1. General Description
1-1 About This manual
1-2 Warranty and Liability
We warrant the product you have purchased for one calendar year after the date of purchase. In case
of any malfunctions caused by the manufacturer during this period of time, ASTEC will be responsible for
repair or replacement of failed parts or even the instrument itself as circumstance allows for free of
charge.
However, this warranty excludes the following conditions.
Any damages due to fire, earthquake, thunderstorm, and other catastrophic events and pollution and
abnormal electrical supply.
Any damages due to unduly repair, adjustment, calibration and modification.
Any damages due to unduly usage or mishandling.
Any damages caused by moving, dropping, transportation of the instrument.
Replacement and repair of expendables and consumables.
For service, please contact our agent that you have purchased the instrument from.
1-3 Warnings
Connect the instrument to a line plug according to local or county standards and to the specifications.
Do not use the instrument for different purposes from those shown by the manufacturer. (See Chapter 1
for General Descriptions)
Do not switch on the instrument without having waited at least 20 seconds after switching off.
Do not open the instrument before reading the servicing specific conditions described in the manual.
Follow instruction contained in the manual in case of changing any parts
To move the instrument, at least two persons are required. We suggest to contact the distributor or
directly to the manufacturer for servicing.

1-4 Warning Labels
AR-3100GC has an intrinsic danger due to the structure designed for the purpose. For the safe use of
the instrument, the following mark is labeled on the instrument. Please use this instrument under a
caution indicated on the labels.
Power inlet on the back of the incubator is labeled with the Warning sticker.
The following symbols are used in this manual.

Ventilate the room in which you use carbon dioxide with the instrument. It can harm your
body without adequate air ventilation. Do not breath carbon dioxide directly.
Do not place flammable objects in the incubator, such as alcohol, benzene, ethanol or
propane gas. Especially when you use oxygen, do not use matches or lighters or
cigarettes into the laboratory.
Do not use this instrument outdoors. It could cause an electrical short or a lethal shock.
Ask the manufacturer or your sales agent for the setting up. This will prevent from the
events such as gas leaks, water leaks or any other unexpected problems caused by
mishandling.

The location where the instrument will be placed must be steady, firm and leveled.
Do not leave this instrument in humid or wet conditions. It could cause an electrical short.
When double-stack the incubators, use the optional brackets. The top incubator may fall
by earthquake or other unexpected cause or accidents.
Connect only to a suitable power source specifically assigned for the instrument(s)
Using the instrument outside of Japan may require a transformer. Please ask the
manufacturer or your sales agent.
The power plug should be inserted all the way into the power outlet. Make sure that the
power plug is clean and dust-free when used.
Make sure that the instrument is securely earth-grounded.
Do not earth the instrument to the gas pipe, water pipe and lighting rod. This will cause
an electrical shock.
Never use the flammable objects with the instrument.
Make sure the type of gas and appropriate gas pressure are used with the instrument. All
the gas connections with gas tube(s) must be firmly connected and tightened with
cable-tie.
Do not plug any wire or pin into gas ports, sampling port.
Toxic, pathogenic and radioactive substances should be kept away from the instrument.
During maintenance and cleaning, make sure that power off the instrument and unplug the
power code.
Do not take apart or modify the instrument without first-hand supervising.
In case of malfunctions, turn off the power switch in the back of the instrument, and then,
unplug the power cord.
When the incubator is disposed, please be concerned for environment safety.

Pedestal, bench, desk or table where the instrument will be set up MUST be solid and
leveled even to prevent leakage of the water in the Jacket.
Using the correct power source specifically assigned for the instrument is extremely
required.
Do not unplug the power cable with soaked hands.
In case any cuts or worn-out was found on the power cable of the instrument, stop using
the cable and replace it with brand-new power cable.
Do not operate the instrument with soaked hands.
Do not put heavy objects or bottles that contain liquid.
Do not get on the instrument.
When close the inner and outer doors please hold the knob and handle in appropriate
manner to avoid pinching your hands or fingers.
Do not intentionally step on, bend or twist the power cable.
Do not lean against the instrument.
Do not lean against the door of the instrument, especially when it is open.
Use appropriate gloves when cleaning the instrument, especially for cleaning the chamber.
Do not inhale any chemicals, organic compounds, dust or moisture when cleaning.
Power off and unplug the power cable when moving the instrument to different location.
Remove the water from the water pan when moving the incubator to the different location.
In case the instrument will be unused for a long period of time, remove the water in the
water pan and dry out the chamber completely.
Do not splash the water to the outside of the instrument especially on the top of it.
The receiver of O2 sensor should be kept away from condensation.
Used O2 sensor should be properly discarded in order to avoid contamination of the natural
environment.

2. Bio Hazard
If the product has been used with any substances that may be hazardous to human health, and it needs
to be sent back for maintenance, the product must be completely and thoroughly decontaminated before
being sent back.
3. Important Notice
(Read this section to the end prior to use for your own safety)
AR-3100GC requires separate CO2 and N2 cylinders. Premixed gas (ex. CO2: 5% O2: 5% N2: 90%) is
prohibited.
A power outlet should be solely assigned to the incubator. Use of power strip or same kind of
connections is not recommended.
Acid, benzene, soap, toothpaste are not recommended to use for cleaning.
Use of brush to any part of the incubator for cleaning is not recommended.
Do not wipe the plastic and rubber area with volatile liquid such as benzene. Use neutral detergent and
wipe off completely with water.
Do not clean the incubator by poring water (especially on top panel) and wiping it off.
Pure distilled or even cleaner water is recommended for the humidification in the chamber in order to
avoid any chance of contamination. Check the volume of remaining water at least once in 3 days.
Water for the humidification must be replaced or refilled as needed. The water should be pre-warmed
to the same temperature as the set temperature of the incubator.
Pedestal, bench, desk or table where the incubator will be set up MUST be solid and leveled.
Do not place flammable substances and volatile compounds in the incubator, such as alcohol, benzene,
ethanol, adhesive or propane gas. Especially when you use oxygen, do not use matches or lighters
and/or smoke cigarettes into the laboratory.
CO2 cylinder, which is equipped with siphon inside, cannot be used with the incubator.
Gas pressure to supply into the incubator should be maintained in the ranges between 0.03~0.05Mpa
(0.3~0.5 bar / 4.35~7.25 PSI).
In case the incubator will be kept powered off, remove the water for the humidification and dry out the
chamber completely.
The incubator MUST be properly earthed to ground.
Worn out O2 sensor should be properly discarded in order to avoid contamination of the natural
environment.

9. Prior to Installation
� The place (bench, table or stand) where the incubator will be set up MUST be steady, firm and
leveled. Make sure the surface supports the weight of the incubator in operational conditions.
� To move the incubator, two or more persons are needed.
� Please keep 10 inches or more of clearance in the back and both sides of the incubator.
� Avoid placing an incubator where cold air drifts directly to the incubator such as air conditioning or
air vent. It may cause condensation in the chamber.
� All the surface of the chamber should be cleaned and disinfected before operation.
� The incubator should be placed in a clean room environment.
When placing the drawer type incubator on the optional base stool (P/N: AR-3100STD) balance the level
of incubator adjusting the height of the foots located at each bottom corner of the incubator and secure it
to the shelf using M6 screw bolts included in the base stool package. Drawer Type incubator has
dedicated threaded holes on the bottom surface where indicated by red circles in the below diagram.
9-1 Initial Cleaning and Disinfecting
Cleaning and disinfecting the incubator before its use are critical steps. The initial process and
subsequent maintenance should be done properly and following the steps included in this manual.
The incubator should be cleaned and disinfected with substances that can be removed with proper
rinsing. Any toxic substance or its residue could be biologically toxic which potentially affecting the
culture conditions end specimens development.

9-2 Procedure
Wipe all incubator surfaces (internal and external) with a 3% solution of 7X detergent in distilled water
using a damp sponge or a soft cloth (Kim wipes).
Rinse with distilled water using a damp sponge or soft cloth.
Wipe all surfaces (internal and external) with OH Peroxide solution (3%).
Rinse with distilled water using a damp sponge or soft cloth.
Dry the incubator chamber and all incubator parts with a lint-free soft cloth.
Wrap aluminum incubation block for sterilization (use steam sterilizer or oven only - DO NOT USE
ETYLHENE OXIDE METHOD for this purpose).
Once the sterilization of the incubator parts is completed and the parts are at room temperature open the
wrapping and install it.
The power outlet
Please double check for the appropriate power voltage suitable for the IVF CUBE you have purchased.
The power outlet must be individually assigned to the unit; we do not recommend multiple connections
for any circumstance.
The product must be properly grounded to the earth to avoid electric shock when short circuit.
Connect the incubator to a grounded dedicated circuit only. The power cord connector is the
main disconnect device for the incubator. Position the unit so that it can be easily disconnected.

10. Humidity for the Incubation Chamber
10-1 Filling the Humidity Slot
Water in the humidity water slot should be distilled water and supplied in the slot with approximately
20ml of the water. Humidity water, because it is stagnant, can be common source of contamination
within the incubator. So we strongly recommend that the humidity water should be replaced once in at
least 3 days or more often, and the water slot should be autoclaved as often as possible.
It is also recommended to warm up the humidity water to about 37C before it is poured into the slot for
faster evaporation.
When the incubator is powered off more than 30 minutes, make sure to remove the water
from the chamber. When incubator is powered back up, make sure there is NO humidity in
the chamber. Humidity can harm CO2 and O2 sensor when incubator is powered off.

11. Connecting gas
11-1 Connecting CO2/N2 Gas Supply
The Gas port is located on the back of the incubator as shown in the figure below. Connect plastic
tubing (supplied with the incubator) and firmly tie them up with cable ties or clamps. The other end of
the tubing needs to be connected to gas regulator of gas supply tank or gas manifold.
11-2 Gas Pressure
The optimal gas supply pressure is approximately 0.03 Mpa (0.3 bar / 4.35 psi).
Excess gas pressure may cause malfunction and/or damage of the incubator.

12. Main Power
Main power switch is located in the back of the unit as shown in the figure below. Make sure that the
AC cable is plugged into the power outlet assigned individually for the incubator, then, turn on the unit.
13. Setting up AR-3100GC
Gas-Stop Mode
The Gas Stop Mode is recommended for setting up the incubator for the first time, or setting it up again
after a complete cleaning and disinfection process, or after a long-time unused period.
Gas-Stop Mode: Press and hold the SET key on the control panel for about 5 seconds.
The ON/OFF control of Gas Stop Mode is independent for each chamber.
During the Gas Stop Mode, the Temp display shows the actual temperature and “G.ST” intermittently,
and both CO2 and O2 solenoid valve operations completely stop but warm up the chamber(s) in order to
bring the temperature and humidity to the stable condition before the gas supply.
The Gas Stop Mode needs to be turned off manually. To return the chamber(s) into normal operation,
press and hold the SET key on the control panel for about 5 seconds while the chamber(s) is in Gas Stop
Mode.

14. Gas Concentration Check
CO2 / O2 Concentration check
The CO2 and O2 gas concentration must be checked frequently using an analyzer. It is always
recommended to use an analyzer that has air-circulating function between the device and the incubator
to minimize the environmental change in the incubator during the measurement.
There are 2 sampling ports on each drawer, and either port can be used for sampling. The left
sampling port is covered by a cap. Remove the cap and connect one of the ports to the sampling port
of the gas analyzer, and connect the other port to the outlet port of the gas analyzer. Let the analyzer
read the concentration continuously until you finish the calibration. Ambient air remaining in the analyzer
may dilute the chamber concentration and the display values slightly drop from the current values. Wait
about 2 minutes for both incubator’s display and analyzer’s reading to become stable and compare the
difference in the concentration.
If the displayed CO2 and/or O2 concentration differs from the concentration measured with calibrated
analyzer, adjust the concentration on the display of the incubator to the actual concentration measured
with the analyzer. For the CO2 display adjustment, change the CO2 Span parameter to the same value
as that of actual CO2 concentration at A-4 menu. For the O2 display adjustment, calculate the
difference between the two values (actual O2 concentration - O2 display) and add this number to the
current value in A-6 menu. Refer to page 22 and 23 for more detail of adjustment menu.
Note: It is strongly recommended to use an analyzer which can circulate air between the incubator and
the device to prevent significant gas concentration change in the drawer chamber.

Sampling Mode: <UP> key pressed for 5 seconds *Factory Use Only
During the Sampling Mode, Temperature display shows “SP” and the current temperature for 3 minutes.
Concentration check must be done during this period. After 3 minutes, the Sampling Mode
automatically cancels and goes back to normal function.
15. Drawer type Incubation Chamber
Circulation of the gas in the Chamber
Regardless of the lid of the drawer is opened or not, once the drawer is pulled out and pulled back in,
AR-3100GC starts circulating the gas in the drawer chamber
As soon as the drawer is pushed back in, it starts circulating the gas in order to mix the gas in the
incubation chamber and CO2 sensors (and tubes). This helps the sensor to detect the correct gas
concentration and much faster concentration recovery. The gas circulation continues for 1 minute, and
during this circulation, O2 display shows “CYC”. During this 1 minute period gas supply into the
chamber is stopped.
16. Automatic Calibrations
AR-3100GC performs calibrations on CO2 and O2 sensors once in every week. During the calibration
of CO2 sensor, CO2 display shows “CAL” and actual concentration alternately. When O2 sensor is
calibrated, O2 display shows “CAL” and actual O2 concentration alternately.

17. Basic Settings
Every operation and parameter can be changed by using <SET> key on the front control panel. Using
<UP> and <Down> cursors allows changing the value for the settings or changing the parameters.
After changing the value or parameter, if the operation is left unchanged about 7 seconds, it goes back to
the normal display, which indicates the current condition of the incubator.
17-1 Temperature Setting
Press <SET> key once, the TEMP SET indicator lights on. Use <UP> and <DOWN> cursors to set the
temperature to the value you wish to set.
Press <SET> key once more, the control panel brings you to Over Temp Setting. If the control is left
unchanged for about 7 seconds, it returns to normal display.
17-2 Overtemp Setting
Press <SET> key until the Over temp set point lights on. Use <UP> and <DOWN> cursors to set the
temperature to the value you wish to set. In case the incubator reaches the temperature you set here in
the chamber, it will activate the Over Temp alarm.
For detailed descriptions for Over Temp Alarm, see the Alarms section on Page 24.
Press <SET> key once more, the control panel brings you to CO2 concentration setting. If the control is
left unchanged for about 7 seconds, it returns to normal display.
17-3 CO2 Setting
Press <SET> key until the CO2 set point lights on. Use <UP> and <DOWN> cursors to set the CO2
concentration you wish to set.
Press <SET> key once more, the control panel brings you back to the Temperature Setting.
If the control is left unchanged for about 7 seconds, it returns to normal display.
17-4 Gas Selection
Always choose N2 by Use <UP> and <DOWN> cursors
17-5 O2 Setting
Press <SET> key until the O2 set point lights on. Use <UP> and <DOWN> cursors to set the O2
concentration you wish to set.
If the control is left unchanged for about 7 seconds, it returns to normal display.

18. Adjustment and Calibration
Adjustment / Calibration Mode is utilized for modifications to Temperature and CO2 concentrations by
changing factory pre-set parameters as needed. The modifications should be made accordingly to
adjust to the environment in which the incubator is used.
Some of the pre-set parameters should be modified only by a qualified service technician, as they are
critical to overall performance of the incubator. These parameters are indicated here below in this
section.
By pressing <SET> key for about 0.5 sec with <UP> and <DOWN> keys pressed and held, the
Adjustment / Calibration Mode will be activated.
18-1 Adjustment / Calibration Mode:
<UP> + <DOWN> Pressed and Hold + SET pressed for 0.5 sec
Temperature Adjustment (In-chamber temperature) A-1
When Temperature display shows ST1 in Adjustment / Calibration Mode, press SET key to go to
section A-1. You will see a value in this section. Increasing or decreasing the value here allows
you to adjust the actual temperature in the chamber adjusted to the actual temperature.
18-2 Lid Temperature Adjustment A-3
When Temperature display shows ST1 in Adjustment / Calibration Mode, press SET key until you
see section A-3, Increasing or decreasing the value here allows you to adjust the temperature.
For example, when you need to increase the displayed temperature by 0.2 C, decrease the value
shown by 0.2 using <UP> and <DOWN> keys.

This function allows the incubator to carry out manual ZERO Calibration CO2 concentration. As a
default, A-4 shows OFF on the display. Change this to ON with <UP> and <DOWN> cursors and
wait for 10 second until the incubator starts manual calibration.
18-4 CO2 SPAN Adjustment A-5
When measured CO2 concentration is different from the CO2 display concentration and it needs to
be adjusted, change the value on A-5 to actual CO2 concentration by using <UP> and <DOWN>
cursors. It will allow the incubator to automatically adjust the value to the measured value.

18-5 O2 ZERO Adjustment A-6
When measured O2 concentration is different from the O2 display concentration and it needs to be
adjusted, calculate the difference between these two values (A) and change the current value in A-6
by the same amount using <UP> and <DOWN> cursors. After changing the A-6 value, go back to
the normal display and confirm that O2 display shows the same value as that of the measured O2
concentration.

This function allows the incubator to carry out manual ZERO Calibration for O2 concentration. As a
default, A-7 shows OFF on the display. Change this to ON with <UP> and <DOWN> cursors and
wait for 10 second until the incubator starts manual calibration.
Make sure to use a calibrated thermistor when temperature in chamber is re-calibrated.
Do not change the value on A-2 and b1~Gb as these setting are only used by the
manufacturer.

19. Chamber Temperature Calibration
1. Prepare a calibrated thermometer and attach the sensor at the center of the dish plate surface where
indicated as “CAL Point” (Figure A-1). An electronic thermometer with thin lead is recommended.
2. After setting a thermometer in the chamber, close the lid and wait at least 1 hour. Once lid is opened,
it should be left running at least another 1 hour until the temperature is stabilized completely.
3. If the temperature on the thermometer indicates different value from the Temp display, go to "ST1"
and "A-1" for adjustment. Increase the value on "A-1" brings the temperature down, and decrease
the value brings the temperature up. For example, when set temp is 37.0 C and the actual temp is
36.6, you need to set the control temp up by 0.4. In this case, decrease the value by 0.4 in order to
bring up the actual temperature by 0.4.
Lid Temperature Adjustment
When Temperature display shows ST1 in Adjustment / Calibration Mode, press SET key until you see
section A-3, Increasing or decreasing the value here allows you to adjust the temperature.
For example, when you need to increase the displayed temperature by 0.2 C, decrease the value shown
by 0.2 using <UP> and <DOWN> keys.

CAL Point
In case you need to carry out the Calibration more than once, make sure that you have
at least 1 hour between Calibrations for the chamber temperature to stabilize.

20. Alarms
20-1 CO2 Alarm
Press <SET> key in Function suspending mode until Temperature display shows ALA. Then, CO2
display shows ON or OFF. Use <UP> and <DOWN> key to change the setting. This function only
suspends CO2 alarm.
20-2 Overtemp Alarm
OVERTEMP. Alarm activates when the temperature inside the chamber reaches the temperature set on
OVERTEMP setting. The alarm triggers a beeping sound that can be shut off by pressing the
<SILENCE> key. However, OVER TEMP setpoint indicator keeps blinking until the temperature in the
chamber actually decreases. While the alarm is being activated, incubator partially shuts down the
power to all chamber heaters for safety reasons.
20-3 CO2 Alarm
CO2 Alarm activates when CO2 concentration is off ±1.0% or more from the concentration that is set on
CO2 concentration setting for 15 consecutive minutes. The alarm triggers a beeping sound and CO2
Alarm indicator blinks. By pressing <SILENCE> key, beeping shuts down and the CO2 indicator turns
off.
20-4 O2 Alarm
O2 Alarm activates when O2 concentration is off ±1.0% or more from the concentration that is set on O2
concentration setting for 15 consecutive minutes. The alarm triggers a beeping sound and O2 Alarm
indicator blinks. By pressing <SILENCE> key, beeping shuts down and the O2 indicator turns off.

21. Cleaning and Disinfectant
Disinfecting the incubator interior
The incubator should be cleaned and disinfected with substances that can be removed with proper
rinsing. Any toxic substance or its residue could be biologically toxic which potentially affecting the
culture conditions end specimens development. The instructions here included have demonstrated to
clean and disinfect the incubator and its components allowing proper culture conditions as demonstrated
by validation testing and Mouse Embryo Assay (MEA).
Preparation
It is recommended to use gloves, safety goggles as well as a protective coat. Make sure the incubator is
unplugged.
Procedure
1. Turn the incubator off and unplug the power cord from the outlet wall. Shut off the CO2 supply from
the gas cylinder or gas manifold.
2. Open the lid of the incubation drawers and remove water from the slot.
3. Remove the aluminum incubation block out of the drawers.
4. Wipe all incubation drawer surfaces (internal), lids with a 3% solution of 7X detergent in distilled
water using a damp sponge or a soft cloth (Kim wipes).
5. Rinse with distilled water using a damp sponge or soft cloth.
6. Wipe all incubator surfaces (internal), lids with OH Peroxide solution (3%).
7. Rinse with distilled water using a damp sponge or soft cloth.
8. Dry the incubator chamber out with a lint-free soft cloth.
9. Wrap the aluminum incubation blocks for sterilization (use steam sterilizer or oven only - DO NOT
USE ETYLHENE OXIDE METHOD for this purpose).
10. Once the sterilization of the aluminum incubation block is completed and the parts are at room
temperature open the wrapping and install it back in the drawers.
11. Plug in the incubator and turn the incubator on. Pour humidity-water to the water slot with water at
room temperature (or higher temperature) for humidification of the drawers.
12. Initiate a Mode. The incubator will start heating until it reaches the set temperature. The chamber will
increase its humidity gradually. Open the CO2 flow from the gas cylinder or manifold. The incubator
will not inject CO2 until the Set Up Mode cycle is completed.
13. After Set Up Mode is completed (12 hours) the incubator will initiate CO2 injection into the chamber
to a pre set 5%. Adjust the incubator to the desired setting if different than the factory setting (5%
CO2 and 37C).

CO2 Sensor Replacement
� Remove gas control box from the incubator by loosening screws on each side.
� Unscrew 4 screws and pull out the CO2 sensor when you need to replace it.
O2 Sensor Replacement
� Remove gas control box from the incubator by loosening screws on each side.
� Pull out the O2 sensor from the gas control box. The O2 sensor can be replaced without removing
sensor holder and O-rings.
� Place a new O2 sensor to the original position. Make sure that sensor completely fits to the sensor
slot. Gas leakage may occur if the sensor is not properly positioned.

� Remove 4 screws located on the heating surface and remove the aluminum plate. Temperature sensor, heating element and magnet switch for incubation box can be replaced.
� Simply remove the screws that hold the magnet switch for drawer and incubation box when these switches need to be replaced.
� Use a hexagonal wrench to remove the hinge.

23. Warranty Information
We warrant the product you have purchased for two years after the date of purchase. In case of any
malfunctions caused by the manufacturer during this period of time, ASTEC will be responsible for repair
or replacement of failed parts or even the instrument itself as circumstance allows for free of charge.
However, this warranty excludes the following conditions.
Any damages due to fire, earthquake, thunderstorm, and other catastrophic events and pollution and
abnormal electrical supply.
Any damages due to unduly repair, adjustment, calibration and modification.
Any damages due to mishandling.
