READY-TO-USE MEDIA 17
The agri-food industry routinely enumerates microbial flora, which indicatesthe microbiological quality of the entire production process, from raw materialsto finished products. Quality indicators are of major importance in establishinga HACCP approach and guarantee the commercial value of food products fromleaving the factory through to the end of their shelf life.Quality indicators enumerated in routine usually are:Total count, total/thermo tolerant coliforms, E. coli, S. aureus,Enterobacteriaceae, yeast & moulds…
Quality indicators
AUTOMATED METHODS
TEMPOTEMPO®
TEMPO® is an automated solution for the enumeration of quality indicators in food. One TEMPO configuration performs up to 500 tests per day. The TEMPO solution iscomposed of two ergonomic works stations:
• The preparation station to inoculate TEMPO cardswith the food sample and specific culture media.
• The reading station to read TEMPO cards and givefinal result in CFU/g of initial product.
It comprises of: • computer with a waterproof mini-keyboard, barcode
scanner and a WIFI card;• intuitive preparation software;• 144 tests capacity media stand;• 4 dispensers for media rehydratation;• TEMPO Filler instrument for inoculating and sealing
the cards;• 4 filling trays;• 12 incubation/reading trays.
It comprises of: • computer with a barcode scanner and a mouse;• intuitive software for reading and managing results;• administration software for managing the
configuration;• TEMPO Reader instrument for reading the cards;• wireless WIFI transmission router;• printer;• bidirectional connection with the lab computer
(optional).
PRINCIPLE
14
Relayed through a wireless WIFI connection, the two stations communicate in real time although they are not in the same room.
Each medium containsa fluorescent indicator basedon the formula of traditional
culture media and bioMérieux’sexpertise in bacteriology.
The TEMPO cardoffers the precision
of the MPN method in 16 tubes.
Ease of use
Using intuitive icons and userfriendly software, TEMPO® is veryeasy to operate and requiresless than two days of training to beroutinely used.
Standardisation
As most of the analytical stepsare automated, TEMPO improvesthe consistency of results fromplant to plant, day to day, countryto country and therefore operator variability is removed.
Time savings
With TEMPO results are one totwo days faster than conventionalmethods (i.e: 24 to 48 h), providing more rapid selection and orientation of raw materials, accelerated corrective actions in case of non compliance, speedier product release, improvedstock rotation…As the routine workflow is fullyoptimized, time is saved for otheractivities such as statistical andtrend analysis, monitoring ofcritical control points, reinforcing ofpathogen control plans or simplyperforming more routine analyses.
Cost savings
The TEMPO card covers severaldilutions, so only one TEMPO testis needed instead of multipleplates to obtain the most relevantresult according to each criteria. It also eliminates time consumingtasks such as media preparation, manual dilutions and identifications.
Traceability
Every consumable item is tracedthrough a barcode identification. The unique card barcode ensuresfull traceability throughout
the analytical procedure, from the sample ID until the final result. All data is saved and can be easilyretrieved for quality assurance purposes.
Innovation
bioMérieux using its experience and know-how in culture media, instrumentation, VITEK® cards and fluorescent reading, has developed TEMPO, the first automated solution for quality indicator enumeration.
15
After distribution of the culture media-sample mixture inthe wells, the TEMPO cards are placed into a normal incubator. The bacteria multiply during incubation and metabolisethe culture media.
According to the number and size ofthe positive wells (fluorescent or non-fluorescent), the TEMPO systemuses statistical methods to calculate the number of microorganisms present in the initial sample.
The finalresult isexpressed inCFU/g

A wide range of culture media, specifically designed and evaluatedfor food testing, is available toenumerate quality indicator microorganisms such as Escherichiacoli, Enterobacteriaceae,Staphylococcus, yeasts/molds, sulfite-reducing anaerobes, etc..bioMérieux is ISO 9001-certified andcontrols all the stages of production, frommedium design (selection to each rawmaterial) through to manufacture, withperfect traceability.By means of rigorous raw material selec-tion, significant control of production andshelf-life validation according to a proce-dure including thermal shocks, bioMérieuxcan guarantee the performance androbustness of its ready-to-use media.
The standardized formulas comply withfood industry standard recommendations(generally NF, EN and/or ISO). They areadjusted and supplemented, as authorizedby the standard ISO/TS 11133-2:2003,to achieve the expected performancelevels and allow satisfactory equivalencebetween lots. Similarly, the quality controlprocedures performed on these mediacomply with the recommendations of the standard ISO/TS 11133-2 and thiscompliance has been evaluated byAFAQ/AFNOR Certification (2006).Chromogenic culture mediaOn the basis of API® technology used todetect enzyme reactions, bioMérieux hasdevised a wide range of chromogenicmedia, named “chromID™”, including aspecific range for food testing.
These media, some of which are standar-dized, offer very easy use and better per-formance criteria than conventional media. The range of chromogenic media intendedto screen for pathogens currently consistsof five media (see page 9).ChromID Coli medium is specifically designed for the simultaneous detectionand enumeration of E. coli and coliforms. It can be used at 2 incubation temperatures(37°C or 44°C) according to a protocolvalidated by AFNOR Certification.

Innovation
API completely revolutionizedthe field of bacteriology. The API range introduced a standardized and miniaturizedversion of existing techniques, which,up until then, were complicated toperform and difficult to read. WithAPI, bacterial identification becamesimple, rapid and reliable.
API 20E was the first identificationsystem to be developed, combininga strip of biochemical tests and adatabase. The range was rapidlyexpanded, in collaboration with leading international reference centres in the USA, Japan,Australia and Europe…
Evolution of the technologiesenabled bioMérieux to improve itsIdentification range with the rangeID 32 strips which further increasedstandardization accuracy in theidentification of microorganisms. The innovative concept of API alsoinvolved the development of a calculation method, known asnumerical identification. This method in turn led to the design of software programs, making identification easierand more accurate.
API®/ID 32API®/ID 32
Today API continues to innovatemaking these interpretation toolsavailable anytime anywhere in theworld with access via the Internetto APIWEB.
The reference
Chosen by microbiologistsworldwide for ease-of-use andhigh-performance, the API range rapidly became the global reference for identification:
• Over 2000 scientific publicationrefer to API products.
• API systems have long beenestablished as the referencetechnique used to assess theperformance of any newidentification arriving on the market.
• API products have led to theidentification of previouslyunknown bacterial species.
• API systems are used to teach microbial identification in many higher education centres through-out the world.
The identification of allthe microorganisms
Today, the API® /ID 32 range is themost extensive range available. It includes 15 identification systemscovering all groups of bacteriaencountered in industrial microbiology laboratories: Listeria, Staphylococci, Entero-bacteriaceae and non fermentingGram negative rods, Bacillus, Lactobacillus… API can identify over 600 different species.
Reliability
Over its 30 years of existence, API has gained invaluable expe-rience in bacterial identification:around 1000 different biochemicaltests in routine use, an in depthknowledge of bacterial metabolismsand expertise in manufacturingtechniques. The scope of thisexperience makes bioMérieux the world leader in bacterial identification. In addition to regularly updating our databases and softwareapplications, we are dedicated toproducing high quality products and providing comprehensivecustomer service.

AUTOMATED METHODS
BacT/ALERT® 3D Microbial Detection SystemBacT/ALERT® 3D Microbial Detection System
Fast, reliable, sensitive, andproblem-free microbial spoilagedetection is possible and is inroutine use in several well-knownaseptic production facilities throughout the world. They are using bioMérieux’sBacT/ALERT® 3D MicrobialDetection System.
With the introduction of theBacT/ALERT, rapid testing andall the benefits it can providehave arrived. BacT/ALERT can beimplemented with relative easeinto any laboratory performingpresence/absence testingof spoilage microorganisms andeffectively improve the quality oftesting for many product types. The BacT/ALERT 3D also reduceshands-on time and eliminates
When a bottle is placed intoa cell in the detection unit, a small light-emitting diode(LED) exposes the sensor to a red light beam. The reflected-light is collected every ten minutes by a photodiode builtinto each cell, where it istransformed and amplifiedbefore being transmitted tothe computer system forinterpretation.
The computer software generatesa growth curve based on production of CO2 and othermetabolic by-products that are

dissolved in the culture media foreach sample. Through the useof complex algorithms, theBacT/ALERT 3D MicrobialDetection System discriminatesbetween constant CO2 and other metabolic by-products production and an acceleratedrate of CO2 and other metabolicby-products production causedby growing microorganisms.
As additional CO2 is generatedwithin the culture bottle, thebuilt-in sensor changes froma grayish color to a lighter yellowcolor. Even the slightest color

false positives, re-testing, lengthysample pre-incubation andinstrument standardization byproduct. There is no longer theneed to run batch controls or todevelop baselines to compensate
for background readings. All of this can be accomplishedwithout sacrificing accuracy andreliability.
Wide range ofproducts can be tested usingBacT/ALERT® 3D
• Different product types can berun simultaneously.
• Culture bottles available for highand low acid products.
• Each bottle is compared to itsprevious readings, not pre-setthresholds determined for eachproduct.
• Measurements not taken directlyfrom media/product mixture.
• No false positives due to initial pHor initial brix/acid ratio of product.
• No need for negative controls runwith each product to establishcutoffs.
• Results easily verified by wet prepor Gram stain performed frombottle.
BacT/ALERT® reduceshands-on time
• Sample bottles already containappropriate media for high or lowacid products.
• Bottles are bar-coded for easydata entry and bottle loading.
• Barcodes permit ease of sampleidentification and tracking.
• No pre-treatment of sample toremove non-microbial backgroundinterference.
• Once inoculated, the bottle isloaded into the instrument andrequires no additional handlingduring testing.
• May reduce staffing requirementsor allow increased productioncapacity without additionalpersonnel.
Large sample volumesoften eliminate the need for lenghtypre-incubations
• Sample size up to 10-20 mldepending on the bottle typeused.
• Large sample volumes provide for20-100 fold increase in sensitivity.
• Media developed to facilitategrowth in high acid or low acidproducts.
• Detection level as low as1 organism per sample.
• Instrument continuouslyincubates, agitates and monitorseach sample.
• Alerts lab as soon as bottle isdetected as positive.
• Spoilage organisms generallybecome positive in 24-48 hoursor less.
Standardization
• No need for negative controlsto set baseline or threshold fordifferent product types.
• Each bottle must pass strict QCprocedures during and aftermanufacture.
• Instrument is automaticallyQC’d regularly to ensure accuratereadings.
• Each bottle is compared to itsprevious readings, not pre-setthresholds determined foreach product.
• Sophisticated computeralgorithms to detect growthanalyze changes in bottlereadings.
change is detected by theinstrument and immediatelytransmitted to the BacT/ALERTcomputer. Since each sampleis compared to its own past performance rather than a preset threshold, true positivesare detected rapidly without anincrease in false positives.
When a sample is determinedpositive, the computer systemalerts the user and a lightappears at the cell of the bottleto be removed.

The BacT/ALERT system utilizesdisposable culture bottles - in both aerobic and anaerobicmedia formulations - for testing avariety of low and high acid foodand beverage products. Each self-contained BacT/ALERTculture bottle is sterile andcontains an internal colorimetricsensor. Caps and labels arecolor-coded for easy identification.
All bottles feature a bar code label, which assigns a unique ID numberand identifies it by media type. While providing a convenienttracking system, these labels alsoreduce data entry and clericalerrors. With the bar code reader, bottles can be quickly and easilylogged into the system, andplaced directly in the instrumentfor monitoring.
