LewishasbeeninvolvedinhealthprojectswithNativeAmericansinnorthwestNewMexicosince1991.ShecurrentlyhasfundingfromNIEHStostudytheimpactofchronicuraniumexposureonkidneyfunction,andfromtheAgencyforToxicSubstancesandDiseaseRegistrytostudytheeffectsofuraniumexposureonpregnancies,birthoutcomes,anddevelopmentofchildren.

Inconversationswithcommunityrepresentatives,includingthevicepresidentoftheNavajoNation,BirnbaumbrieflydescribedtheNIEHSvision,mission,currentresearchactivities,andthenewfive-yearstrategicplan.SheemphasizedtheimportanceofcontinuingrelationshipswithNativeAmericancommunitiesasoneofthemostimportantelementsoftheplan.

AsanexampleofongoinginterestatNIEHSinNativeAmericanenvironmentalhealthissues,BirnbaumpointedtotheDecember2012webinaron“EnvironmentalJustice:ANativeAmericanPerspective,”whichattractedmoreparticipantsthananyotherNIEHSwebinar.“Protectingourhealthmeansprotectingourenvironment,”shesaid.“Cleanair,cleanwater,andcleanlandareessentialtopreventingdisease.”

BirnbaumandGantobservedreclamationeffortsintendedtoreduceexposuresforresidents.Theyalsoheardfromcommunitymembersabouthighconcentrationsofuraniuminsurfacewaterimmediatelydownstream.

Otherpollutantsdetectedintheareaincludearsenic,barium,chromium,cobalt,copper,lead,manganese,vanadium,selenium,andzinc.TheLagunapeoplehaveaskedtheU.S.EnvironmentalProtectionAgencytodeclaretheareaaSuperfundsite.

ScientistsandpolicymakersfromaroundtheglobemettodiscusscurrentresearchonthepotentialhealtheffectsofflameretardantsApril7-10attheSixthInternationalSymposiumonFlameRetardantsinSanFrancisco.NIEHSandNTPDirectorLindaBirnbaum,Ph.D.,iswellknownforherownresearchinthisarea.NIEHS,alongwithothersponsors,providedfundingforthemeeting.

Flameretardantsarefoundinmanyconsumerproducts,suchasbabybedding,foamcushions,carpetpadding,clothing,andelectronics.Thecompoundsareeverywhereintheenvironment,andresearchersareconcernedthatexposuretothesechemicals,atanearlyage,mayleadtoarangeofdetrimentalhealtheffectslaterinlife,eitherbyincreasingsusceptibilitytodiseaseorbyinitiatingdiseaseprocesses.

Birnbaumpresentedanoverviewoftheprevalenceandtoxicityofflameretardants,duringaplenarylecture.Shestressedtheneedtomovetoresearchthatispredictiveratherthanreactive,withknowledgeofhealtheffects,beforechemicalsareaddedtoconsumerproducts.Ifalternativechemicalsareneeded,Birnbaumsaidweneedtoavoidregrettablealternativesthatarejustasharmfulastheoriginals.Shealsoemphasizedthatmoreattentionshouldbepaidtotheneedforflameretardantsinproducts.

Birnbaumdescribedmethodstomeasurecompoundsandidentifytheirtoxicity.Shesaidthechemicallyactivatedluciferasegeneexpression(CALUX)cellbioassayisanexcellentexampleofaninnovativetooltoimproveunderstandingofenvironmentalexposures.CALUXisarapidassayusedtodetectandquantifydioxinanddioxin-likechemicalsthatwasdevelopedthroughtheUniversityofCalifornia(UC),Davis,SuperfundResearchProgram(SRP).

Superfund scientists lead the way“Theconferencebroughttogetherasmallcommunityfromaroundtheworldinterestedinthesametopic,”saidCandaceBever,Ph.D.,fromtheUCDavisSRP.“Reportsofflameretardantsinenvironmentalorhumansampleswerepresentedfrom16countriesonfivedifferentcontinents.”

NIEHSSRPgranteesHeatherStapleton,Ph.D.,fromDukeUniversity,andThomasWebster,D.Sc.,fromBostonUniversity,servedassessionchairsandmembersofthenationalorganizingcommittee.Stapletonalsogaveaplenarytalk.

“AlmosthalfoftheconferencepresentationsreferencedworkbyStapletonorWebster,forcomparisonorjustificationofresearch,”saidBever.“ThenumerouscitationsandacknowledgementsdemonstratetheircontributionstothisfieldofscienceandtherelevanceofSRP-fundedresearch.”

An emphasis on multidisciplinary research“Duringtheconference,itwasgreattohavealloftheattendeesinthefieldsofchemistry,biology,industry,andpolicyinthesameroom,”saidBever.“Chemistsneedbiologiststoexplaintheissuessurroundinghealtheffects,biologistsneedchemiststoexplainhowflameretardantstravelthroughtheenvironment,andeveryonehastoworktogethertoinformpolicy.”

Becausetherewerenoconcurrentpresentations,allresearchersfrommultipledisciplinesparticipatedinallsessions.Theprogressionoftalksbeganwithscientistsdescribingwheretheyarefindingflameretardants,movementofflameretardantsintheenvironment,howthesechemicalsaregettingintohumansandaffectingthem,andendedwithadiscussiononpublicpoliciesthataddressflameretardantsinconsumerproductsandtheenvironment.

Conferencepresentersstressedtheemergingneedtounderstandthefateandtransportofflameretardantchemicalsinthenaturalenvironmentandhumanbodies.Theyagreedthatpublicpolicyhasfocusedonflameretardantsinbuildingandconsumerproductsinthepast,butacknowledgedthatmanyoftheseproductsareendingupinlandfills,releasingflameretardantchemicalsintotheenvironment.

Training directors gather at NIEHS By Eddy Ball

Representativesofthe49NIEHS-fundedtraininggrantprogramsmetMarch27-28tolearnaboutemergingtrendsinfunding,workforcedevelopment,anddiversity.Thegrants,awardedundertheRuthL.KirschsteinNationalResearchServiceAwards(NRSA),supporttrainingprogramsforgraduatestudentsandpostdoctoraltrainees.AlsoinvitedtoattendwerethedirectorsofthethreeNIEHSshort-termsummerresearchprogramsformedicalstudents,andthefourdirectorsoftheshort-termsummerresearchprogramsforhighschoolandundergraduatestudents.

FollowingawelcomefromNIEHSProgramAdministratorCarolShreffler,Ph.D.,NIEHSandNTPDirectorLindaBirnbaum,Ph.D.,addressedthecriticalroleoftrainingintheNIEHSstrategicplan,developmentsinthebudgetprocess,andherone-NIEHSvisionforadvancingtheenvironmentalsciences.

Birnbaumwascandidabouttheeffectsofbudgetcutsongrants.“Thepaylinesaregoingtodrop,”shesaid.ButBirnbaumwasalsoeagertoassurethetrainingdirectorsthattheburdenwouldfallevenlyacrossalltheprogramsNIEHSsupports.“Intramuralprogramsarealsotakingahit,”sheadded.

RespondingtoBirnbaum’senthusiasmfortrainingsuccessesandthedeepcommitmentofNIEHStotrainingprograms,severalattendees,afterward,describedherpresentationasupbeatandreassuring.

New initiatives in training and diversityDirectorsandrepresentativesofnewtrainingprogramsreportedontheiroperationsandcurriculumdesign,aswellaseffortstoenrichtheirprogramswithexistingresourcesattheirinstitutions.Eachoftheprogramsincludesaproactiveagendaforincreasingthediversityofparticipants,andimplementinginnovativeapproachestobetterpreparetraineesforachangingjobmarket.

ThesethemesweredevelopedevenfurtherinvideocastsfeaturingrepresentativesofNIHadvisorycommitteestothedirector(ACDs)ontheissuesofbiomedicalresearchworkforcedevelopmentanddiversityinthebiomedicalworkforce(seetextbox).Theyalsoemergedindiscussionsofevaluationandfuturedirections,laterinthemeeting.

Funding decisions — a view from the other sideInherreport,NIEHSDivisionofExtramuralResearchandTraining(DERT)DirectorGwenCollman,Ph.D.,quicklyestablishedanatmosphereoftrustandsharedinterests,byhelpingattendeesunderstandhowDERTscientificadministratorsmaketheirdecisions.Shepresentedacandiddescriptionofthedifficultprocessofreviewinggrantapplicationsandsettingfundingpriorities.

“Wearesobroad,”Collmanexplained,pointingtotherangeofprogramsthatcomplicatesdecision-makingbyDERTstaff.“Wespendalotoftimearguingaboutthesechoicesin-house,”shesaid.

Insearchofafairandjustwaytorecommendawards,andmakethegreatestnumberofawardswithavailablefunds,Collmansaidherstaffmustbalancepotentiallyhigh-impactproposalswithconsiderationforongoingprogramsthataremakingsteady,ifnotheadlines-grabbing,progresstowardtheirobjectives.

Toaddressthatuncertainty,Drewoutlinedapilotsystem,establishedlastyear,todescribetraineeoutcomes,andoffertrainingprogramsameasureofaccountability,withamainfocusonthepasttenyears.Withnearly7,000NIEHStraineesloadedintothesystem,Drew’steamiscontinuingtosolicitinputforrefiningendpoints,suchasscientificandtechnicalemphasiscodes,andemploymentsectorandstatusclassifications,withthegoalofhavingdataentrycompletedbySeptember.

“Ithinkforthefirsttime,we’rereallygoingtogetsomeanswers,”Drewconcluded.

AttendeesdevotedtheremaininghalfdayoftheirmeetingtodiscussionsoftheNIEHStrainingandworkforcedevelopmentstrategicplan,inworkgroupsmoderatedbyShrefflerandNIEHSProgramAdministratorMichaelHumble,Ph.D.Programrepresentativesconsideredundergraduatecurriculum

Workforce training and supportNIHDirectorofPolicyandLiaisonActivitiesHenryKhachaturian,Ph.D.,reviewedstatisticsaboutthecurrentbiomedicalresearchworkforce,beforepresentingACDrecommendationstoadvanceNIHsupportforafuturesustainablebiomedicalinfrastructure.

KhachaturianpointedtothelargeupsurgeinbiomedicalPh.D.s,andlongtrainingtimesthatcomplicatelaunchingatraditionalresearchcareer.Comparatively,lowstartingsalaries,hesaid,contributetomakingthebiomedicalresearchcareerlessattractive,andcurrentprogramsinadequatelypreparetraineesforanythingotherthananacademicresearchcareer.

Therecommendationsaddressissueswithgraduatestudents,physicianscientists,andstaffscientists,butareespeciallypointedintermsofthepostdoctoralexperience,wheretrainingneedstobeenhancedtobettermatchavailableoff-the-benchcareers,wherenearlyone-thirdormoreoftraineesendupworking.

ImplementationofACDrecommendations,includinganincreaseintraineestipendsandbenefits,isprogressing,Khachaturianexplained,buttheexactdetailsandtimingwillstillbeundergoingreviewandclearancefollowingtheApril22deadlineforpubliccomment.

Workforce diversityNIHExtramuralProgramScientificWorkforceDiversitySpecialistLisaEvans,J.D.,openedherpresentationwithtwopiechartsthatpaintedapicturetrulyworththeproverbialthousandwords.Whilethepercentageofwhiteprincipalinvestigators(PIs)receivingNIHresearchprojectgrants(RPGs)isalmostequaltothepercentageofwhitesinthepopulation,African-Americans,with12.6percentofthepopulation,representjust1.1percentofPIswithRPGs.Latinosfarealittlebetter,16.3versus3.5percent,whileNativeAmericans,with0.9percentofthepopulation,representbutasliveroftheRPGpie.

Withrecommendationsfromitsworkinggroup,theACDforworkforcediversityhasidentifiedfourinterrelatedapproachesforaddressingtheseglaringdisparities,byincreasingsupportforstudentsthroughspecialawardstoexpandmentorship,andensuringfairnessinpeerreviewthroughimplicitbiasanddiversityawarenesstraining.Therecommendationscallforrecruitmentofapermanentchiefofficerforscientificworkforcediversity,tocoordinateNIHinitiativesandoverseerigorousprospectiveevaluationofexistingNIHprograms.

Norris outlines NIH Big Data initiative By Richard Sloane

NeweffortsareunderwaytomanagethehugeamountsofscientificdatabeingproducedbyresearchersthroughouttheNationalInstitutesofHealth,accordingtoNIHChiefInformationOfficerAndreaNorris,whogaveatalkMarch29atNIEHSabouttheNIHBigDatatoKnowledge(BD2K)initiative.

NorrisexplainedthatBD2Kaimstocreateimproveddataandsoftwaresharingpolicies,catalogsofresearchdata,andthedevelopmentofdataandmetadataqualitystandards.Shesaidsheexpectstoseeasignificantlong-terminvestmentbyNIHforacceleratedsoftwaredevelopmentandenhancedtrainingthroughnewbiomedicalbigdatacentersofexcellence.

AccordingtoNorris,theBD2KinitiativewouldcreateanadvancedcomputingenvironmentcalledInfrastructurePlus,whichwouldultimatelymodernizetheNIHnetworktomeetdatahandlingrequirementsthroughamuchmorerobustnetwork.Ideally,InfrastructurePluswouldadvancehigh-performancecomputing,andagilehostingandstorageapproachesfordifferentdatadomains.

BigDataismeasuredinterabytes,ortrillionsofbytes,andpetabytes,orquadrillionsofbytes,butaccordingtosomeexperts,withinadecade,eventhosenumbersmaybeinadequate.

AccordingtoestimatesbyEricSchmidt,Google’sformerchiefexecutiveofficer,theworldcreates5exabytes,orquintillionsofbytes,ofdataeverytwodays—roughlythesameamountofdatacreatedbetweenthedawnofcivilizationand2003.

It’sestimatedthatNIHgenerates4petabytesofdataeachday.

“Fundamentalchangeinthewaywegatherandusemassiveamountsofdataisoverdue,”saidNorris.TheagingNIHcomputernetworkcurrentlyrunsat80to90percentcapacityduringpeakutilization,farhigherthanthedesirable30to40percent.AccordingtoNorris,meetingthechallengeofmanaginggrowingamountsofdata(seetextbox)involvesmoresophisticatedtechnology,adedicatedresearchnetwork,betterharmonizationtools,andevenculturalevolution.

Issues and opportunities“Bigdataischangingdramaticallyhowwedoscience,”Norrissaid.“Accessingthesemassivepoolsofdatawillmostlikelyrequirenewskillsetsbyscientists.”

Morescientistsareincreasinglyusingpooleddata,insteadofworkingwithonlytheirown.Inmanycirclesofscience,teamsofresearchersareleveraginglarge,andevenmassive,amountsofdata(seestory).

Thisnewkindofshareddataapproachchallengesthecultureofscientificresearch,Norrissaid,becauseitwillrequirethecommunitytorecognizethevalueofgeneratinggooddata,andallowingaccesstothatdata.TheresearchcultureatNIHwillneedtochange,torespondeffectivelytonewdevelopmentsinanever-changingtechnologylandscape.

Many questions remainBioinformatics,genetics,andgenomicsstudiesproduceandconsumemassiveamountsofdata.Yetmanyquestionsarise.Howwillthisdatabestored?Howwillitbeaccessed?Whowillbeincontrol?Whatarethehardwareandsoftwarechallengestofacilitatebigdatamanagement?Howwilldatabeshared?Howcandataqualitybeassuredandadaptabletotheneedsofscience?Howlongshoulddatabestored?

“Wearelearning,”Norrisexplained.“Infiveyearswe’lllookbackandrealizehownaïvewewereonsomeoftheseapproaches.Whateverwe’redoingtoday,weexpecttoadapt,mature,andevolveovertime.”

Polyurethane: promise and perilAsubiquitousaspolyurethaneisnow,itwasinventedin1937bytheGermanchemistOttoBayer.Itsoonbecamewidelyused,becauseofitsversatilityandutility,accordingtokeynotespeakerSharonFeng,Ph.D.,oftheUniversityofChicago.Differentisocyanatecompounds,mostcommonlytoluenediisocyanate(TDI)andmethylenebisphenylisocyanate(MDI),areused,dependingonthedesiredqualitiesofthefinalproduct.

Thediversewaysthatparticipantsinteractwithisocyanatesintheirwork,bothindirectlyordirectly,enrichedthediscussionsconcerningresearchgapsandfuturepriorities.Attheendoftheconference’ssecondday,chairsofthethematicareasreportedoutdraftlistsofresearchpriorities,basedonthediscussionsandapre-conferencesurvey.ThemeetingsummaryandresearchprioritieswillbepublishedintheJournalofOccupationalandEnvironmentalMedicine.

Moving forwardAworkshopforagencyrepresentatives,chairsofthethematicareas,andtheconferenceplanningcommitteeimmediatelyfollowedthelargertwo-dayconference.AccordingtofacilitatorAndrewMaier,Ph.D.,ofToxicologyExcellenceforRiskAssessment(TERA),theworkshopwasdesignedtoshareinformationondifferentmodelsofcollaborationandleveragingofresourcestoenhancetheimpactofresearchfundingonisocyanatesandhealth.

Representativesfromseveralorganizationsinvolvedincollaborationsandpartnerships,includingNIEHS,explainedtheirmodelsforpossibleuseorexpansionrelatedtoisocyanates.Millersaid,theNIEHSmissionandpriorityareasencompassisocyanateactivityandresearch,includingtheWorkerEducationandTrainingProgram,theNationalToxicologyProgram,andfundedresearchfocusingonneurotoxicityandthedevelopmentofbiomarkersofisocyanateexposure.

Twenty years of research suggests asbestos regulations need updating By Pamela Kidron

Asbestosregulationswerelastupdatedinthemid-1980s,andmorethan20yearsofhealthresearchsuggestsit’stimetoupdatethoseregulationsagain,saidNIEHSSeniorMedicalAdvisorAubreyMiller,M.D.,duringaMarch23keynotespeechattheannualinternationalAsbestosAwarenessConferenceinWashington,D.C.Newresearchshowsthatmuchlowerexposurestoasbestoscancausedisease,Millercontinued.

Exposuretoasbestosisdangerousforminingandindustryworkers,anditisalsoariskforcommunitiesexposedtoitthroughtheenvironment.Examplesofasbestosintheenvironmentincludeasbestosdustbroughtintohouseholdsbyworkers,aswellasasbestos-contaminatedproductsandgravelsusedforroadpaving.

Thisissueattractedwideattentionin1999,whenanewspaperarticlereported192peopledeadand375sickfromasbestos-relateddiseasesinLibby,Mont.,eventhoughthevermiculiteminetherehadclosedadecadeearlier.ThevermiculitefromthemineinLibbywascontaminatedwithnaturally-occurringasbestos.

Attheconference,MillerusedLibbyasacasestudy,toshowtheassembledasbestospatients,caregivers,andmedicalexpertshowexistingasbestosregulationscouldbeimprovedtoprotectworkersandcommunities.Millersaidthatasbestoshealthstandardshavenotkeptupwithscience,andthatnewrealitiessuchastherisksfromenvironmentalexposuresnowconfrontthepublic.

Remembering the worst public health incident in EPA historyTheLibbydisasterunearthedgapsinasbestosregulations,Millerexplained.Lawswereprimarilyaimedatprotectingmineworkers,notcommunitiesthathadbeenputatriskbyhouseholdcontactwithworkersortheirclothes,fromlivingnearasitewhereasbestoswasmined,orfrombeingexposedtoasbestos-contaminatedproducts,suchasvermiculiteinsulationusedinLibbyandinmillionsofresidencesthroughouttheUnitedStates.

“Howcouldadisasterlikethis[inLibby]havehappened,whenwealreadyknewaboutthedangerofasbestosforalmostahundredyears?”askedMiller,whowaswiththeU.S.EnvironmentalProtectionAgency(EPA)atthetime,coordinatingmulti-agencyemergencyresponsestopublichealthproblems.“Theselow-doseexposures

MillerandtheEPAteamscreenedthetownforhealthproblems,andfoundthat18-20percentoftheadultpopulationhadasbestos-relateddisease,andthatmostofthesecaseswerenotrelatedtominingexposures.

Researchersalsofoundthatthecurrentmethodsbeingusedtoidentifyhazardousproducts,materials,orsoilscontaminatedwithasbestos,werenoteffectiveinprotectingworkersorthepublicfromdangerousasbestosexposures.Currentlawpermitstheuseofuptoonepercentofasbestosinmaterialsthatreadilyreleaseasbestosfiberswhendisturbed.Yetmaterialscontainingasbestos,wellbelowonepercent,canbeverydangerous.

Additionally,currentstandardsdonotregulateotherdangerous,asbestos-likemineralfibers,suchaserionite,apotentcarcinogen.“Erionite-contaminatedgravelswereusedinDunn[County],N.D.,topavemorethan300milesofroads,including32milesofschool-busroutes.EPAtestingofschoolbusesdrivingonthegravelroadsfoundelevatedairlevelsoferionite,similartotownsinTurkeywithhighratesofmesothelioma,”notedMiller.

U.S. surgeon general warns about dangers of asbestos

U.S.SurgeonGeneralReginaBenjamin,M.D.,hasurgedAmericanstolearnaboutthedangersofasbestosexposure.ThewarningcameinanApril1statementsheissuedinsupportofNationalAsbestosAwarenessWeek.

TheAsbestosDiseaseAwarenessOrganization(ADAO)applaudedthesurgeongeneral’sstatement,andtooktheopportunity,onceagain,tourgeCongresstobanasbestosandstopasbestosimports.

BenjaminstressedthatpreventingthedamagecausedbyasbestoswaskeytokeepingAmericanshealthyandsafeintheirhomes.“Together,wecanpreventthedangersassociatedwithasbestos,”shesaidinherstatement.

ADAOhasbeenworkingwithCongressandtheWhiteHousesince2004,topreventasbestosexposure.Respondingtothesurgeongeneral’sstatement,ADAOco-founderandpresidentLindaReinsteinnotedthatthereisconsensusamongtheEPA,NationalInstituteforOccupationalSafetyandHealth,NationalToxicologyProgram,WorldHealthOrganization,InternationalLabourOrganization,andInternationalAgencyforResearchonCancerthatasbestosisacarcinogenandthatthereisnosafelevelofexposure.

Hennig receives prestigious professorship at the University of Kentucky By Carol Kelly

BernhardHennig,Ph.D.,directoroftheUniversityofKentucky(UK)SuperfundResearchCenter(SRC)andprofessorofnutritionandtoxicologyintheCollegeofAgriculture,istherecipientofaUniversityResearchProfessorship.TheseprofessorshipsrecognizeoutstandingUKresearcherswhohaveestablishedanationalreputationforsustainedcontributionstotheirfield,andcarryanawardof$40,000.

Hennig’sdistinguishedresearchprogramisbroadlyfocusedonunderstandingmechanismsunderlyingthepositiveeffectsofselectednutrientsonhumanhealth.AlthoughHennighasalwaysbeeninterestedinhownutritioncanaffecthealthanddisease,hisworkwiththeSRChasdrivendiscoveriesabouthownutritionalsciencesandtoxicologycanoverlaporbeinterrelatedscientificdisciplines.

“TheconceptthatnutritioncanmodulatehowpeoplerespondtodiseaserisksassociatedwithexposurestoSuperfundpollutantsisfascinating,”saidHennig.

Blending nutritional and toxicological environmental researchHennig’sresearchprovidesevidencethatpeoplemayreducetheirrisktoenvironmentalpollutantexposuresthroughhealthynutrition.Hecontendstheresearchsupportsaparadigmshiftwherenutritionordietaryhealthisacriticalplayerinoverallvulnerabilitytoenvironmentalstressors.

Hennig’sresearchgroupiswellknownforafindingthatsuggeststhetypeoffatweeatcanalterhowourbodiesrespondtoenvironmentalstressors.Theyfoundthatdietsrichinomega-6fattyacids,foundinplant-derivedoils,suchascornandsafflower,canfurtherpromotethepro-oxidativeandproinflammatoryeffectsofcertainpolychlorinatedbiphenylcompounds(PCBs),prevalentenvironmentalpollutants.Incontrast,Hennig’sgroupalsofoundthatdietsrichinomega-3fattyacidsandantioxidantpolyphenols,compoundsfoundinmanyfruits,vegetables,andwholegrains,canlessentoxicityofPCBstothecelllayerthatlinebloodvessels.

“Thesebeneficialcompoundsworkinthebodytohelprepairdamagedcellsandpreventcertaindiseasemechanismsfromoccurring.Nutritionshouldbeconsideredinoverallriskassessment,”saidHennig.

Hennigbelievesthatnutritionalinterventionsmayprovidethemostsensiblemeanstodevelopprimarypreventionstrategiesofdiseasesassociatedwithmanyenvironmentaltoxicinsults.

Envisioning future research“Thisprofessorshipwillallowmetoaddressfutureissuesrelatedtonutritionalmodulationoftoxicologicalinsults,”saidHennig.

HennighasbeenamemberoftheUniversityofKentuckyfacultysince1984.Since2001,hehasservedaseditorinchiefoftheJournalofNutritionalBiochemistry,ahighlyrespectedjournalforresearchintothebiochemicalbasisofnutrition.Since2003,hehasdirectedtheSRC,fundedbytheNationalInstituteofEnvironmentalHealthSciences.Hisresearchhasproducedasteadyseriesofpublicationsinprominentjournalsandhasearnedhimothernationalawards.Hennigalsoregularlyparticipatesinnationalandinternationalconferencesasaninvitedspeaker.

TheUniversityofWashington(UW)CenterforEcogeneticsandEnvironmentalHealth(CEEH)hostedthe2013NIEHSEnvironmentalHealthSciences(EHS)CoreCentersmeetingApril17-19inSeattle.ThemeetingbroughttogetherNIEHSstaffandmembersfrom20corecenterstosharetheirlatestworkinenvironmentalhealth,fostercollaborations,andhighlightnovelresearchandcommunityengagementactivities.

Themeetingfeaturedscientificsessionsongene-environmentinteractionsandglobalenvironmentalhealth,breakoutsessionswithtopicsrangingfromhydraulicfracturingtobidirectionalcommunication,andasessionhighlightingresearchofyounginvestigators.ThemeetingalsoincludedacommunityforumtolearnabouttheDuwamishRivercleanupproject,whichendedwithaneducationaltourofPugetSound,toinformparticipantsaboutlocalSuperfundsitesandremediationefforts.

Duringanothersession,aninter-corecenterworkinggrouponhydraulicfracturingconvenedtodiscussitsresearchandcommunityengagementrecommendations.TrevorPenning,Ph.D.,fromtheUniversityofPennsylvania(UP),facilitatedthediscussiononthecurrentresearchgaps.Participantsexplainedtheneedforbaselineairandwaterqualitydata,andhighlightedacollaborativestudy,betweenUPandColumbiaUniversity,tocollectenvironmentaldatainPennsylvania,wherehydraulicfracturingisprevalent,andinNewYork,wherehydraulicfracturinghasnotyetbegun.

Thegroupdiscussedtheimportanceofcompilingdatafromrelatedstudiesandpreservingitsquality.HarvardUniversity’sEHScorecenterhasinitiatedthisprocesswithFrackMap,acompilationofdatarelatedtohydraulicfracturing,throughtheHarvardUniversityWorldMapproject,anopen-sourcewebmappingsystem.

COEC effortsCommunityoutreachandengagementcore(COEC)leaderssharedpastaccomplishmentsandconsideredfutureeffortstosustainandstrengthentheirwork.TheystartedthemeetingbyconveningahistorywalltocapturethemanyCOECmilestoneeventsandaccomplishmentssincethebeginningoftheEHSCoreCenterprogram.ThehistorywallservedasawaytointroducemanyofthenewCOECmemberstotheprogram.Throughoutthemeeting,COECleadersfocusedonevaluation;emergingtopics,suchasenvironmentalhealthliteracy,socialmedia,andhealthimpactassessments;andpeermentoring.

Oneofthesessionsfocusedonpreparingthenextgenerationofenvironmentalhealthscientistsandbuildingadiverseworkforce.Theyallagreedthatmakingsciencerealcommunityresidentsandyouth,startingasyoungaselementarystudents,wascentraltothiseffort.

ThePortofSeattle,Boeing,andotherindustriesarelocatedontheriver.TheDuwamishSuperfundsite,a5.5milestretchoftheriverthatflowsintoElliottBay,isoneofthemostpollutedplacesintheUnitedStates.TheU.S.EnvironmentalProtectionAgency(EPA)releaseditsproposedcleanupplanforthesiteonFebruary28,andthistimelyforumoccurredduringthe90-daypubliccommentperiodonEPA’sproposedplan.

Thediverse,historicGeorgetownandSouthParkneighborhoodsalongtheDuwamisharehometoadisproportionatenumberoflow-income,Hispanic,andrecentimmigrantresidents.TheriverisalsopartofthetraditionalfishinggroundsofthreeNorthwesttribes.

AlthoughaWashingtonState,DepartmentofHealthadvisorywarnedresidentsnottoeatfishorshellfishfromtheDuwamishRiver,healthofficialsknowthatmanypeoplestillsubsidizetheirdietswiththecontaminatedfish.Theydosobecauseofeconomicnecessity,amisunderstandingofthedanger,oranacceptanceofthehealthrisks.

Yakel inspires future scientists By Robin Mackar

Itwasn’tjustatalkaboutcholinergicreceptorsinthebrainthatgotthestudentsatSaintAugustine’sUniversity(SAU)excitedaboutcareersinscience.Itwasmoreamessageofhopeandpursuingdreamsthatcaughttheirattention.

“Iwaswhereyouarenow,studyingformybachelor’sdegreeinscience,andfeelinglikeIwasn’tsmartenoughorgoodenoughtobeascientist,”saidJerrelYakel,Ph.D.,headoftheIonChannelPhysiologyGroupandActingChiefoftheLaboratoryofNeurobiologyatNIEHS.“Fortunately,Ihadfabulousmentorsthatkeptencouragingmetopursuemyloveofscience.AndI’msogladIlistenedtothem.”

YakelwasinvitedtogivethekeynoteaddressApril18atSAU’s10thannualUndergraduateResearchDayfortheSchoolofSciences,Mathematics,andEngineering.Yakelspokefromexperience,whenhesaidstudentsfromunderrepresentedgroupssometimesthinkgraduateschoolsaretooexpensiveandoutofreach.

Exciting time for brain researchDuringhistalk,Yakelalsomentionedwhyitwasanexcitingtimetobeascientist,especiallystudyingthebrain.

“Justafewweeksago,thePresidentandthedirectoroftheNationalInstitutesofHealthstoodtogetherandannounceda$100millioninitiativethatisgoingtorevolutionizeourunderstandingofthebrain,”saidYakel.ThenewBrainResearchThroughAdvancingInnovativeNeurotechnologies(BRAIN)Initiativeaimstohelpresearchersfindnewwaystotreat,cure,andevenpreventbraindisorders,suchasAlzheimer’sdisease,epilepsy,andtraumaticbraininjury.

YakelreceivedhisB.S.fromOregonStateUniversityandhisPh.D.fromtheUniversityofCalifornia,LosAngeles,wherehestudiedligand-gatedionchannelsandserotoninreceptors.Heiswellknownforhisworkonnicotinereceptorsinbrainfunction,andhisrecentworkinmicemayhaveimplicationsforlearningandmemory.Hisgroupdemonstratedthat,inthebrain’shippocampus,thetimingofthereleaseoftheneurotransmitteracetylcholinemayplayaroleinregulatingnervecellconnectionsorsynapses(seestory).

Yakel,aswellasotherscientistsandpostdoctoralstudents,workcloselywiththeNIEHSOfficeofScienceEducationandDiversity,toreachouttotalentedstudentsfromunderrepresentedgroupstogettheminterestedinenvironmentalhealthsciences.TheirworkhaspaidoffasthreeSAUstudents—KaceyMcHoney,JenniferPlair,andMalcolmRichbourg—arecurrentlydoingresearchthroughtheNIEHSScholarsConnectProgram(seestory).

DuringthefinalweekofMarch,NIEHSmarkedasocialmediamilestone,asthenumberofitsFacebooklikesexceeded1,000forthefirsttime.

“LikingaFacebookpagesignifiesamuchgreaterlevelofapprovalandtrustincontentreliabilitythanmerelyvisitingapage,”saidJosephPoccia,whojoinedtheNIEHSOfficeofCommunicationsandPublicLiaison(OCPL)in2012asaWebinformationdevelopmentspecialist.

AccordingtoOCPLDirectorChristineFlowers,formanypeople,especiallythoseinthe18-34agerange,oftenthefirstthingstheycheckfornewsinthemorningareFacebookandTwitterposts,whichcanappealtopeoplewithspecialinterests,suchasenvironmentalhealth.

“PeopleareincreasinglygettingtheirnewsandinformationfromtheInternetontheircomputers,tablets,andcellphones,”Flowerssaid.“Socialmediagivesusagreatwaytocommunicatehealthandscienceinformationquicklyandaccurately.We’reexcitedaboutreachingyoungpeopleandsparkingtheirinterestinresearchandsciencecareers.”

Anotherupsidetosocialmediaistheabilityofitscontenttogoviralthroughpostingandrepostingbydirectandsecondaryfollowers.

OCPLlauncheditsTwitterpageinDecember2010andhasaccumulatedmorethan3,100followers.TheInstitute’sFacebookpage,launchedinDecember2011,currentlydrawsmorethan4,000visitsperweek.

AccordingtoPoccia,asofApril2,NIEHSFacebookhadatotalof1,028likes.“AftertheU.S.with553,wehavethemostlikesinEuropewith55,CentralandSouthAmericawith35,Indiawith31,Pakistanwith28,andEgyptwith23,”Pocciasaid.“Intermsofdemographics,38.7percentofouraudienceonFacebookisbetweentheagesof18-34,and57.1percentisfemale.OurTwitterfeedhasmorethan5,800followers.”

Oneofthethingsthatmakessocialmediasoappealingisitsabbreviatedformat.InthecaseofTwitter,userposts,referredtoastweets,maynotexceed140characters.Thatputspressureoncommunicatorstowritecontentthatisbothenticingtothereaderandconcise.ThetweetconventionhaseveninfluencedthewayOCPLputstogetherthelandingpageofitsmonthlynewsletter,withstoryleadsgenerallyfallingwithinthe140-characterrange.

Asaresearchinstitute,NIEHSfocusesonunderstandinghowtheenvironmentaffectspeople’shealth,butitaddstothecausebyparticipatinginprogramsthatencourageyoungpeopletoentercareersinenvironmentalhealthsciences.

Inoneofthoseoutreachevents,NatashaClayton,abiologistintheNIEHSCellularandMolecularPathologyBranch,joined11otherprofessionalsattheClarenceE.LightnerYMCAAchieversCareerFairApril20inRaleigh,N.C.SponsoredbytheYMCAoftheTriangle,theeventexposedteenagers,fromunderrepresentedgroups,toanumberofprofessionsavailabletothemoncetheyfinishhighschool.

InadditiontohearingfromClayton,approximately70middleschoolandhighschoolstudentsweretreatedtotalksfrompeopleinthefieldsofcosmetology,law,medicine,militaryservice,music,psychology,andeducation.

Being an exampleAsarepresentativeofoneofthemyriadofcareersinscience,technology,engineering,andmathematics(STEM),Claytondiscussedhowtobecomeabiologistandhowtostayontoponcethere.ShepresentedanoverviewofNIEHS,herday-to-dayworkinsupportofthemission,andinformationaboutseveraltrainingopportunitiesavailableattheInstitute.Claytonexplainedwhyshewascompelledtoparticipateinthecareerfair.

“Ifeelitismyresponsibilitytogiveback,bysharingmyexperienceswiththeyouthinourcommunities,”Claytonsaid.“IcouldseemuchpromiseintheteenswithwhomIinteracted,andseemanyofthemasfutureleaders.”

KendallHarris,YMCAoftheTrianglecommunityoutreachdirector,thankedalloftheparticipantsformakingthecareerfairasuccess,andconveyedsinceregratitudetothespeakerswhotooktimeoutoftheirbusyschedulestoattend.

“Itisgoodtohavepeoplelikeyouthatcareaboutourteensandtheirfutures,”Harrissaid.

Earlyinhiscareer,MichaelCahalan,Ph.D.,wantedtocombineimmunologyandbiophysics,twoseeminglyunrelatedfieldsofscience,tocreateauniqueresearchnichethatwashisown.Sincethen,hisvisionhasledtotheabilitytoobservelivingimmunecellsintheirhostenvironment.

CahalandiscussedhisworkApril9aspartofthe2013NIEHSDistinguishedLectureSeries.“CrackingCRAC—thecalciumrelease-activatedcalciumchanneloftheimmunesystem,”washostedbyJamesPutney,Ph.D.,headoftheCalciumRegulationGroupintheNIEHSLaboratoryofSignalTransduction.

CahalanisadistinguishedprofessorandchairoftheDepartmentofPhysiologyandBiophysicsattheUniversityofCalifornia,Irvine.Inhisintroduction,PutneydescribedCahalanasaninnovativeresearcher,whosequesthasnowturnedintoanenterprisingventurethatdemonstratestheimportanceofthecalciumsignalingpathwayinTcellfunction,anditspotentialasatherapeutictargetforimmunedeficiencyinhumans.

Observing immune cells in real time“Iwouldliketotellastorythroughsomemovies,”Cahalansaidashebeganhistalk.Heintroducedtheaudiencetotherevolutionarytwo-photonmicroscopytechnique,whichprovidesamuchhigherphotondensitycomparedtoconventionalmicroscopymethods,andsubsequentlyenablesimagingwithgreatersensitivityanddepthpenetration.Cahalanandcolleaguestookadvantageofthistechnologytoproducethree-dimensionalmoviesofliveimmunecellsinvarioushostenvironments,fromisolatedmouselymphnodestomice.

Cahalan’simagingexperimentsrevealedhowdifferenttypesofimmunecellsmoveinthelymphnodes,theirhomingbehavior,andtheirmodesofactiontoeliminateforeignantigens.

“TheTcellsmoveincrediblyfast,sothattheycanscantheenvironmentforantigensthatarecapturedandbroughttothelymphnodesbydendriticcells,”Cahalanexplained.“Oncethetwocelltypesmeet,thedendriticcellsinteractwiththeTcellsthroughlongextensions,formingtheimmunologicalsynapses.”

Hesaidtheseinteractions,inturn,activatetheTcells,causingthemtodividerapidlyandexecutetheireffectorfunctions.

Calcium signaling in T cell functionThediscoveryofvariousionchannelsinTcells,manyofwhicharecalcium-regulated,sparkedCahalan’sinterestinthesignificanceofcalciumsignalinginTcellfunction.HisgroupfoundthatcalciumsignalingisrequiredtoenhanceTcellproliferationandthestabilityoftheimmunologicalsynapses.Thegroupalsomeasuredasmall,butsignificant,inwardcalciumcurrentwhichrestoresthecalciumsourceinsidethecells.

Throughsystematicandgenome-wideRNAiscreening,usingculturedinsectcells,Cahalanandcolleaguesdiscoveredtwoproteins—thestromalinteractingmolecule(STIM)andOrai—whichconstitutewhattheytermedtheCa2+release-activatedCa2+(CRAC)channel.Homologuesoftheseproteinsarepresentinhumans,andthechannelishighlyselectiveforcalciumions.Moreimportantly,theCRACchannelsareresponsibleforengagingTcellsanddendriticcellsintheimmunologicalsynapsesuponantigenrecognition.

CahalanemphasizedthattheCRACchannelsaretightlyregulated.Singleaminoacidmutationsintheirproteinsequencesmayaltertheirflexibilityandionselectivity,leadingtodireconsequences.

“Manyofthesemutationshavebeenimplicatedinpatientswithsevere,combinedimmunedeficiency,”headded.

Healsostressedthatthestoichiometry,aswellasthelocalizationofSTIMandOraiinthecells,iscriticalfortheproperfunctioningofthechannels,andhasdevotedsubstantialeffortintodissectinghowtheseaspectsareregulated.Ultimately,CahalanhopestocapitalizeonhisfindingsonCRACchannels,bydevelopingtherapeuticagents.

“Thisisalongandarduousprocess,butweareoptimisticthattheseagentswillmaketheirwayintoclinicaltrials,andeventuallybenefitpatientswithimmunedeficiencydisorders.”

NTP webinar informs Report on Carcinogens By Robin Mackar

TheNationalToxicologyProgram(NTP)successfullyusedaweb-basedmeetingformatApril11tobringtogetherscientificexpertsandmembersofthepublicforadiscussiononpentachlorophenol(PCP)exposureandcancer.

PCP,whichisprimarilyusedasawoodpreservativeintheUnitedStatesforitemssuchasutilitypoles,crossarms,fenceposts,andrailroadties,isacandidatesubstancebeingformallyreviewedforpossiblelistingintheReportonCarcinogens(RoC).Thewebinarcreatedavirtualmeeting,allowingmorethan70registeredparticipantstowatch,listen,andparticipateindiscussions,byphoneandcomputer,fromacrosstheglobe.

Thehalf-daysessionwasmoderatedbyepidemiologistGlindaCooper,Ph.D.,fromtheU.S.EnvironmentalProtectionAgency.ThemeetingincludedpresentationsfromfouroutsidescientistswhohavebeeninvolvedinPCPhumanstudies.Theobjectiveofthemeetingwastoreceivescientificinputonhowbesttoevaluatetheepidemiologic,orhuman,studiesrelatedtoPCPexposure.NTPwasspecificallyinterestedinhearingaboutPCPcomponentsandcontaminants,anddistinguishingbetweencancereffectscausedbyPCPandthosethatmightbecausedbyotherchemicalsorcontaminantstowhichworkersmayhavebeenexposed.

Purpose of webinar“ThiswebinarisintendedtohelpinformourcancerevaluationofPCP,”saidRuthLunn,Dr.P.H.,directoroftheNTPOfficeoftheReportonCarcinogens,whileprovidinganupdateontheRoCprocess.“ItisnottoreceiverecommendationsfrominvitedspeakersorthepubliconwhetherornotPCPshouldbelistedintheRoC.”Thecancerevaluationcomponent,developedbyLunnandherstaff,laysoutalltheinformationusedtomakeapreliminarylistingdecision,andisanintegralpieceofthemonographthatNTPpreparesaspartofitsevaluationofasubstance.

Aftersomebriefintroductorycomments,Cooperintroducedthefirstpanelist,andfacilitatedaquestionandanswersessionamongparticipantsaftereachpresentation(seetextbox).

Presentation HighlightsKevin Dunn, from NIOSH,presentedanoverviewofoccupationalexposurestoPCP.HesaidPCPwasproducedintheUnitedStatesfrom1936to2006,butsince1984,itsuseintheU.S.hasbeenrestrictedtowoodpreservationandcannolongerbeusedonwoodinresidentialoragriculturalbuildings.DunndiscussedhowexposureforPCPmanufacturingworkersmightoccur,includingthroughdustorvapors,dependingonthefinishingprocessusedtopreparethePCPandexposureinthewoodpreservativeindustry.HealsoidentifiedcommonPCPcontaminants.

Avima Ruder, Ph.D., also from NIOSH,focusedonoccupationalexposuretoPCPandotherchemicals,whilefurtheraddressingsomeoftheissuesthatneedtobecarefullyconsideredwhenevaluatinghumanepidemiologystudies.ShediscussedtheseissuesastheyrelatetowhatNIOSHrecentlyfoundinamortalitystudythatincludes2,122U.S.PCPproductionworkers,fromfourlargechemicalplantsoperatingfrom1936-2006.Theworkerswereexposedtootherchemicals,aswell,whileworkingattheplants.

James Collins, Ph.D., from the Dow Chemical Company,focusedhistalkonbiomonitoringandepidemiologicstudiesofPCPproducers.HetalkedabouthowdifficultitistodetermineexposureassessmentforPCP.HesharedbiomonitoringdataonserumdioxinsfromdifferenttypesofPCPworkers,andsuggestedthatthedioxins,whicharelong-livedinthebody,mayserveasanindicatorforpastexposuretoPCP.HediscussedfindingsfromanepidemiologystudyofPCPmanufacturingworkersattheDowchemicalplant.

Thefinalpresentationcamefrom Paul Demers, Ph.D., a professor at the University of Toronto Dalla Lana School of Public Health and the Cancer Care Ontario Occupational Cancer Research Centre.DemersfocusedmuchofhisremarksonalargestudyhehasbeeninvolvedwithinBritishColumbiathatincludes27,464workers,employedby14sawmillsfor1yearormore,between1950and1995.HealsoreviewedfindingsofotherepidemiologicstudiesofPCP.

Symposium features cutting-edge stem cell research By Ernie Hood

NIEHSanditsneighboringresearchuniversitiesarehotbedsofscientificactivitywhenitcomestothevastpotentialofstemcells,soitwasonlynaturaltheInstitutewouldholdasymposiumonthetopic.OnApril11-12,NIEHShostedUnlockingthePromiseofStemCells,ameetingthatbroughttogethersomeofthenation’sleadingresearchersinthefield.

RajaJothi,Ph.D.,andGuangHu,Ph.D.,tenure-trackinvestigatorsintheNIEHSLaboratoryofMolecularCarcinogenesis(LMC),explorethebiologicalmechanismsofstemcellsandco-organizedthegathering.Theybothbelievedhavingadiscussiononhowstemcellscouldbeusedtostudyenvironmentalhealthsciencewasneeded,giventherecentstemcellbreakthroughsinregenerativemedicine,drugdiscovery,andtoxicitytesting.

“Wethoughtthiswouldbeagoodopportunitytoinviteresearchers,whoareleadersinthefield,toexaminethecurrentstateofstemcellresearchandtothinkabouthowtomovethedisciplineforward,”Husaid.“Thepossibilitiesareendless,fromscientificresearchtoolstotherapeuticapplications.”

Thetwodaysofpresentationsdisplayedawiderangeofareasinwhichstemcellsarepoisedtomakemajorcontributions(seetextbox).AsNIEHSScientificDirectorDarrylZeldin,M.D.,notedinhiswelcomingremarks,“HereatNIEHS,we’rejustbeginningtoappreciatehowstemcellsandtheirdifferentiatedprogeniescanbeusedasnewtoolsandasmodelsfortoxicologicstudies,withtheprimarygoalofunderstandinghowenvironmentalfactorsinfluencehumanhealthanddisease.”

Gathering of like mindsThesymposiumattractedmorethan250attendeesonbothdays.Participantscamefromlocalgovernmentagencies,universities,andindustries,aswellasfromuniversitiesinVirginiaandtheNationalInstitutesofHealth(NIH)inBethesda,Md.

“Themeetingprovidedagreatopportunityforlocalstemcellresearcherstointeractandnetwork,whichwehopewillleadtomanycollaborativeeffortsinthefuture,”Jothisaid.

DNA methylation marker shows promise for detecting breast cancer risk By Pamela Kidron

DNAmethylationcouldprovetobeaneffectiveindicatorofbreastcancerrisk,accordingtoanewstudypublishedonlineApril11intheJournaloftheNationalCancerInstitute.

DNAmethylationisaformofepigeneticmodificationthatmaycontrolgeneregulation,butisnotdirectlypartoftheDNAsequence.Inthisstudy,NIEHSepidemiologistsJackTaylor,M.D.,Ph.D.,andZongliXu,Ph.D.,discoveredthat,bymeasuringsuchepigeneticmodifications,it’spossibletocapturechangesinsusceptibilitytobreastcancerthatresultfromfactorssuchasagingortheenvironment—changesthatgeneticstudiesofDNAsequencedonotcapture.

Taylor’sgroupfoundthatevidenceofthismodificationwasabetterpredictorofbreastcancerriskthanotherapproaches,suchastheGailmodel,whichreliesonknownriskfactorsincludingage;reproductive,medical,andfamilyhistory;andcommonpolymorphisms.However,Taylorcautionsthattheworkisthefirststudyofitskindandthatthesemeasurementsarenotyetaccurateenoughforclinicaluse.

NIEHS Sister Study yields valuable dataWorkingwithDNAextractedfromwhitebloodcellsinbloodsamples,Taylorandhiscolleaguesassessedthemethylationprofileof27,000sitesacrossthegenome.TheyusedsamplesfromacohortofwomenintheNIEHSSisterStudy,anationwidestudyofwomenwhohavehadabiologicalsisterwithbreastcancer.TheSisterStudyisdesignedtoexploregeneticandenvironmentaldeterminantsofbreastcancer.

TheNIEHSteamusedthedatatoexaminewhetherthesesitesweredifferentinthosewholaterdevelopedbreastcancerthanthosewhodidnot.Theyalsoexaminedthepredictivepowerofbloodmethylation,comparedwiththeGailmodel,and10commonpolymorphismsthatareassociatedwithbreastcancerrisk.

ThedataprovidedevidencethattherearedifferencesinbloodDNAmethylationinthosewhoultimatelydevelopbreastcancer.Thesedifferencesaredetectablemonthstoyearsbeforetheclinicaldiagnosisofbreastcancer.

Ofthe250differentiallymethylatedsitestheteamidentified,188,or75.2percent,wereundermethylated.Thepredictionaccuracyviamethylationwas66percent,comparedto56percentfortheGailmodeland59percentforgenome-wideassociationstudypolymorphisms.

Producing a diagnostic tool Thestudymayhelpresearchersmoveclosertodevelopingabloodtestthatcanpredictawoman’schancesofdevelopingbreastcancer.Womenwithelevatedriskcouldthenbemonitoredclosely.Earlydiagnosisofbreastcancer,beforeitspreads,isessentialforsuccessfultreatment.

Breastcanceristhemostcommoncancerinwomenandtheleadingcauseofcancermortalityworldwide.Some40,000peopledieannuallyfrombreastcancerintheUnitedStates.

DarshiniTrivedi,Ph.D.,lefttheNIEHSLaboratoryofToxicologyandPharmacologyinApriltostarthernewcareerasaclinicalresearchscientistatImpactPharmaceuticalServicesInc.(IMPACT).Thecompanyisalocalcontractresearchorganizationthatspecializesinprovidingregulatoryanddrugdevelopmentconsulting,medicalwriting,andprojectandprogrammanagementtothepharmaceuticalandbiotechindustries.

Perfect timingTrivedi’sjobsearchisastoryofnetworking,hardwork,andperfecttiming.ShewasoneofthefirstmembersoftheRhoTauChapterofGraduateWomeninScienceinResearchTrianglePark,N.C.,andwhenoneofthechapterofficerstransitionedtoIMPACT,shenotifiedTrivediaboutajobopening.ThatconnectionledtoaninterviewinFebruary2012,butTrivediwasnotreadytostartanewjobatthattime.Shewasworkingonaprojectthatneededafewmoremonthsofadditionalexperiments,andshesuggestedlatesummerasapotentialstartdate.

ManyoftheworkshopsthatTammyCollins,Ph.D.,directorofNIEHSOfficeofFellows’CareerDevelopment(OFCD)andLoriConlan,Ph.D.,directoroftheNIHOfficeofPostdoctoralServices,broughttoNIEHShelpedTrivedianswerbehavioralquestionsshewasaskedduringthejobinterview.

TrivedialsorecognizedthequalityofthementorshipshereceivedwhileatNIEHS.Duringthosefouryears,TrivediandLangenbachidentifiedthesignalingproteinbeta-arrestin-2asamajorplayerthatcouldbeusedasadrugtargetfornewtreatmentsinaneurysmdevelopment(seetextbox).

“I’vehadexcellentmentorswhoalwaysofferedmegreatsupportandadvice,”Trivedisaid.“IwasfortunatetoworkwithBob[Langenbach],whogavemeindependenceinthelab,andbasedonotherongoingbeta-arrestinstudies,thefreedomtodevelopanewaneurysmdiseasemodel.”

Casey presents at workshop on stems cells in cardiotoxicity studies By Cathy Sprankle

Pollutantsandtoxicantsintheenvironmenthavethepotentialtodamagecellsintheheart.Knownascardiotoxicity,itisalsoamajorreasonfordrugdevelopmentfailure.Becauseofitsimportance,scientistsfromallovertheworldgatheredinBostonMarch18-19tosharethebestandlatestresearchmethodsinthefield.

OneofthosescientistswasWarrenCasey,Ph.D.,actingdirectoroftheNationalToxicologyProgram(NTP)InteragencyCenterfortheEvaluationofAlternativeToxicologicalMethods(NICEATM).Caseyjoinedaninternationalgroupofpresenters,representingresearchinstitutions,pharmaceuticalcompanies,andgovernmentagencies,attheStemCell-DerivedCardiomyocytesasModelsofCardiacPathobiologyandToxicologyWorkshop.

TheHealthandEnvironmentalSciencesInstitute(HESI),anonprofitinstitutionthatbringsscientiststogethertoaddressglobalhealthandenvironmentalissues,sponsoredtheworkshopasawaytoassessthecurrentstatusandpotentialapplicationsoftheuseofstemcell-derivedcardiomyocytes,orculturedheartcells,forstudyingcardiotoxicity.

Casey’spresentationdescribedhowtestingapproaches,thatuseculturedcells,mightbeusedinregulatoryorsafetydecision-makingcontexts.

Understanding how aneurysms formTrivediandLangenbachstudiedabdominalaorticaneurysm(AAA),apotentiallylife-threateningdiseasethataffectsbloodvessels.Sincenocurrentpharmacologicaltreatmentexistsforthecondition,TrivediandLangenbachsetouttounderstandAAAformation.

OtherresearchersutilizedamousemodeltostudyAAAformationandestablishedthatangiotensinII(AngII),adrugthatraisesbloodpressurebyconstrictingbloodvessels,bindstoaG-proteincoupledreceptorcalledAT1a.Additionalstudiesfoundthatthemultifunctionalscaffoldingprotein,beta-arrestin-2,alsobindstoAT1a,soTrivediandLangenbachwantedtoknowifbeta-arrestin-2wasinvolvedinAAAformation.

Theydeterminedthatbeta-arrestin-2contributestoAngII-inducedAAAformation,byactivatingextracellular-signal-regulatedkinases(ERKs),cyclooxygenase-2(COX-2),andotherfactorsinvolvedintheinflammatoryresponse.Theirresultssuggestthatbeta-arrestin-2maybeanoveltargetforpharmacotherapeuticdesigninAAAtreatment.

“Stemcellsandothertechnologiescanhelpusbetterunderstandthemechanismsunderlyingtoxicityanddisease,”hesaid.“Forexample,theycanhelpusunderstandhowthegeneticdiversityoftargetpopulationsaffectstoxicity.But,aswemovetowardusingemergingtechnologies,suchasstemcell-derivedcardiomyocytesforsafetytesting,it’simportantthatwedevelopappropriatevalidationcriteria,sothatthedatathesetestmethodsprovideareusefultoregulators.”

Theworkshop’sgoalwastoevaluatehowsuchtechnologiesmaybeusedtoevaluateriskstohumancardiachealthfrompharmaceuticalsandenvironmentalchemicals.Topicsdiscussedincludedthebiologyofculturedcardiomyocytes,specificapproachestousingthemtoassesstoxicity,howthoseapproachesmightbeusedtobenefitpublichealth,andfutureresearchanddevelopmentneededtoachievethosepublichealthbenefits.

Farmland application of sewage sludge raises environmental justice concerns By Nancy Lamontagne

Peoplewholiveclosetofarmlandwheretreatedsewagesludgeisappliedtothesoilsaidtheyexperiencephysicalsymptoms,smelloffensiveodors,andareangryaboutthepractice,accordingtoanewstudyfromNIEHSgranteesattheUniversityofNorthCarolinaatChapelHill(UNC)GillingsSchoolofGlobalPublicHealth.Thestudy,whichwassupportedbytheNIEHSPartnershipsforEnvironmentalPublicHealth,indicatesaneedforinvolvingcommunitymembersinthedecision-makingprocessaboutlandapplicationofsewagesludge.

Researchersfoundthathealthconcernsrecordedinthisstudywereconsistentwithotherreports,buttheydidnotexpectsomanyrespondentstoexpressfeelingsofinjusticeoverthespreadingofurbanwastescalled“sludge”inruralareas.

Responding to a community need“Ruralresidentswhoknewaboutourpreviousstudiesofhealthimpactsofhogwastecontactedusaboutillnessandnoxiousodorsthattheyassociatedwiththeuseoftreatedsewagesludgeonfarmlandneartheirhomes,”saidSteveWing,Ph.D.,associateprofessoratUNCandaco-authorofthestudy.Inresponse,theresearchersinterviewed34peoplefromNorthCarolina,SouthCarolina,andVirginiawholivednearfieldswheresludgeisappliedasasoilamendment.

Morethanhalfofthestudyparticipantsreportedphysicalsymptomssuchaseye,nose,andthroatirritationsandgastrointestinalsymptoms.Mostofthemsaidthattheunpleasantodorsfromthesludgedisruptedtheirdailyactivitiesandsocializingwithfamilyandfriends.

“Researchersandpublicofficialsresponsibleforpermittinglandapplicationofsludgefrommunicipalwastewatershouldnotbesodismissiveofneighbors’reportsofillness,”Wingsaid.

Inadditiontobeingangryovernotbeinginformedthatsludgewasbeingappliedneartheirhomes,thepeopleinterviewedalsosaidthatregulatorsseemedunconcernedwithviolationsoflandapplicationrules,andthatpublicofficialsdonotrespondtoreportedconcerns.Studyparticipantswantedimprovedmonitoringandbetterenforcementofregulations,andseveralsaidtheywouldlikepublicofficialstodirectlynotifyresidentsnearapplicationsitespriortoeveryapplication,sotheycanprepareandtakeprecautions.

“Itmayalsobehelpfultoprovideawayforneighborsoftreatedsludgefieldstoreportpossibleviolations,suchasapplicationduringraineventsorrunoffofsludgeintowaterways,sopotentialproblemscanbepromptlyaddressed,”Lowmansaid.

Wingoutlinedseveralchangesthatcouldlessentheimpactoflandapplicationofsludge.Intheshortterm,thesemodificationsincludedecreasingthefrequencyandamountsofsludgeapplied,increasingbuffersbetweenapplicationsitesandneighbors,andimprovingthetreatmentofsludgetoreducepathogensandbacterialregrowth.

“Inthelongrun,itwillbeimportanttoeliminateindustrialwastesthatarereleasedtodomestictreatmentplants,andtoaddressthesustainabilityofusingpotablewater,whichwillbeincreasinglyscarceasclimatechangeoccurs,toflushhumanwaste,”Wingsaid.

Fatty acid metabolite inhibits tumor growth in mice By Sara Mishamandani

AccordingtoarecentNIEHS-fundedstudy,aproductfromthemetabolismofomega-3fattyacidhelpscombatcancer,bycuttingofftheoxygensupplyandnutrientsthatfueltumorgrowth.Withthisfinding,ateamofUniversityofCalifornia,Davis(UCDavis)scientistsprovidesmoreevidenceabouthowomega-3fattyacids,whicharecommonlyfoundincertainfishandplantoils,maybeusedtofightcancer.

“Ourinvestigationopensupanewunderstandingofthepathwaysbywhichomega-3fattyacidsexerttheirbiologiceffect,”saidGuodongZhang,Ph.D.,leadauthorofthestudy.ZhangisapostdoctoralresearcherinthelaboratoryofBruceHammock,Ph.D.,NIEHSSuperfundResearchProgramcenterdirectoratUCDavis.

Fatty acids and tumor growthZhangexplainedthattumorsgrowandspreadbydisruptingthenormalprocessofangiogenesis,theformationofnewbloodvesselsinthebody.Heandhiscolleaguesdeterminedthatthemetaboliteepoxydocosapentaenoicacid(EDP)inhibitsangiogenesis,whichreducesthegrowthandspreadoftumorsinmice.Italsoblocksangiogenesisbyadifferentmechanismthanmanycurrentanti-cancerdrugs.

EDPisproducedinthehumanbodyfromtheomega-3fattyaciddocosahexaenoicacid(DHA),whichisfoundincold-wateroceanicfishoilsandvarioussupplementsandpharmaceuticals.EDPisalsoincreasinglymadeindustrially.

“Asfarasweknow,EDPsarethefirstsignalinglipidsthathavebeendiscoveredtohavesuchpotentanti-cancereffects,”saidZhang.“ResearchersmaybeabletouseEDPsasstructuraltargetstodevelopstableanalogsasanti-canceragents.”

Researchersalsofoundthatametaboliteofarachidonicacid(ARA),anomega-6fattyacid,hastheoppositeeffectofEDP.TheARAmetaboliteslightlyincreasesangiogenesisandtumorprogressioninmice.ARAinthehumanbodyusuallycomesfromdietaryanimalsources,suchasmeat,eggs,anddairy.Thisincreaseinangiogenesisencourageswoundhealingandtissuerepair.

Patent law career optionsKieferandSchwartzmanexplainedthatascientistcouldbehiredbyalawfirmasatechnicalspecialist,patentagent,orpatentattorney.Atechnicalspecialisthasnolawexperienceandisrequiredtounderstandthesciencebehindaninvention.

“Afterall,it’seasiertoteachascientistsomelawthanalawyersomescience,”Schwartzmanpointedout.

Whilebothapatentagentandanattorneyneedtopassapatentbarexam,onlyanattorneyhasalawdegree.Althoughanagentisnotabletosignoffonalllegaldocuments,bothagentsandattorneyspreparepatentapplicationsandcommunicatewiththeU.S.patentoffice.Many,butnotall,lawfirmstreattheiragentsandattorneysthesame,andexpectthesameamountofworkfromthem.Attorneyscananticipatetheirsalariestobehigher,makingtheinvestmentofgoingtolawschoolworthwhile.

ThefourthoptionofpracticingpatentlawistoworkasapatentexaminerfortheUnitedStatesPatentandTrademarkOffice.Examiners,whomustbeU.S.citizens,reviewpatentapplicationsforcompliancewithbasicrulesandlegalrequirements.Referringtohisowntimeattheoffice,Schwartzmancalleditaveryvaluableexperiencethatmakesfindingthenextjobeasy.Heexplainedthatlawfirmsarelookingforformerexaminersfamiliarwiththepatentapplicationprocess,andreviewerswithfouryearsofexperienceareexemptfromthebarexamrequirement.

KieferandSchwartzmandescribedthejobashardandextremelydeadlinedriven.Theworkloadisexpressedinbillablehoursthatcanamountto1,800-2,100ayear.Tobillaneighthourworkday,anagentorattorneymayneedtoputin10-11hours,whichmayincludenonbillablemeetingsandlunchbreaks.But,exceedingexpectationscouldbearoadtopartnershipinalawfirm.

BothKieferandSchwartzmanspokehighlyofjobsatisfaction,suchasthejoysofhavingapatentapplicationapproved,talkingtohighlyengagedinventors,orworkingforastartupcompanythatispurchasedbyabiggercorporationforitsprofitablepatents.

New protein mutated in neurodegenerative disease By Robin Arnette

Wheninjurytothemammaliangenomeoccurs,suchaswhenDNAstrandsbreak,specializedproteinscalledpoly(ADP-ribose)polymerases,orPARPs,raiseanalarm.UponencounteringDNAdamage,PARP1tagsahostofDNAdamagerepairfirstresponderproteins,asameansofcontrollingtheiractivity.ThesemoleculartagsareformedbygeneratingrepeatingchainsofthesmallmoleculeADP-ribose,tocreatepoly(ADP-ribose)orPARtags.

PropercoordinationofresponsestoDNAdamagealsorequirescompleteremovalandrecyclingofsuchtags.Forseveraldecades,though,scientistsdidnotknowwhichproteinclipstheADP-ribosedirectlyattachedtotaggedproteins.Now,ateamthatincludesseveralNIEHSresearchershasidentifiedandcharacterizedonesuchprotein,anduncoveredanothersurprisingdetail—amutationinitsgeneleadstoarareformofneurodegenerativedisease.

Working together to find a solutionTheresearch,whichappearedonlineMarch12inTheEMBO[EuropeanMolecularBiologyOrganization]Journal,wasamultidisciplinaryeffortfromscientistsandcliniciansfromtheUnitedKingdom,UnitedStates,Germany,andIran.ScottWilliams,Ph.D.,oftheNIEHSLaboratoryofStructuralBiology,ledtheNIEHScontingentthatcharacterizedtheactivityandstructureoftheprotein,originallyknownasC6orf130.

Williamssaid,heandhisgroupwereinterestedinC6orf130,becauseitisamemberofapoorlyunderstoodfamilyofproteinsthatcontainADP-riboseinteractingmacrodomains.IncollaborationwithIvanAhel,Ph.D.,ofthePatersonInstituteforCancerResearchattheUniversityofManchester,andRezaSharifi,M.D.,Ph.D.,ofSt.George’sUniversityofLondon,EuropeanteammembersidentifiedamutationintheC6orf130gene.

Whenthemutatedgenewasexpressed,theresultingC6orf130proteinwaschoppedinhalf,renderingitinactive.Asaresult,peoplebornwithtwocopiesofthemutatedgenewereboundtowheelchairs.Theysufferedseizures,anddisplayedalackofatendonreflexandapartialabsenceofaswallowingreflex.

“WerenamedC6orf130,terminalADP-riboseproteinglycohydrolase1,orTARG1,becauseitcleavesatthestemofwheretheADP-ribosemodificationismadeonaprotein,”Williamssaid.“TheneurodegenerativeconditionthatoccursisaTARG1deficiency.”

Sofar,theonlypeoplewhohavetheunnamedillnesscomefrommembersofonefamilyinIran,butWilliamssaidothermutationsinTARG1mayproduceotherinfirmities,somoreresearchisneeded.

Seeing TARG1 in actionUsingX-raycrystallography,aswellasavarietyofbiochemicaltechniques,Williams’researchgroup,whichincludedDeniseAppel,JunoKrahn,Ph.D.,andMatthewSchellenberg,Ph.D.,generatedhigh-resolutionmolecularsnapshotsshowinghowTARG1engagedandprocessedtheterminalADP-ribose.Withadditional

AppelisabiologistintheWilliamsgroupandsharedfirstauthorshiponthejournalarticle.Shesaid,definingthemolecularmechanismofTARG1andunderstandinghowonemutationinitsgeneledtodiseasewassignificant,butthesestudiesmayalsohavethepotentialtohelpcancerpatients.

“Researcherscanselectivelykilltumorcellsbyaffectingtheirabilitytometabolizepoly(ADP-ribose)correctly,”Appelsaid.“SinceTARG1actstodegradepoly(ADP-ribose),itmaybeanattractivetargetfordrugdevelopmentincancerchemotherapy.”

WilliamswasjustasexcitedabouttheTARG1research,andsaidonerarelyseesastorylaidoutinonepublication—identificationofacriticalenzymaticactivityandtheproteininvolved,atomicresolutionsnapshotsofitsmodeofaction,andthedocumentationoftheindividualswithinthehumanpopulationwithdefectsinthatactivity.

“It’safineexampleofscience’smovetowardpowerfulmultidisciplinaryresearch,”Williamssaid.“It’soneofthemostcomprehensiveprojectsI’veeverbeeninvolvedin.”

Community beliefs regarding dioxin exposure pathwaysAstudy,partiallyfundedbyNIEHS,foundthatresidentsofcommunitieswithknowndioxincontaminationmadeincorrectassumptionsabouthowandwheretheymightbeexposedtodioxins.Despitethefactthatdioxinsaggregate,ratherthandisperse,inwater,79.3percentofsurveyrespondentsincorrectlybelievedthatdioxinscouldbefoundinriverwater,evenaftertheremovalofallsoilandsediment.Thestudyshowstheimportanceofclearlycommunicatingthespecificexposurepathwaysofcontaminants.

Theresearchersmailedsurveysto904Michiganresidentswholivedincommunitieswheredioxincontaminationhadoccurred.Theresearchteampointedoutthatresidentshaverepeatedlyheardthatindustrialdischargeintotheriverwasthesourceofmostofthedioxincontamination.Asaresult,theresidentsconcludedtheriverwaterwastainted,eventhoughdioxinswerelikelytoquicklybindtoparticlesinwaterandaccumulateinsedimentatthebottom.

Forfuturecommunicationwithresidents,theresearcherssuggestedusingananalogy,suchasdioxinsaremorelikeoilthanwater,tohelpmitigateconcernsaboutdrinkingwaterandcontactwithriverwater.

TheresearcherslookedaturinaryBPAconcentrationfor55infants,andcategorizedeachinfant’smedicaldeviceuseasloworhigh,basedonthenumberandinvasivenessofdevices.TheyfoundthatthemedianurinarytotalBPAconcentrationofinfants,whohadrequiredfourormoremedicaldevices,forthreepreviousdays,wassignificantlyhigherthanforinfantsrequiringthreeorlessdevices.Theresearchersalsocollectedandanalyzedbreastmilkorformulasamples,andfoundthatincreasedBPAconcentrationwasnotassociatedwiththeinfants’nutritionalintake.

A novel mechanism underlies glucocorticoid resistanceArecentstudyonglucocorticoidresistance,conductedbyresearchersatNIEHS,identifiedanovelmechanismthatcouldhavebroadimplicationsforvariousdisorders,suchasinflammation,autoimmunediseases,andcancer.Glucocorticoidresistanceiscausedbytherepressionofglucocorticoidreceptor(GR)genetranscriptionbyglucocorticoids.Forthefirsttime,theydemonstratedthatGRDNAsequencescouldactasaproteincodingsequenceintheabsenceofglucocorticoids,butrepresstheexpressionofthesamegeneintheirpresence.

Glucocorticoidresistanceoftenaffectspatientsundergoinglong-termorhigh-doseglucocorticoidtreatmentforinflammatorydisorders.Usingbothmouseandhumancells,theauthorshaveshownthatbindingofligand-boundGRtoafunctionalnegativeglucocorticoidresponseelement(nGRE)inexon6enablesassemblyofaGR-NCoR1-histonedeacetylase3-containingrepressioncomplex.Theassemblyismediatedbychromatinloopingoftheintragenicelementsatthetranscriptionalstartsite,therebyinhibitingtranscriptionalinitiation.

TheauthorsproposethatGRanditsligandcancoordinatetherepressionofGRtranscriptionbasedontheirconcentration,irrespectiveofthecombinatorialassociationsoftissue-specifictranscriptionfactors.Thus,theirstudysuggeststhatlong-termglucocorticoidadministrationmayleadtoconstitutiverepressionofGRgenetranscriptionandtoglucocorticoidresistance.(MM)

COX-2 involved in the development and worsening of asthma symptomsNIEHSscientistshavediscoveredthattheenzyme,cyclooxygenase-2(COX-2),isresponsibleforregulatingaspecificallergicresponseinpersonswithasthma.TheyfoundthatCOX-2blocksanimmunecelltypethatgathersaroundtheairwaywhenanallergicreactionoccurs.Theimmunecell,calledThelpertype9(Th9),normallyfightsinfection,butalsoreleasessmallmessengermoleculesthatpromoteinflammation,whichinturn,intensifiesasthmaticsymptoms.

Theteamtestedtheallergicresponseinthreesetsofmice—COX-2knockoutmice,normalmicegivenCOX-2inhibitors,andwild-typemicewithfullyfunctionalCOX-2.Themicewereexposedtoovalbumin,themainproteinineggwhites,togenerateanallergicreaction,andthentheteammeasuredtheTh9cellcount.ThewildtypemiceshowedlowTh9cellcounts,whereastheinhibitedCOX-2miceandthemicewithouttheCOX-2genehadsignificantlyincreasednumbersofTh9cells.

Thescientistswerealsoabletoreplicatetheirfindingsinhumans.TheydeterminedthatasthmaticpatientshadsignificantlyhighernumbersofcirculatingTh9cells,comparedtotheirnonasthmaticcounterparts.(MK)

Early life socioeconomic factors influence the development of rheumatoid arthritisUsingdatafromtheNIEHSSisterStudy,Instituteresearchershaveexaminedearlylifeandcumulativelife-coursesocioeconomicstatus(SES),andfoundconnectionswithadult-onsetrheumatoidarthritis(RA).ThisstudyprovidesadditionalevidencethatRAmaybelinkedwithloweradulteducationalattainmentandoccupationalclass,andisthefirsttoreportassociationsofRAwithyoungermaternalageandchildhoodfoodinsecurity.

Thestudyincluded50,884women,aged35-74yearswhentheyenrolledintheSisterStudy,anationwidecohort.WomenwereaskedifadoctorhadeverdiagnosedthemwithRA,theirageatdiagnosis,andspecificsymptomsandmedicationuse.InvestigatorsfoundthatseveraladversechildhoodSESfactorsweremorecommoninRAcases—youngmaternalage,foodinsecurity,andlowerparentaleducationandhouseholdincome—withasignificantdose-responseacrossanincreasingnumberoffactors.Thisassociationremainedinwomenwithloweradulteducationalattainmentandwasindependentfromage,race,andsmoking.

ThefindingswereconsistentwithacumulativeeffectoflowerSESthroughoutthelifespan.Researcherssuggestthatthesefindingssupportfurtherinvestigationofearlyandsustainedsocioeconomicadversity.(BS)

DNAlesions,whicharecreatedbyreactiveoxygenspeciesafterexposuretoenvironmentalstress,canleadtocancer-causingmutagenesis,ifnotrepairedthroughbaseexcisionrepairandotherDNArepairpathways.8-oxoGisconsideredthemostabundantoxidativestress-inducedlesioninDNA.When8-oxoGisinthetemplatebasepositionduringDNAsynthesis,itcanbindtoDNApolymerasebetaintwodifferentconformations.Thisdualbindingcapacityleadstoeitheramutagenic(syn)pairingwiththeincomingadeninebase,ornon-mutagenic(anti)basepairingwiththeincomingcytosinebase.

Byintroducingapointmutationintothepolymeraseactivesitethatdestabilizesthesyn-conformation,theresearcherswereabletocomparethespecificityandkineticsofinsertionwithatemplating8-oxoGinthemutantandwild-type.TheyfoundthemutantDNApolymerasetobelesserror-pronethanthewild-typeenzyme,reflectingmorefrequentincorporationofcytosineopposite8-oxoG.

Themutantpolymerasestructure,incomplexwiththe8-oxoG-containingtemplatebaseandanincomingbase,revealedthatDNAminorgrooveinteractionsbythepolymerasearenecessarytostabilizethesyn-conformationof8-oxoG.Thus,thesingleactive-sitemutationchangedthecodingpotentialoftheenzymebymodulatingthe8-oxoGlesionconformation.(MF)

Inside the InstituteNIEHS ends weeklong Earth Day celebration with local highway adoption By Ian Thomas

NIEHSconcludeditsweeklongcelebrationofEarthDayApril26byadoptingthe1.2-milestretchofHopsonRoadbetweenthemaincampusandKeystonebuildinginResearchTrianglePark(RTP),N.C.AspartoftheAdopt-A-HighwayProgram,theeffortisintendedtoreducelitteronNorthCarolinaroadsides,andhelppreservethenaturalbeautyofthestate.

“Thisprogramletsouremployeestakeanactiveandimmediatepartinprotectingourenvironment,”saidPaulJohnson,amemberoftheNIEHSEnvironmentalAwarenessAdvisoryCommittee,whodonnedanorangevestwithseveralofhiscolleaguestopickuptrash.“Atthesametime,italsosetstheexampleforourcommunitythatit’suptoustokeepourhomebeautiful.”

Establishedin1988,asaresponsetogrowingpublicconcernsonlitter,theAdopt-A-HighwayProgramismanagedbytheNorthCarolinaDepartmentofTransportation,andcurrentlyhostsrepresentativesfromgovernmentandprivate-sectorentitiesacrossthestate.

A week of educationInadditiontothehighwayadoption,NIEHSstaffmembersweretreatedtoanumberofothereducationaleventsthroughouttheweek.Amongthemwasaseminar,“Theeffectsofinvasiveplantspeciesintheaquaticenvironment,”byRobEmens,anaquaticplantspecialistwiththeNorthCarolinaDepartmentofEnvironmentandNaturalResources(NCDENR).Thehour-longtalkexplainedhowcertaininvasiveplantspecies,suchasHydrilla,canhavedevastatingecologicaleffectsonlocallakesandwaterways.

“Ihadnoideahowquicklysomeofthesespeciescoulddisplacethenativeplantsinagivenecosystem,”notedJohnson.“Thatreallyunderscoresthesophisticationoftheworldaroundus,andhowinterconnectedeverythingis.”
