Confocal Detection Principle
Confocal Laser Scanning Raman Microscope hasbecome a widely recognized research instrumentin recent years. Confocal microscopy offersseveral advantages over conventional wide-fieldoptical microscopy, including the ability to controldepth of field, elimination or reduction ofbackground information away from the focal planeand the capability to collect serial optical sectionsfrom thick samples. The image of the extendedsample is generated by scanning the focused laserbeam across a defined area.
The pinhole aperture rejects the residual scatteredrays originated from any out-of-focus points on a sample.

Recognition of living and dead cells
Raman spectra of the cell nucleus have significantly lower peaks in dead
cells as compared to living cells. A decrease in intensity is observed in
almost the full range, especially at the peaks of 783 and 827 1/cm and at
the peaks of 1003 and 1337 1/cm, which corresponds to the breaking of the
internal phosphodiester bonds in the DNA molecules and a decrease in
the level of protein expression.

However, in the nuclei of dead cells, lines increase by 1578 1/cm and new
Raman peaks appear at 1607 or 1114 1/cm, which corresponds to Phe and
Try, respectively. These results indicate the degradation of DNA molecules
and the presence of new aromatic amino acids rich in proteins, which
usually occur during cell apoptosis

Hemozoin is an iron-containing pigment present in parasites that cause
malaria in humans. Raman spectroscopy can identify hemozoin in
erythrocyte cells.

on the bottom right one. Different FLIM signals come from different
fluorescent proteins, which produced by e-coligenetically modified
in different ways. Spectrum shape is very similar. Intensity
and lifetime is different. FLIM provides ufficient information to identify
different proteins in bacteria.

The distribution of proteins, lipids, DNA and RNAin HeLa cells
Proteins and lipids were observed in the CARS mode at their characteristic-1 -1vibrations of 2930 cm and 2840 cm , respectively. Nucleic acids, stained
with acridine orange, were acquired in the red (RNA = green) and green
(DNA = blue) fluorescence channels in TPEF mode.

Micro Raman Spectrometer NAµRAMS-01 provides the acquisition of
two images within a single scan: a Rayleigh image (using laser light
reflected from a sample) and a spectral image by Raman scattering.
Ultrafast imaging option allows to get confocal image in 3 sec
(3 μs/pixel).
NAµRAMS-01 uses fast beam scanning by galvano mirrors.
