Naming fungi involved in spoilage of food, drink, and water

Correct identifications are the key to all data on an organism. Fungi cultured or sequenced from a foodstuff may not always be the spoilageagents, and there are potential pit-falls in culturing and molecular identification. Molecular phylogenetics is resulting in major changes infungal classification, and substantial changes in the rules on naming fungi were agreed in 2011. Different morphs of a single pleomorphicspecies can no longer have different scientific names, and stability is to be fostered through lists of protected names. The naming of fungi isbecoming increasingly fit-for-purpose by taking advantage of the possibilities arising from advances in molecular and digital technologies. Alist of the current names of 100 species of spoilage fungi is included.

Communities of arbuscular mycorrhizal fungi on a vegetation gradient in tropicalcoastal dunes

The community composition of arbuscular mycorrhizal fungi (AMF) was studied related to AMF species richness, abundance and frequency incoastal dune areas, northeastern Brazil. Soil samples were collected periodically in 2010 and 2011 in three dune areas (herbaceous, shrubbyand arboreal) that form an environmental gradient from the beach to the inland tropical rainforest. Fifty AMF species were identifiedbelonging to 18 glomeromycotan genera and 10 families. Several remarkable observations were made: higher AMF spore density wasobserved in the arboreal dune compared to the shrubby and herbaceous dunes, while the highest number of infective propagules wasobserved in the herbaceous dune, followed by the shrubby and arboreal dunes. Although AMF species richness and AMF root colonizationwere similar in all dunes, the Shannon's diversity and Pielou's evenness indices were higher in the herbaceous dune than in the shrubby andarboreal dunes, which was the opposite in relation to the Simpson dominance index. Thus, surprisingly, the areas closest to the sea hadgreater diversity compared to the later successional dunes in farer distance from the sea. Funneliformis halonatus was the indicator speciesof the arboreal dune, Ambispora appendicula, Gigaspora gigantea and Paradentiscutata maritima were the indicator species of the shrubbydune, while two undescribed Cetraspora species and Sclerocystis sinuosa were the indicator species of the herbaceous dune. The studyareas can be considered as hotspots for the conservation of diversity of AMF.

Abstract: Spadicoides sylvatica sp. nov. and Sporidesmiella mammillata sp. nov., both collected on decaying plant material in a Mexican drytropical forest, are described and illustrated. Spadicoides sylvatica is distinguished by its ovate to obclavate, 3–4-septate, smooth, goldenbrown to brown conidia. Sporidesmiella mammillata is characterized by conidia that are solitary acrogenous, obclavate to fusiform, sub-rostrate, 4–7-distoseptate, smooth, and golden brown.

Abstract: Endophragmiella selenosporellaria sp. nov. was collected on dead branches of an unidentified broadleaf tree from southern Chinaand is characterized by its branched conidiophores and Selenosporella-like synanamorph developed from the apical cell of the conidium. Abstract: A new wood-decaying polypore, Melanoderma disciforme, is described from tropical China based on morphological characteristicsand rDNA ITS sequences. The new species is characterized by perennial effuse-reflexed basidiocarps, a narrow pileus with a thin black cruston the pileal surface, a dimitic hyphal system with dextrinoid and cyanophilous skeletal hyphae, and the presence of fusiform cystidioles andcylindrical basidiospores. Molecular analyses indicate that the new species and M. microcarpum cluster together with full support.

Abstract: Eight poorly known or unusual crustose and pileate basidiomycete species were studied. These included the type specimens ofthree monotypic genera: Chrysoderma alboluteum from Réunion is conspecific with Cerocorticium molle; Dendrophysellum amurense fromthe Russian Far East is a species of Vararia; and Hyphoradulum conspicuum belongs in Pseudolagarobasidium and is the first representativeof the genus from Europe. Corticium murrillii, from Mexico, is congeneric with Corneromyces kinabalui. New combinations Varariaamurensis, Pseudolagarobasidium conspicuum, and Corneromyces murrillii are proposed. Mycobonia flava and M. brunneoleuca aremacroscopically similar species with diagnostically distinct basidiospore shape and size. Mycobonia disciformis is accepted in Mycothele, andMycobonia winkleri represents a species of unknown affinities. Recent molecular phylogenetic studies indicate that Mycobonia is embeddedin Polyporus sensu stricto and is a synonym of Polyporus. Transferring M. brunneoleuca and M. flava to Polyporus requires the creation ofthe replacement names, P. polyacanthophorus and P. epitheloides.

Abstract: Two new species of Rosellinia from China are illustrated and described. Rosellinia sigmoidea differs from other species mainly inhaving a white entostroma and broadly rounded ascospores with sigmoid germ slits extending over half of the spore length. Roselliniacamphorae is unique because of its large ascus apical apparatus and ascospores with slimy sheaths. The herbarium and living culture aredeposited in the Collection of Guizhou University (GZUC).

BackgroundAmong the understudied fungi found in nature are those living in close association with social and solitary bees. The bee-specialist genera Bettsia, Ascosphaera and Eremascus are remarkable not only for their specialized niche but also for their simple fruitingbodies or ascocarps, which are morphologically anomalous in Pezizomycotina. Bettsia and Ascosphaera are characterized by a unicellularcyst-like cleistothecium known as a spore cyst, while Eremascus is characterized by completely naked asci, or asci not formed within aprotective ascocarp. Before molecular phylogenetics the placement of these genera within Pezizomycotina remained tentative;morphological characters were misleading because they do not produce multicellular ascocarps, a defining character of Pezizomycotina.Because of their unique fruiting bodies, the close relationship of these bee-specialist fungi and their monophyly appeared certain. However,recent molecular studies have shown that Bettsia is not closely related to Ascosphaera. In this study, I isolated the very rare fungusEremascus fertilis (Ascomycota, Pezizomycotina) from the bee bread of honey bees. These isolates represent the second report of E. fertilisboth in nature and in the honey bee hive. To establish the systematic position of E. fertilis and Bettsia alvei, I performed phylogeneticanalyses of nuclear ribosomal LSU + SSU DNA sequences from these species and 63 additional ascomycetes. ResultsThe phylogeneticanalyses revealed that Eremascus is not monophyletic. Eremascus albus is closely related to Ascosphaera in Eurotiomycetes while E. fertilisbelongs in Myxotrichaceae, a putative member of Leotiomycetes. Bettsia is not closely related to Ascosphaera and like E. fertilis apparentlybelongs in Leotiomycetes. It grows on volcanicrock in temperate region. ITS nrDNA sequence analysis supports the taxonomic distinctness of this species.

Abstract: Eleven Graphis species are newly recorded from Hong Kong. Graphis hongkongensis is new to science, G. centrifuga and G.hyphosa are new to China, and G. assimilis, G. descissa, G. duplicata, G. immersella, G. immersicans, G. japonica, G. parallela, and G.plagiocarpa are new to Hong Kong. Graphis hongkongensis possesses lineola-morph lirellae, a completely carbonized exciple, small tomedium-sized ascospores, and the presence of stictic acid. A brief description of each species is given.

Abstract: Species of the genus Pleurotus are very important edible mushrooms and many of them can be cultivated in commercial scale.Although P. abieticola was originally described from Russian Far East, and then reported from northeastern China and northwestern Russia,its distribution range is still largely unknown. Our morphological and molecular phylogenetic evidence indicated that this species is alsodistributed in subalpine mountains of southwestern China. This paper documented the taxon based on morphological and ecologicalfeatures, and DNA sequences generated from materials collected from Sichuan Province and the Tibet Autonomous Region.

Abstract Research over the past two decades shows that both recombination and clonality are likely to contribute to the reproduction of allfungi. This view of fungi is different from the historical and still commonly held view that a large fraction of fungi are exclusively clonal andthat some fungi have been exclusively clonal for hundreds of millions of years. Here, we first will consider how these two historical viewshave changed. Then we will examine the impact on fungal research of the concept of restrained recombination [Tibayrenc M, Ayala FJ(2012) Proc Natl Acad Sci USA 109 (48):E3305–E3313]. Using animal and human pathogenic fungi, we examine extrinsic restraints onrecombination associated with bottlenecks in genetic variation caused by geographic dispersal and extrinsic restraints caused by shifts inreproductive mode associated with either disease transmission or hybridization. Using species of the model yeast Saccharomyces and themodel filamentous fungus Neurospora, we examine intrinsic restraints on recombination associated with mating systems that range fromstrictly clonal at one extreme to fully outbreeding at the other and those that lie between, including selfing and inbreeding. We also considerthe effect of nomenclature on perception of reproductive mode and a means of comparing the relative impact of clonality and recombinationon fungal populations. Last, we consider a recent hypothesis suggesting that fungi thought to have the most severe intrinsic constraints onrecombination actually may have the fewest.

Abstract: A name has been found for a common species in Pholiota subg. Flammuloides fruiting during and soon after snowmelt in thesubalpine Abies forests of California: Pholiota olivaceophylla is characterized by rather pale slime-covered basidiocarps, relatively pale brownellipsoid to slightly phaseoliform spores, 6.0–8.5 × 3.5–5.0 μm, with an inconspicuous germ pore, and abundant lageniform pleurocystidia.The ITS sequence of the type collection of Ph. olivaceophylla matches those of recent collections. From phylogenetic analyses andmorphology, it is clear that the secotioid Nivatogastrium nubigenum [= Pholiota nubigena] is nested within Pholiota; this species hasretained ballistospores and the typical curved sterigmata for active spore dispersal.

Production of oospores by Peronospora effusa, the causal agent of downy mildew on spinach (Spinacia oleracea), was reported on spinachseed over three decades ago. In view of the rapid proliferation of new races of P. effusa worldwide, seedborne transmission of this pathogenhas been suspected, but methods to test the viability of seedborne oospores have not been available. Eighty-two seed lots of contemporaryspinach cultivars were evaluated for the presence of P. effusa using a seed-wash method and the sediment was examined by microscopy.Sixteen percent of the analyzed seed lots were positive for oospores and an additional 6% for sporangiophores characteristic of P. effusa.Application of a P. effusa-specific quantitative PCR (qPCR) assay showed that 95% of the 59 tested seed lots were positive for P. effusa. Theviability of oospores from five seed lots that were proven to carry the pathogen from the above tests was tested using two independentmethods, one involving plasmolysis and the other by trypan blue staining. The oospores plasmolyzed in 4 M sodium chloride and were de-plasmolyzed in water, demonstrating an active and viable cell membrane. Similarly, viable oospores failed to take up the trypan blue stain.Overall, 59% of the oospores were viable in the plasmolysis test, and 45% with the trypan blue test. These results indicate the presence ofP. effusa oospores in contemporary spinach seed lots, and suggest that the transmission of viable oospores of P. effusa in spinach seed canoccur. Elimination of the pathogen on seed, in addition to other management approaches, will be useful in reducing the extent and severityof downy mildew on spinach crops and diminishing pathogen spread through seed.

A diversidade dos fungos no planeta Terra é imensa, com as extrapolações dos dados de trabalhos já concluídos estima-se cerca de 1,5milhão de espécies relatadas, onde apenas 74 mil espécies foram realmente descritas. No Brasil e no Rio Grande do Sul, vários trabalhosvêm contribuindo com o conhecimento sobre os fungos macroscópicos da Mata Atlântica. Porém, ainda não existem trabalhos sobre adiversidade micológica para o Parque das Sequóias, no qual este trabalho relata a ocorrência de famílias de fungos para esta região. Oparque possui uma área de 10 hectares e está localizado no bioma Mata Atlântica, este bioma guarda a maior micodiversidade por hectareentre as Florestas Tropicais. O objetivo deste trabalho foi realizar o levantamento das espécies de fungos na região do Parque das Sequóias,localizado na cidade de Canela/RS, visando contribuir com o conhecimento da micobiota local. A coleta do material foi realizada emsetembro/2013, seguindo a metodologia proposta em literatura especializada. Após a coleta, os basidiomas foram levados para oLaboratório de Botânica da Universidade Federal do Pampa, onde foram analizados. As análises das características macroscópicas forambaseadas nos dados propostos na literatura especializada, feitas a olho nu e para maiores detalhes com o auxílio de um microscópioestereoscópico. Para análise de características microscópicas o material foi reidratado na solução 5% KOH e submetido a reagentes, taiscomo o vermelho congo, azul de cresil e reagente de Melzer. This research indicated various fungal infections in different parts of several medicinal plants that grown in arid woodlands of Vayots Dzorregion of Armenia. In this territory we investigate 289 species of medicinal plants belonging to 175 genera and 58 families. 39 species ofmicroscopic fungi were found from 41 species of medicinal plants from this area. Among the taxa of fungi 18 species belong to Ascomycota,7 belong to Basidiomycota and 1 belongs to Oomycota divisions. Golovinomyces genus from powdery fungi was the dominant for 11 higherplants, the most common species of micromycetes in this territory is G. galeopsis (about 15.4% of the total), which causes of medicinalplants from the family Lamiaceae.

Sensitivity of Mycosphaerella pinodes to pyraclostrobin fungicide

Mycosphaerella blight, caused by Mycosphaerella pinodes, is a destructive disease of field pea that is managed using foliar fungicides.Strobilurin fungicides have been used in western Canada for disease management since 2003. To assess the baseline sensitivities of M.pinodes isolates to the strobilurin fungicide pyraclostrobin, the effective concentration to reduce mycelial growth by 50% (EC50) wasdetermined for 70 isolates collected prior to 2003 from Alberta, Saskatchewan, North Dakota, and Washington State. Each of these baselineisolates was sensitive to pyraclostrobin, with EC50 values ranging from 0.03 to 0.29 mg L-1. The pyraclostrobin concentrations required toreduce conidia germination by 50% was lower, ranging from 0.008 to 0.041 mg L-1. Three hundred and twenty-four isolates collected in2010 and 2011 were tested for high levels of insensitivity by examining mycelial growth using a discriminatory dose of 5 mg L-1. Nineteenisolates, were highly insensitive to pyraclostrobin, with EC50 values ranging from 80 to 216 mg L-1. Conidia of these isolates germinatedwhen exposed to a discriminatory dose of 0.1 mg L-1. Insensitive isolates infected field pea plants treated with pyraclostrobin, but sensitivieisolates did not. The identification of insensitive isolates indicates that insensitivity may be emerging in the pathogen population.

Abstract: The basidiomycetous genus Kriegeria Bres. 1891 (Kriegeriaceae), typified by K. eriophori, is legitimate. An ascomycetous genus“Kriegeria” was originally published by Rabenhorst in 1876 as an invalid provisional name based on Ombrophila kriegeriana. Subsequently,the ascomycetous genus was validly published as Kriegeria Höhn. 1914 (Rutstroemiaceae), typified by Peziza elatina; this genus is anillegitimate later homonym, currently treated as a synonym of Rutstroemia. Additionally, Rabenhorst's invalid name was “revived”(validated) as Kriegeria Rabenh. ex Seaver 1943, typified by O. kriegeriana, to accommodate species currently placed in Chloroscypha.Revised nomenclators are presented for both of the genera, as well as for Ombrophila kriegeriana.

Out of 15 isolates, two white-rot basidiomycetes, namely MLK-01 and MLK-07, showed good growth in the pH range of 5.0 to 10.0. Bothisolates produced maximal fungal growth and xylanase activity with minimal cellulase contamination at incubation periods of 8 and 9 days,respectively, and pH 10.0 with wheat bran as substrate under submerged fermentation conditions. The maximum xylanase, CMCase andlignin peroxidase activitiesof isolate MLK-01 were 30.32, 0.32 and 0.25 IU/mL, respectively, with protein concentration of 1.76 mg/mL at pH7.5 and temperature of 75 °C, while those of isolate MLK-07 were 36.37, 0.25 and 0.15 IU/mL with protein concentration of 1.69 at pH 8.0and temperature of 65°C. Hg2+ and Cu2+ inhibited the xylanase activity for isolate MLK-01 to 17.6 and 20.20%, and for MLK-07 to 82.2and 70.5%, respectively. All the metallic ions like Ca2+, Mn2+, Fe2+, Pb2+, Mg2+, Ni2+, K+, Na+ and Zn2+ improved the enzymeactivity, but the enzyme activity doubled with Zn2+for isolate MLK-01and with Fe2+ for isolate MLK-07.

DNA sequence analysis has shown Candida abiesophila (NRRL Y-11514T, CBS 5366T) and Candida asiatica (NRRL Y-63747T, CBS 10863T)to be members of a small clade that is phylogenetically separate from other yeasts. In view of their isolation from neighboring genera, suchas Pichia and Saturnispora, the two anamorphic species are proposed for transfer to Martiniozyma gen. nov. (MycoBank MB 812061) withMartiniozyma abiesophila designated as type species (MycoBank MB 812062). In keeping with the International Code of Nomenclature foralgae, fungi, and plants, which specifies that related anamorphic and teleomorphic species can be assigned to the same genus, the followingCandida species are transferred to Saturnispora to conform with their phylogenetic placement: Candida diversa (NRRL Y-5713T), Candidahalmiae (CBS 11009T), Candida sanitii (CBS 10864T), Candida sekii (CBS 10931T), Candida siamensis (CBS 11022T), Candida silvae (NRRLY-6725T) and Candida suwanaritii (CBS 11021T).

El centro de origen y domesticación de Jatropha curcas L. es México. Este arbusto se distribuye en varios ecosistemas tropicales y tiene granpotencial para la producción de biodiesel. Sin embargo, este cultivo puede ser afectado por la presencia de diversos hongos fitopatógenosque causan diferentes enfermedades y disminuyen la calidad de sus semillas. Además, algunas plagas insectiles causan daños en J. curcas.El objetivo de este estudio fue evaluar la actividad antagonista y entomopatógena de bacterias rizosféricas contra Fusarium verticillioides yLeptoglossus zonatus. La actividad antagonista de Bacillus subtilis, Bacillus mojavensis, Bacillus thuringiensis y Lysinibacillus sphaericus fueevaluada contra Fusarium verticillioides por la técnica de cultivo dual en medio papa dextrosa agar. Las variables estudiadas fueron elcrecimiento micelial y la morfología hifal. Se realizaron diferentes cinéticas de crecimiento con las cepas bacterianas y la observación decristales proteicos de B. thuringiensis y L. sphaericus. Se estableció la cría de L. zonatus en condiciones de laboratorio. Se evaluó el efectode B. thuringiensis y L. sphaericus en la mortalidad y desarrollo de L. zonatus. Los resultados obtenidos demostraron que todas las cepas debacterias rizosféricas inhibieron el crecimiento micelial y afectaron la morfología hifal de F. verticillioides. Las cepas de bacterias rizosféricasmostraron actividad antagonista contra F. verticillioides con independencia del medio de cultivo utilizado y de las fases de crecimientoprobadas. En estas condiciones de estudio, B. thuringiensis y L. sphaericus no afectaron la mortalidad ni el desarrollo de L. zonatus. Engeneral, las cepas de bacterias rizosféricas evidenciaron actividad antagonista contra F. verticillioides y no mostraron actividadentomopatógena contra L. zonatus

Analysis of fungal endophytes associated with rice roots from irrigated and uplandecosystems in Kenya

Background and aim Fungal endophytes are commonly associated with plants, and are considered an important component of cropproduction. They can influence plant growth and tolerance to biotic and abiotic stresses. The aim of this study was to analyse and identifyendophytic fungi associated with rice roots in irrigated and upland ecosystems in Kenya, as an inventory for a future search for biologicalcontrol and growth promoting agents. Methods Fungi were isolated from the roots and selected based on culture characteristics. All selectedisolates were sequenced using primers targeting the internal transcribed spacer (ITS) region, intergenic spacer (IGS) region and the geneencoding the translation elongation factor (TEF-1α). The species were determined by comparing their sequences with those of wellcharacterised or type strains. Phylogenetic relationships among the species were used to identify their taxonomic groups, and distribution inthe agroecosystems, especially for the Fusarium spp. Results Based on sequencing of the ITS region, 75 fungal isolates were identified asFusarium-like, while the remaining 98 isolates were found to belong to different species representing other genera than Fusarium. A furtheranalysis of the Fusarium spp., using concatenated IGS and TEF-1α sequences showed that these isolates belong to the Fusarium oxysporum(FOSC) and Gibberella fujikuroi (GFSC) species complexes. Within the FOSC isolates, a clear divergence was observed between isolates fromirrigated and upland ecosystems, while in the GFSC this phenomenon was not observed. When the total number of species was considered,27 species were identified in the irrigated ecosystems, while only 18 species were found in the upland ecosystems.
