Who said stress could be productive ?In the case of our adipocytes stress in fact goes
with the production…of fat.
Without really noticing it, the externalenvironment is often hostile for our skin.Repeated and daily exposure to exhaustfumes, UV radiation, pesticides, cigarettesmoke…generates oxidative stress in ourskin. It causes the production of freeradicals, veritable cellular missiles whichattack our cells, oxidise proteins andmembrane lipids, degrade DNA, disturbnormal cell function, and accelerate their
ageing.
Free radicals are implicated in fat storage through several ways:
• Alteration of the working order of adipocytes which become insensitive tolipolytic agents (whether produced by the organism or supplied by slimming
treatments)• Stimulation of the production of prostaglandins, especially PGE2 which favours
the production of VEGF, the development of blood vessels and therefore thesupplying of oxygen and fatty acids to adipocytes.
• The decrease in the synthesis of adipokin, especially adiponectin, a moleculeresponsible for the limitation of the expansion of adipose tissue.
At this stage, the adipocytes lose their lipolytic activity and continue to store fatswithout ever hydrolysing them.
Lotusmaritimus
Still called “PeaLotus” (Dragon’sTooth or SeaBirdsfoot Trefoil) itgrows particularly in wetregions near the seaacross all of western Europebut it is also found inland,including in the mountains up to 1600maltitude.
It is a small vigorous plant with a woody taproot and a horizontal stem which thenstraightens up. Its flowers are goldenyellow giving it the name “Golden Slipper”,or “Bride’s Slipper” in France.
Lotus has calming properties, in thepast it was given to reduce anxiety,nervousness and muscular spasms.Some use the flowers as a sleepingdraught. It was also taken to be themagic plant which made Ulysses’companions lose their memory in theOdyssey.
For the more mystical amongst us,Lotus corniculatus assistspsychic powers and helpsclairvoyance. An ancientSwedish tradition says thatyoung betrothed who putLotus flowers under theirpillows on Midsummer’sNight will have revealed tothem the face of theirfiancé.

Adipocytes are generally considered to have the capacity tostore fatty acids. However they possess all the meansnecessary to hydrolyse fats, this mechanism being calledLipolysis. Lipolysis is induced by internal factors such asadrenaline, but can also be amplified by external lipolytic suchas caffeine.

2 factors are mainly involved in lipolysis :
• cAMP (cyclic Adenosine MonoPhosphate), which issynthesised from ATP and activates lipase byphosphorylation,• lipase which once it is activated catalyses triglyceridehydrolysis into fatty acids and glycerol.
cAMP is therefore at the heart of lipolysis and is the firsttarget of agents which activate or inhibit this mechanism.Phosphodiesterase (PDE) inhibits cAMP, while adrenalinefixation on its beta receptor stimulates its activity.
One of the first targets of free radicals in the cell is the mitochondrion. When they are damaged,mitochondria produce less ATP, the cAMP concentration falls and the lipolytic activity is therefore reduced.By protecting adipocytes and their mitochondria from oxidative stress, Hydrofiltrat Marine Lotus Gmaintains an optimal level of cAMP and restores the efficacy of lipolytic agents.
Oxidative stress and ageing are accompanied by a loss of adipocyte sensitivity to lipolyticagents, and therefore a reduction in lipolysis.Protecting the adipocytes from free radicals forearms them against any celllular dysfunctioning,delays their ageing, and therefore maintains and reinforces the effectiveness of lipolytic agentslike caffeine in slimming treatment.

In vitro, the lipolytic action of adrenaline on adipocyte cultures is recreated using isoproterenol ornoradrenaline. Like adrenaline, these two lipolytic agents fix on the β adrenergic receptor and stimulatethe hydrolysis of fatty acids. Caffeine, a lipolytic agent widely used in cosmetics, inhibits PDE, removing theinhibition previously exercised by cAMP.

Protocol :Culture of mature adipocytes with or without oxidativestress in presence or absence of Hydrofiltrat MarineLotus G 2% for 1 week.
Incubation in presence of lipolytic agents ( isoproterenoland caffeine)for 5 hours
Measure of lipolytic activity (amount of glycerol liberatedby lipolysis)
Thanks to its strong antioxidant capacities, HydrofiltratMarine Lotus G neutralizes free radicals, restores thelipolytic activity of isoproterenol and boosts the efficacyof caffeine.
By protecting fibroblasts from oxidative stress, Hydrofiltrat Marine Lotus G limits thesecretion of PGE2 and therefore reduces the supplying of fatty acids to adipocytes.
Protocol :Culture of adipocytes with Hydrofiltrat Marine Lotus G for 7days, followed by UV exposure (oxidative stress).
Adiponectin is a factor released by adipocytes themselves,which limits the expansion of the adipose tissue. Under theeffect of oxidative stress, its production is significantlyreduced.
First of all, Hydrofiltrat Marine Lotus G has astimulating effect on the synthesis of adiponectin,then, thanks to its antioxidant capacities, it has aprotective effect when adipocytes are submitted tooxidative stress.These both complementary actions limit in apermanently way the expansion of the adipose tissue.

The strong antioxidant effect of Hydrofiltrat Marine Lotus G protects adipocytes from thenegative effects of free radicals. It restores the lipolytic capacity of fat cells, limits the supplyingof fats to adipocytes, controls the expansion of the adipose tissue and strengthens the slimmingproperties of caffeine. Used alone as a 2% solution or in combination with another slimming agent such as caffeine,Hydrofiltrat Marine Lotus G removes unsightly curves, sharpens the silhouette and removescellulite.
Significant decrease in the volume of thebuttocks observed by 73% of volunteers:-0.6% on average (p<0.01) and up to-2.65%
Significant decrease in the perimeter of thethighs observed by 73% of volunteers: -0.5%on average (p<0.01) and up to -1,8%.Significant decrease in the volume of thebuttocks by -4,4% (-0,9% on average).
A statistically significantcentimetre loss is measuredon the thighs, the buttocksand the hips, accompanied bya smoothing effect of theorange peel aspect.
