Flow Check®

Flow Check® Particles and Compensation Particle SetsFlow cytometers are complex instruments that require the fluidic system, excitation optics and detection optics to be in optimal working order. The precise alignment of these systems insures the highest levels of sensitivity and repeatability. In addition to optimizing the overall performance of the flow cytometer, it is also necessary to set up the instrument for the particular type of samples being analyzed. To facilitate these processes, Polysciences offers a wide range of fluorescent microspheres for flow cytometry calibration and set up that contain the actual dyes used in antibody conjugations. These particles are “surface dyed” and will closely mimic cells that have membrane bound antibodies conjugated to dyes.

Rhodamine Intensity SetComposed of 6µm particles labeled with four levels of fluorescence intensity: Low-Brite, Med-Brite, High-Brite and Maxi-Brite. These particles can be used for detection in channels FL2 and FL3 of the flow cytometer. The Flow Check® Rhodamine Intensity Set is supplied with an undyed particle for comparison. All particles are supplied at ~1x107 particles per ml (5ml per component) in an aqueous suspension.

Flow Check® Microspheres & KitsFlow Check® beads have been designed to assist flow cytometry users in checking the suitability of their instruments and their procedures. Particles are made from a polystyrene base and stored as an aqueous suspension. Components of the Flow Check® kits are also available individually.

Flow Check® Sulforhodamine Intensity Set Composed of 6µm particles labeled with four levels of fluorescence intensity: Low-Brite, Med-Brite, High-Brite and Maxi-Brite. These particles are specific for green excitation and detection in the FL3 channel of the flow cytometer. Flow Check® Sulforhodamine Intensity Set is supplied with an undyed particle for comparison. All particles are supplied at ~1x107 particles per ml (5ml per component) in an aqueous suspension.

Ruby Red Fluorescent Microspheres 6µm narrow distribution polystyrene particles that can be used for flow cytometry applications where fluorescence emission in the range of 600nm to 710nm is needed. Particles are supplied at an approximate concentration of 5x106 microspheres per ml in a convenient dropper bottle. See Technical Data Sheet #624. Packaging: 5 ml

• Excitation maximum: 475nm• Emission maximum: 663nm• Suitable for 488nm excitation, detection in FL3 and 633nm excitation,

detection in FL4, APC, APC-Cy™7• Check and adjust instrument settings, check instrument sensitivity,

YG Kit 2.0 Particles in this kit allow the comparison of various levels of green fluorescence from uniform polystyrene beads. Kit contains one bottle each of a full intensity bead, two intermediate intensity beads and an unlabeled bead. All particles are supplied at ~1x107 particles per ml (10ml per component) in an aqueous suspension. The CV’s of these particles offer one of the best combinations of narrow size and fluorescence CV control.

Absorption spans approximately 380nm to 520nm.Emission was recorded at 488nm excitation.

Flow Check® BB/YG Compensation Kit Contains two types of 0.5µm particles supplied at ~1x109 particles per ml (10ml per component) and is designed for checking the suitability of systems using both UV excitable dyes with blue emission and 488nm excitable dyes with green emission. The small size has been chosen to coincide with the use of Hoechst dyes for the detection of bacteria. The green particles, which mimic FITC fluorescent signals, are similarly sized and can be separated when using blue or green detection parameters.
