Protected amino acids are pre-packaged with equimolaramounts of dry activator (e.g. HBTU) in a specially designedvial*. The dry mixture is shelf-stable for years. Upon additionof solvent to the vial, activation takes place immediately. Theactive HOBT-ester is quickly transferred to the reaction vesseland a highly efficient coupling ensues.
The PS-3 takes advantage of this amino acid delivery method bydrastically reducing the number of hardware componentsrequired for automated synthesis.
