EDUCATIONAL TRAINING WORKSHOPS − Must pre-register in order to attend −

WORKSHOP 1 8:30 am–12:30 pm Location: Commonwealth A-1 (2nd floor) Preparative Chromatography for the Purification of Intermediates and API Lecturers: Dr. Kathleen Mihllbachler, Bristol-Meyers Squibb, and Dr. Olivier Dapremont, Ampac Fine Chemicals This workshop will focus on development of method for the preparative purification of small molecules for the pharmaceutical industry. After an introduction of the basics of liquid chromatography for small molecule, the instructors will present a practical approach to the development of preparative separation for Batch HPLC and Simulated Moving Bed through a series of examples. The attendees will learn valuable information and techniques to apply in the laboratory to increase throughput and performances. A book of the course will be provided to each attended at completion of the training. The book will contain copies of the slides as well as additional tables of data relevant to preparative chromatography. Dr. Dapremont has worked on the development of SMB technology since 1992. He is Director of Process Technologies at AMPAC Fine Chemicals where his role encompasses the development of SMB separations using multiple SMB units ranging from 4.6 mm to 1 m in diameter as well as developing continuous processes for the manufacturing of APIs. Bio: Ph.D. from the University Pierre & Marie Curie in Paris; Prochrom (France), a manufacturer of chromatographic equipment; Chiral Technologies Europe (France), in charge of kilo scale separation service using SMB; and Aerojet Fine Chemicals, now AMPAC Fine Chemicals. Dr. Dapremont is co-author of a dozen publications and 5 patents related to SMB technologies. Dr. Mihlbachler has worked in the field of separation technology with an emphasis on chromatography for the last 15 years. In her current role as a Sr. Research Investigator II in PRD at BMS (New Brunswick, NJ), she develops purification/separation processes for pharmaceutical intermediates and API, in particular for chiral compounds and peptides. Dr. Mihlbachler held positions at Pharmacia/Pfizer (kg-scale chiral separation) and Eli Lilly and Company where she also evaluated multi-column continuous chromatography for bio-molecule purification. Dr. Mihlbachler received a doctoral degree in chemical engineering from the O.-v.-Guericke University Magdeburg, Germany, under the direction of Profs. Guiochon and Seidel-Morgenstern. She authored a chapter on SMB in “Advances in Large-Scale Biopharmaceutical Manufacturing and Scale-Up Production” and co-authored 11 articles. Since 2003 she has co-lectured workshops on “Adsorption Isotherms". She has also served as the 2009-2010 Vice-Chair of section Bioseparations at AIChE.

WORKSHOP 2 1:30–5:30 pm Location: Commonwealth A-1 (2nd floor) Bringing Biomolecules to Market (and keeping them there) Lecturers: Dr. Joan Newburger, Johnson & Johnson Consumer Products Worldwide, and Ms. Lois Ann Beaver, LAB Enterprises New drug products start with innovation. Frequently the most impactful drugs addressing critical medical needs, must overcome significant process and regulatory hurdles to gain approval and reach the patient. Everyone in the industry understands that the costs and timeframes are huge, that numerous problems can derail progress and that setbacks can affect the viability of the product or even the viability of the business itself. The outcome of a decade’s long and continuing concerted effort by members of the regulatory community and industry carried out within the ICH (International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use) and in other venues, (ISPE, WHO, AAPS, International Society of Pharmaceutical Engineers, World Health Organization, American Association of Pharmaceutical Scientists) resulted in certain approaches derived from principles of Quality and Risk Management that encompass all aspects of bringing drugs to market, from product development to commercial production. Quality by Design, Quality Systems Management, Science-based Risk Management., Process Analytical Technology and Continuous Process Improvement are tools that can be incorporated into basic drug development processes. The potential benefits to industry are diverse and can include lower production costs, faster approvals, fewer recalls, new patents and increased regulatory flexibility. Time and dollars invested in implementation should yield measurable value. In this workshop we will examine the relationship between good drug development science, quality and regulatory flexibility, with emphasis on application to the biopharmaceutical industry. Particular attention will be placed on the practice of Quality by Design. A practical study of how the integration of quality and risk management concepts and tools is used in an application to bring a MAb to market will be presented. In addition, we will address the challenges for technology and regulation created by the emerging biosimilars industry along with the current status of internationally developed guidance. Dr. Newburger has worked in pharmaceutical development for over 25 years at Wyeth Ayerst, Bristol-Myers Squibb and, currently, Johnson & Johnson. During the first half of her career, she led separations groups supporting medicinal, formulations and process chemists. For the past 7+ years, Dr. Newburger has led cross-functional teams, including clinical, CM&C, toxicology, regulatory and commercial members, having the responsibility of developing and implementing strategies for all Phases of prescription drug development and post-Approval support. Dr. Newburger has a Ph.D. in organic chemistry from the University of New Hampshire, where she was one of the first practitioners of "modern" preparative HPLC, using a prototype of the first commercialized preparative HPLC instrument (Waters Associates Prep 500). While at Wyeth-Ayerst, Dr. Newburger began collaborating with Professor Georges Guiochon on practical applications of non-linear chromatography. She is the principal author of 4 research publications and has presented 12 invited papers on her research and on drug development. Since 1994, Dr. Newburger has taught 18 workshops on the topics of preparative chromatography and FDA CM&C Initiatives. After a multi-decade career with the U.S. Food and Drug Administration, in 2009 Lois Ann Beaver founded a regulatory consulting group, LAB Enterprises. While working in FDA’s Office of the Commissioner, Lois served on the FDA Pharmaceutical Quality Council that conceived and developed quality by design; led activity for FDA’s joining the international Pharmaceutical Inspection Cooperation Scheme; worked as liaison with international organizations such as WHO, most recently focusing on biosimilars; managed the export program for international investigations of unapproved pharmaceuticals; was associate director for international harmonization working on international best practice and standardization activities such as the ICH; was US Coordinator for Veterinary ICH; and served as delegate to the APEC (Asia Pacific Economic Cooperation) Life Sciences Innovation Forum. Bringing Biomolecules to Market (and keeping them there) (continued) Lois also led international projects on anti-counterfeiting of medical products and good manufacturing practices in pharmaceutical firms in emerging countries, and established a pharmaceutical information center in Cairo. With degrees in chemistry and mathematics, Lois joined FDA as an analytical chemist in the lab in Buffalo, NY then was invited to FDA regulatory headquarters to oversee nationwide trace analysis of chemical residues and contaminants. While at FDA, she took advanced studies in analytical chemistry and international law at Georgetown University, and numerous courses at the State Department Foreign Services Institute. During her career, she functioned as a compliance officer advising on FDA law, as a program officer developing a management information system, and as a congressional liaison officer. While working at FDA, she co-founded a successful company that produced preparative chromatographs and analytical instruments that was sold after 10 years.
