Procedure (frozen sections) 1. Fixation: 5 min in 4% PFA 2. Hydration: rinse in cold PBS or distilled water 3. Hematoxylin-Eosin staining: 10 min in Harris Hematoxylin solution 2 min of dipping in warm tap water bath 1-2 dips in bluing solution (lithium carbonate) 2 min of dipping in warm tap water bath 2 min 80% ethanol 5 min Eosin 4. Dehydration 4 min 90% ethanol bath (dips periodically) 4 min 100% ethanol bath (dips periodically) 9 minutes xylene 5. Coverslip with Permount (Toulene based) Allow slide to sit for a few hours to allow curing to complete Slide may be imaged with microscope after cover slipping. Nuclei should be stained dark blue and cytoplasm should be stained pink .
