Procedure (frozen sections)1. Fixation: 5 min in 4% PFA2. Hydration: rinse in cold PBS or distilled water3. Hematoxylin-Eosin staining:10 min in Harris Hematoxylin solution2 min of dipping in warm tap water bath1-2 dips in bluing solution (lithium carbonate)2 min of dipping in warm tap water bath2 min 80% ethanol5 min Eosin4. Dehydration4 min 90% ethanol bath (dips periodically)4 min 100% ethanol bath (dips periodically)9 minutes xylene5. Coverslip with Permount (Toulene based)Allow slide to sit for a few hours to allow curing to completeSlide may be imaged with microscope after cover slipping. Nuclei should be stained dark blue and cytoplasm should be stained pink.
