To protect your HPLC column from contamination, guard columns should be used ideally. Guard columns hold back particles and molecules that could form an irreversible bond with the column material. Guard columns should be replaced regularly. A good indication of the date of exchange is a gradual rise in the back pressure. When using the Altmann Analytik guard column cartridges only the cartridge must be replaced, not the entire guard column. Since cartridges are cheaper than complete guard columns you can save costs!

How can I find a suitable HPLC column?

THE PATH TO A SUITABLE COLUMN DEPENDING ON THE SEPARATION PROBLEM Since the beginning of high performance liquid chromatography (HPLC) in the 1960s, the method has been continuously developed and refined. This applies equally well to the materials used: We carry over 60,000 different HPLC columns by many different manufacturers. This selection has naturally great advantages - among others there are very specific columns for almost every separation problem - simultaneously, the user is spoilt for choice. This text can provide guidance and help in choosing the appropriate HPLC column. HARDWARE OF THE COLUMN:Most HPLC columns are made of 316 steel. Advantage is that the steel is pressure-resistant and also relatively inert to corrosion. For a good separation efficiency, it is important that the inside of a separation column has no roughness, grooves or micro porous structures (Meyer 2009, p. 110).

Base material SILICAGEL:A popular base material in HPLC columns is silica gel; this consists of Si-atoms which are bridged by oxygen atoms (Meyer 2009, p. 119). A disadvantage of silica is that a high pH dissolves the silica skeleton.

Silica gel columns are usable in the pH range from pH 1 to 8 (Meyer 2009, p 120). With a low metal content the chemical stability of high-purity silica gels can be increased (Meyer 2009, p 125). On the surface silica carries OH groups (silanol groups). These silanol groups can be chemically modified so that stationary phases with specific properties can be obtained (Meyer 2009, p 121). In addition to mechanical stability, Silica has the advantage of a low cost and easy synthesis. Like-wise, the surface modification can be made simple, inexpensive and flexible. The pore size in silica can be adjusted to be closely distributed (Kromidas 2014 p.246).

POLYMER COLUMNS:For the RP-HPLC (reversed phase HPLC) polymer-based HPLC columns are also available. Often polystyrene-divinylbenzene columns are used, which have a higher pH stability compared to the silica gel columns (pH 1-13). For highly acidic or basic eluents they therefore represent an interesting alternative (HPLC column 2016). Especially for the gel permeation chromato-graphy styrenedivinylbenzene is a major stationary phase (Meyer, 2009, p 126).

Shape of the column material FULLY POROUS PARTICLES:Nowadays - besides the traditional fully porous phases – there are also Core shell phases, monolithic phases and phases for UHPLC with small particle sizes available.

Most commonly used particles in HPLC are still fully porous particles. Conventional particle sizes are 3, 5, or rarely even 10 µm. In case of fully porous particles the whole internal structure is porous and can be compared with e.g. a sponge (Meyer 2009, p 116).

CORESHELL PARTICLES:Core-shell particles have a solid core and only the outer shell is porous. This solid core leads to an increased separation efficiency. The particles have a uniform particle size distribution and can therefore be packed better, the A term of the Van Deemter equation is lowered (Figure 1, page 5). In addition, the mass transfer of the molecules in the pores (C-term of the Van Deemter equation) is accelerated. A disadvantage of the core shell particles is the reduced loading capacity compared to fully porous materials.

MONOLITHIC PHASES:Monolithic columns are made of a single piece of porous material, e.g. silica or organic polymer. Thus the chromatographic bed does not consist of individual particles but of a porous rod. Monoliths have a separation performance that is similar to the 3 µm particles packed columns (Meyer, 2009 p.118). So far one disadvantage is that with monolithic phases the availa-bility of selectivities is still limited as compared to total porous materials (Kromidas 2014 p.152).

BONDED PHASE:The large number of available stationary phases (reversed phase, normal phase, phenyl phase ...) of various manufacturers aids in not always making it easy to quickly find the appropriate HPLC column. For many analyses a common reversed phase C18 column can be used. In order to decide which stationary phase is suitable the first step should be to have a look at the molecule to be examined. If one wishes to analyze a very polar molecule there is a possibility that it does not interact with the C18 material and thus elute in dead time. For instance C4, CN, or phenyl-diol phases are suitable in this case. If however the analyte is too non-polar it can irreversibly adhere to the material. To avoid this you can try a silica, CN or NH2 phase. This usually happens when the analyte is only soluble in heptane or hexane (Kromidas 1997 S. 32-33). If that decision is made the first hurdle has been overcome. Now however it is even more complicated because you have to choose between different dimensions and specifications to use for the column such as particle size, pore size, internal diameter, diameter and carbon content.

OVERVIEW BONDED PHASES (HPLC COLUMNS 2016)

Particle size Smaller particle sizes result in better resolution than larger particles. However larger particles produce a lower back pressu-re in the HPLC system. In addition to the size of the particle a uniform distribution of the particles is particularly important. By an increased proportion of small particles the back pressure is increased; if there are too many large particles the mate-rial loses separation performance. If you have a wide particle size distribution the material can be packed poorly (Kromidas 2014 p. 290). The size of the particle has an impact on the quality of the resolution since the size of the particles affect the A and C-term of the Van Deemter equation.

TERMS OF THE VAN-DEEMTER EQUATION (CHEMGEPEDIA 2016) Pore size The pore size to be used depends on the size of the molecule to be analyzed: the larger the molecule the greater the pore should be. Pore sizes are usually given in Å, thereby 10Å = 1 nm. For small molecules a column up to 120 Å can be used, for large biomolecules e.g. columns with pore sizes of 300 Å are suitable (Kromidas 2014, p. 291). For biomolecules choosing the right pore size is particularly important as they are available in a wide size range. The smaller the pore, the more surface is available for interaction Thus it can be loaded higher (Kromidas 2014 p. 291).

Optimal separation performance is obtained at the smallest possible surface level, thereby the total number of theoretical trays is increased in the column. Especially when the A- and C-term can be minimized, it is possible to achieve a good sepa-ration performance over a large flow range.
