How to operate the API 2000 for a single Mass Spectrum
measurement?

Booking of the instrument.
1. Sample LOGIN.
2. Instrument standby � ready with suitable Method.
3. Background observation at a desired m/z range.
4. DILUTED Sample injection.
5. CLEAN-UP.
6. Data observation and saving.
7. Instrument ready � standby.

Did you book the instrument?
Is the instrument free to use?
If yes, register your sample in the LOGBOOK and start the application “Analyst”.
Then the following text dialog box appears:
1- Click once on “Configure”.
and
2- Double-click on “Hardware Configuration”.
1- Select “MassSpec & Syringe”.
and
2- Click on “Activate Profile”.

A “green flag” appears in front of the activated hardware profile chosen, as shown below.
Click on “Close”.
1- Double-click on “Manual Tuning”.
The following dialog box appears
And gas turns on
Press “OK”.
2- Double-click again on “Manual Tuning”.

The Tune Method Editor page appears, as shown below.
A new Tune Method Editor page appears, as shown below.
1- Ctrl+O.
Open a File dialog box appears
2- Select Q1MS Pos
or Q1MS Neg method.
3- Press OK.
4- Press No (about saving
method).
The syringe is already loaded with clean-up solvent.
3- Click on “ Start ” (ACQ).
1- Enter target (s) mass ; with 100 of width .
2- Click on “Start syringe pump”.
4- Observation of mass spectrum background at the desired m/z range.
5- Stop ACQ and Syringe pump.

3- Click on “Start” (ACQ).
2- Click on “Start syringe pump”.
4- After 20 sec , “Stop syringe pump”
and Reload FULLY with suitable clean-up solvent.
1- Bin the clean-up solvent and half load the syringe with
DILUTED SAMPLE (10-20µg.mL-1
) = (1mg. mL-1
diluted 50-100x).
5- Then “Start syringe pump” again.
6- Stop Syringe pump and ACQ, after observation of ions of interest and clean-up process .
10-20 µg.mL-1
<=> 1-2 ng. µL-1
Example: 10µL of sample solution (1mg. mL-1
) + 740µL of suitable
solvent with 0.1% formic acid.

TIC (Total Ionic Current) pane.
Right-Click on “MS pane”.
Select “Open file”.

Select MS pane by one left-click in MS pane (blue frame).
Click on for closing MS pane.
- Select the part of TIC with the mouse left-button.
- Then right-click once in the area chosen.

Then we obtain the following workspace :
Select “show spectrum”.
An average mass spectrum covering the TIC area chosen appears.

Save a mass spectrum:
Print-Screen
Open a WORD or PPT new file and paste (+ Crop picture out)
Save the WORD or PPT document in the following link:
C:\QTRAP 2010\Name_of_supervisor\MS requester\name_of_datafile
1- From Tune Editor page, double-click on
2- Browse.
3- Select file of interest among the last files recorded (sorted by date and time) C:\analyst data\Project\API instrument\Tuning cache

Closing down my session:
- Close panes and windows (WITHOUT saving METHOD CHANGES)

1- Click once on “Configure”.
and
2- Double-click on “Hardware Configuration”.
1- Select “MassSpec & Syringe”.
2- Click on “Deactivate Profile”.
3- Close
and
Close the entire application.
Shut down the monitor.
Leave the computer on.
