When climbing, standard pressure (1013.25 hPa, also known as QNE) is set on the altimeter when passing 3000 feet. On descent, the local altimeter setting is set passing the transition level. Above the transition layer (determined by the controller based on atmospheric pressure at the airport -- the QNH), altitudes are given as flight levels. One flight level is 100 feet, so flight level 60 (written FL60) is 6000 feet as displayed by an altimeter set to standard pressure.
