There are several ways to tackle transportation, one is to hire a car and driver so you can focus on wine tasting and enjoying the sights. We highly recommend Daniel Agnello and his cadre of drivers, which charge approximately U$100 to 150 per day. If you are looking for a more adventurous time with limited street signs and maps that may or may not have been created during the Spanish invasion, then renting a car is a good option.
Within the city, you can easily travel by taxi which are black and yellow and are clearly marked. You can also hop in a remis, which are unmarked taxi's that look like normal cars with a small antennae and a yellow number on the trunk.
If you choose to rent a car, be aware: By U.S. standards traffic in the city is fast and aggressive. Stoplights are normally followed, stop signs are not and many intersections have neither. Generally speaking the car coming from the right has the right of way. So please be careful.
