Histone Modification EnzymesIn a variety of DNA-dependent reactions such as transcription, replication, recombination and DNA repair, nucleosome-based chromatin structures are regulated by histone modification enzymes. Histones are subjected to numerous post-translational modifications. There are at least eight distinct types of modifications found on histones. Modification enzymes have been identified for acetylation, methylation, demethylation, phosphorylation, ubiquitination, sumoylation, ADP-ribosylation, deimination, and proline isomerization.

Histone Modification Enzymes Histone DeacetylasesThese enzymes are a class of enzymes that remove acetyl groups from an -N-acetyl lysine amino acid on a histone. Lysine acetylation is a reversible post-translational modification of proteins and plays a key role in regulating gene expression. Lysine acetylation and deacetylation preferentially targets large macromolecular complexes involved in diverse cellular processes, such as chromatin remodeling, cell cycle, splicing, nuclear transport, and actin nucleation.
