MITOMYCIN-TREATED UNDIFFERENTIATED EMBRYONIC STEM CELLS AS
A SAFE AND EFFECTIVE THERAPEUTIC STRATEGY IN A MOUSE MODEL OF
PARKINSON’S DISEASE

Parkinson’s disease (PD) is an incurable progressive neurodegenerative disorder. Clinical
presentation of PD stems largely from the loss of dopaminergic neurons in the nigrostriatal
dopaminergic pathway, motivating experimental strategies of replacement based on cell
therapy. Transplantation of dopaminergic neurons derived from embryonic stem cells
significantly improves motor functions in rodent and non-human primate models of PD.
However, protocols to generate dopaminergic neurons from embryonic stem cells generally
meet with low efficacy and high risk of teratoma formation upon transplantation. To address
these issues, we have pre-treated undifferentiated mouse embryonic stem cells (mESCs) with
the DNA alkylating agent mitomycin C (MMC) before transplantation. MMC treatment of
cultures prevented tumorigenesis in a 12 week follow-up after mESCs were injected in nude
mice. In 6-OH-dopamine-lesioned mice, intrastriatal injection of MMC-treated mESCs
markedly improved motor function without tumor formation for as long as 15 months.
Furthermore, we show that halting mitotic activity of undifferentiated mESCs induces a four-
fold increase in dopamine release following in vitro differentiation. Our findings indicate that
treating mESCs with MMC prior to intrastriatal transplant is an effective to strategy that
could be further investigated as a novel alternative for treatment of PD.

DEVELOPMENTAL EXPRESSION OF APP SECRETASES IN THE VISUAL
LAYERS OF RAT SUPERIOR COLLICULUS

Neural development refers to the processes that generate, plasticity of the nervous system.
Sensory systems formation is essential for normal brain development and function. Amyloid
precursor protein (APP) plays a central role in neuropathology of Alzheimer’s disease (AD)
but is also important to physiological processes such as synapse formation and neural
plasticity. Our group investigates the relationship between the APP and the plasticity of
visual connection during early postnatal development. Post-translational modifications of
APP includes proteolytic processing by enzymes known as α-, β- and γ- secretases to
generate peptide fragments. In non-amyloidogenic pathway, cleavage by α-secretase releases
soluble neurotrophic α-APP (sAPPα). In amyloidogenic pathway, sequential cleavage by β-
secretase and γ-secretase generates β-amyloid peptide (Aβ), which can aggregate in
neurotoxic oligomers. We have previously reported that the expression of APP and its
fragments in the visual layers of the SC is chronologically correlated with the critical period
of the retinocollicular pathway development, suggesting a functional role for this protein in
the multiple events that occur within this time window. In this pathway, activity-dependent
synaptic plasticity results in axonal/synaptic remodeling and topographic organization of
afferent terminals in the visual layers of the superior colliculus (SC). In this work, we studied
the ontogeny of the enzymes involved in APP proteolysis, ADAM10 (a constitutive alpha-
secretase), BACE (for β-site APP-cleaving enzyme) and presenilin-1 (PS1), a critical
component of theγ-secretase complex, in the visual layers of the SC. Lister hooded rats at
different ages ranging from PND7 to PND70 were used for biochemical experiments.
Animals were euthanized and superficial layers of the SC were extracted and processed for
western blot analysis using specific antibodies to the proteins of interest. All experimental
procedures were approved and conducted in agreement with the Animal Research Ethic
Committee (Protocol CEPA/No. 00205/10). Our results show that similar to APP and its
fragments both BACE and PS1 levels seem to be developmentally regulated, with relative
peaks at PND10 and gradually decline after the end of the critical period. On the other hand,
ADAM10 levels remain constant during the period analyzed. Our findings suggest that the
metabolism of APP may be involved in the multiple steps of retinocollicular pathway
development.

Role of EGFR in ERK, CREB and calcium signaling in P2Y-induced proliferation of
retinal cells in culture

Introduction: ATP is involved in the proliferation of neural precursors in the developing
chick retina. It can induce Ca2+ mobilization in this tissue, a response that declines in parallel
with the decrease in mitotic activity during retinal development. Since ATP-dependent cell
proliferation can rely on trophic factors such as EGF, we investigated the influence of EGF
receptors (EGFR) on P2Y-dependent cell proliferation and calcium mobilization in cultured
retinal cells. In addition, the effect of EGFR activation and blockade on ERK and CREB
signaling pathways was investigated.Methods: Retinal cells obtained from retinas of 7-day-
old chick embryos were cultured for 2 or 3 days. Cell proliferation was estimated by the
incorporation of [3H]-thymidine. Phosphorylation of ERK and CREB was determined by
western blot using specific antisera. Calcium imaging was carried out on confocal
microscopy using the Fluo-3 AM calcium indicator.Results: Cultures treated for 24 h with
250 µM ADP increased [3H]-thymidine incorporation (213 ± 15 %compared to control, n=9),
an effect that was prevented by 5 µM AG1478, an EGF receptor inhibitor (91 ± 17%
compared to control compared to ADP, n=5). Levels of p-ERK and p-CREB were also
increased by treating the cells with 250 µM ADP (respectively 425 ± 71 %, and 152 ±
1%,compared to control levels, n=3) a response that was decreased by the incubation of the
cultures with 10 µM AG1478 for 3 h (ERK: 195 ± 28 % and CREB: 80 ± 4 % compared to
control and ADP n=3). In contrast to ADP, 100 ng/ml EGF did not induce cell proliferation
(127 ±10%, n=6), neither ERK (73 ± 9%, n=3) nor CREB (127 ± 7, n=4) phosphorylation.
Moreover, activation of nucleotide receptors induced transient increases in intracellular
calcium concentration in the retina cell cultures. Peak values of the increases represented 208
± 8% of the resting levels when cells were treated with 250 µM ADP. The ADP-induced
calcium responses were attenuated when cultures were treated with 5 µM AG1478 for 24 h
(143 ± 5% of the resting level,compared to ADP, n=59 cells).Conclusion: These results
suggest that ADP-induced proliferation as well as ERK and CREB phosphorylation in retinal
cultures are dependent on the activation of EGF receptors. Furthermore, since AG1478
attenuated ADP-dependent calcium responses, our results raise the possibility that EGF
regulates nucleotide responses by affecting the expression of P2Y receptors in the retinal
cultures.

EFEITO DA INTERLEUCINA-2 NO SISTEMA COLINÉRGICO EM CULTURAS
DE CÉLULAS DA RETINA DE RATOS

O sistema colinérgico se caracteriza pelo neurotransmissor acetilcolina, de seus receptores
(muscarínicos e nicotínicos), dos transportadores CHT1 e VAChT, enzimas de síntese ChAT
e degradação. Dados prévios do nosso laboratório demonstram que o tratamento de células da
retina de animais neonatos com IL-2 por 48h, regula a captação de [H3]Colina de maneira
dose-dependente, sugerindo um efeito modulatório desta citocina sobre o no fenótipo
colinérgico.
O objetivo deste trabalho foi investigar o efeito da IL-2 na modulação dos marcadores
colinérgicos.
Ratos neonatos (P0-P2), da linhagem Lister Hooded, foram sacrificados e suas retinas
dissecadas em solução salina sem cálcio e magnésio (CMF). Posteriormente, as células
passaram por processo de dissociação química em tripsina 0,1%. Depois de dissociadas, essas
células foram plaqueadas na densidade de 105 cel/cm2 em placas de Petri de duas formas:
culturas controle (1 mL de meio 199 com células, acrescido de 1 mL de meio 199) e culturas
tratadas (1 mL de meio 199 com células, acrescido de meio199 contendo IL-2 50U/mL). As
culturas foram incubadas em atmosfera controlada com 5% de CO2 e 95% de ar, mantidas a
37°C por 48 horas. Posteriormente as células foram lisadas e a quantificação proteica foi
realizada pelo método de Bradford. Os níveis de receptores muscarínicos (M1, M2, M3, M4 e
M5), do transportador vesicular de acetilcolina (VAChT) e do receptor nicotínico alfa 7
foram analisados por Western Blot. Os experimentos foram realizados em triplicata,
considerando diferença significativa com valor de p<0,05. Os procedimentos experimentais
com animais foram aprovados pelo Comitê de Ética no Uso de Animais (CEUA) da UFF
(projeto 00124/09).
Nossos resultados demonstram que o tratamento com IL-2, por 48h, induz uma diminuição
significativa nos níveis dos receptores M1 (CT=100%; IL-2=33,36% EPM=4,73%), M2
(CT=100%; IL-2=34,88% EPM=12,13%) e M4 (CT=100%; IL-2=44,48% EPM=6,73%) e
um aumento nos níveis do receptor M5 (CT=100%; IL-2=125% EPM=2,8%) e do VAChT
(CT100%; IL-2=114% n=2). Entretanto, não interferiu nos níveis do receptor M3, e do
receptor nicotínico alfa 7.
Podemos concluir que a IL-2 é capaz de regular o fenótipo colinérgico através da regulação
dos níveis dos receptores muscarínicos e do VAChT em culturas de células da retina de ratos
neonatos no período de 48h de tratamento, com possíveis efeitos no processo de
diferenciação do tecido retiniano.

MOLECULAR CHARACTERIZATION OF NMDA RECEPTOR IN THE RAT LINE
SELECTED FOR HIGH AND LOW ANXIETY-LIKE RESPONSE

Introduction: Anxiety is the emotional dysfunction that afflicts more the quality of human
life. Therefore, the basic nature of research that seeks to understand the neural mechanisms
underlying these disorders are extremely important. Glutamate is the major excitatory
neurotransmitter in the mammalian brain and is important in neural development, plasticity
for learning and memory, and it is also implicated in pathological conditions such as anxiety.
Its actions are mediated by different receptors, including the ionotropic NMDA receptor
(NMDAr) consisting of different protein subunits. The combination of two subunits GluA1
and GluA2 compose two receptors present in greater numbers in the CNS. The aim of this
study was a comparative neurochemical analysis in animal model with a genetic basis of
anxiety. Methods: For this study, we used male Wistar rats previously selected through the
freezing paradigm and characterized as high (CHF) and low (CLF) anxiety-like response,
compared to control animals (CTL). Behavioral tests to evaluate the levels of anxiety, such as
elevated plus-maze (EPM) and light-dark box and memory tests, such as object recognition
task and Y-maze were also used. Moreover, we evaluated the expression and localization of
GluN1, GluN2A and GluN2b subunits of NMDAr. Results and Conclusion: CHF animals
were more anxious than control rats and CLF animals were less anxious than control rats in
terms of both the number of entries into EPM open arms and the percentage of time spent in
these arms. No statistical differences were found in locomotor activity, as measured by the
number of entries into the closed arms of the EPM. In light-dark box, no statistical
differences were found among the groups. In the object recognition task the results indicated
no difference between the three groups. In Y-maze only CHF animals avoided the new arm,
compared to animals CLF and CTL, with impaired memory in this test. Biochemical analyzes
showed a significant increase in the content of GluN1 subunit for CHF and CLF animals
compared to the control. The results also point to an increase in GluN2A subunit in CLF
animal compared to control and CHF animals and increased GluN2B subunit in CHF animals
for CTL and CLF animals. Taken together, these results support the hypothesis that the
differential composition of NMDA subunits participates in the regulation of anxiety-like
behavior in these animals.

CAFFEINE TREATMENT DURING LACTATION DISRUPTS RETINOTECTAL
TOPOGRAPHY IN PUPS: POSSIBLE INVOLVEMENT OF GLIAL CELLS

Retinotectal pathways have been used as a model to investigate the mechanisms governing
the development and plasticity in the central nervous system. Adenosine is an important
neuromodulator, which are present in neurons and glial cells. In previous studies, our group
demonstrated the presence of the A1 and A2A adenosine receptors in the superior colliculus
(SC) at different stages of development. Moreover, the treatment with caffeine, a non-
selective antagonist of these receptors, led to a disruption in the visual connection. This study
aims to investigate the indirect effect of caffeine during lactation on the distribution of
retinotectal axons of pups as well as possible modulations on the activation of glial cells in
the retina and SC. Methods: Lister Hooded rats were used. From the day of the birth of their
young, mothers were submitted to oral treatment with caffeine (1g/L) to postnatal day 13 or
21 (PND13 or PND21) of the pups. Control animals received water. We establish fluid intake
monitoring standards for infants and weight gain of the animals submitted to indirect
treatment with caffeine, to observe metabolic changes. After treatment, one group of animals
received intraocular injection of HRP, an anterograde tracer to map the retinotectal
connections or were processed for western blot analysis to compare the contents of GFAP, a
biomarker of activated glial cells. Results: Our results demonstrated that caffeine intake by
the mother during lactation leads to an increase in the retinotectal plasticity at PND13 (n=4).
This effect is characterized by the presence of many fibers in atypical regions of the visual
layer in the SC. Furthermore, there was a tendency to fall in pCREB content in the SC of the
pups in the PND13 (7565±51) compared to the control group (8243±62) (n=3/group). At
PND21, pCREB content decreased significantly (1494±90.41) (n=3/group) compared to the
control group (2650±102.6). At PND21 a marked decrease in glial reactivity (n=3) was
observed both in the retina and superior colliculus when compared to the control. These
effects were reversed by treatment withdrawal. Conclusions: Our results indicate that caffeine
treatment during lactation inhibits CREB phosphorylation and leads to a disruption of the
visual connectivity of the pups. Therefore, we hypothesized that blocking A2aR in glial cells
by caffeine decreases the cAMP-PKA-CREB pathway, weakening the synaptic contacts and
leading to a reorganization of retinotectal circuitry.

INTRAVITREAL TREATMENT WITH INTERLEUKIN-4 OR INTERLEUKIN-6
RECRUITS DIFFERENT SIGNALING MOLECULES TO MODULATE RETINAL
ACTIVITY

Objectives: Interleukin- 4 (IL-4) and interkeukin-6 (IL-6), are classically known as
antiinflammatory and pro-inflammatory cytokines, respectively. Some studies have
demonstrated effects of both cytokines in different aspects of CNS development. Accordingly
we have previously shown the effect of exogenous application of IL-4 or IL-6 upon
development of visual circuit, using the retinotectal pathway of rodent as a model. Our data
show that both molecules alter the distribution of retinal axons upon superior colliculus in
different stages of development. However the molecular mechanisms underlying these
responses are unknown. The aim of the present research is to investigate the signaling
pathway that is activated after intravitreal injection of IL4 or IL6 in the retina. Methods:
Lister Hooded rats were injected with either IL-6 or IL-4 in the right eye at PND10. Control
matched-group received PBS injection. At PND11 or PND14, animals were euthanasiated.
Retinas were prepared for western blot or immunohistochemistry procedures. We used glial
markers such as GFAP and Iba1 and intracellular proteins such as pSTAT3, pSTAT6, pERK
and pAkt. Nissl staining was also performed in order to evaluate retinal cytoarchitecture after
intraocular injection. Results: The thickness of retinal layers was similar between PBS- and
cytokines treated groups and no pyknotic nuclei or inflammatory profile cells were detected
with Nissl staining. Also, no difference was observed in Iba1 levels in all groups. Concerning
astrocyte marker, IL-4 treated animals presented a lower level of GFAP content compared to
control (PBS-treated rats). On the other hand, the level of GFAP was unchanged in IL-6
treated animal. The data from biochemical analysis revealed that IL-4 treated animals
presented a transient increase in pSTAT6 24h after injection and a more sustained increase in
pERK levels which was detected at PND11 through PND14. IL-6 treated animals presented
an increase in pSTAT3 levels. No difference was observed in pAKT content in any group.
Conclusion: Our data show that exogenous in vivo treatments with IL-4 or IL-6 modulate
different intracellular pathways in the retina, without inducing an inflammatory response.
Moreover these modifications seem to trigger a different pattern of retinal activity, which in
turns could be responsible for the reorganization of retinal afferent in the target.

ENDOCANNABINOIDS PROTECTION AGAINST ISCHEMIC DAMAGE TO THE
RETINA

Objective: the aim of this study is to investigate the functions of anandamide and 2-
arachidonoylglycerol in retinal ischemic events.
Methods: 2-7 days post-hatching (PH) chicks (Gallus domesticus) are killed (CEUA/UFF
no112/12) and, after enucleation, retinal segments are randomly addressed to be control
(incubated in a Ringer with glucose/aerated with 95%O2 e 5% CO2) or ischemic condition
(incubated in a Ringer without glucose/aerated with 95%N2 e 5%O2 – oxygen and glucose
deprivation, OGD) for 50 minutes. In some cases, antagonists were pre-incubated for 10
minutes. Then, retinal segments and their respective incubation solution were collected to
quantify the activity of the enzyme lactate dehydrogenase (LDH) with a colorimetric
commercial kit (Promega), for the cell viability assay. The percentage of LDH release was
used as a marker of cell viability. We analyzed the data by Kruskal-Wallis test with post-test
SNK through the program BioEstat (versão 5.3, 2007).
Results: We observed that retinal treatments, with different concentrations of the inhibitors of
degradation enzymes of endocannabinoids, URB597 (CTR=100% n=5; OGD=195,2±31,61%
n=5; OGD+URB597 0,1µM=173,1±35,49% n=5; OGD+URB597 1µM=214,7±54,60% n=3;
OGD+URB597 10µM=245,5±47,00% n=5), and URB602 (CTR=100% n=3;
OGD=244,6±17,99% n=3; OGD+URB602 0,1µM=323,8±24,98% n=3; OGD+URB602

reducing the LDH release induced by OGD. However, the co-exposure to inhibitors of

Conclusion: The data suggest that endocannabinoids can have a protective role in this model
of retinal ischemia. Additionally, the protective effect is dependent on the inactivation of at
least one of the CB receptors.

A2A ADENOSINE RECEPTOR ACTIVATION MODULATES SRC KINASE
ACTIVITY IN cAMP-DEPENDENT MANNER

Src kinase is a member of the family of tyrosine kinases involved in many developmental
processes such as cell proliferation, differentiation and survival. The phosphorylation sites
which regulate Src activity include tyrosine 416 (Tyr416), which results in Src activation by
autophosphorylation, and tyrosine 527 (Tyr527) which promotes Src inhibition mediated by
C-terminal Src kinase (Csk) phosphorylation. Thus, Src activity is regulated by
phosphorylation levels at Tyr527 and Tyr416 residues.
Adenosine, a neuromodulator in the CNS, activates different metabotropic receptors,
including the A2a receptor which is coupled to Gs protein and cyclic AMP production. Our
previous results showed that A2a receptor activation decreases Src phosphorylation at
Tyr416. Thus, our objective here was to evaluate the signaling pathways involved in the
regulation of Src activity by A2a receptor activation in retinal cultures.
Retinal cultures were stimulated with CGS21680 (A2a agonist) for 5 minutes and p-Src
levels were evaluated by Western Blot. We observed a decrease in p-Src (Tyr416) induced by
A2a receptor stimulation that was blocked by ZM241385 (antagonist A2a) and SQ22536
(adenylyl cyclase inhibitor). In addition, A2a receptor activation promoted an increase in p-
Src (Tyr 527) and p-Csk induced by A2a receptor stimulation. Similar results were observed
in retinas from 9-day-old chick embryos where CGS21680 promoted a decrease of p-Src
(Tyr416) and an increase of p-Csk.
These results demonstrate that stimulation of A2a receptors induces a decrease of p-Src and
this effect involves the cAMP pathway. Our working hypothesis is that activation of PKA by
A2a receptors leads to Csk activation and Tyr527 phosphorylation, with a consequent

A mente humana sempre foi um dos fascínios ao longo da história, cientistas e filósofos
sempre estiveram em busca por desvelar a mente humana e a relação da mesma com o
funcionamento cerebral. Muitos dos conceitos utilizados atualmente na neurociência moderna
têm suas origens nos antigos médicos e filósofos gregos. Encontramos registros de pesquisas
desde a antiguidade sobre os sintomas de danos no cérebro, porém à época, o coração que era
considerado a sede da alma e o repositório das memórias. Com o passar dos anos houve a
necessidade de dissociar o estudo anatômico da abordagem funcional do sistema nervoso, e
deu-se a criação do termo Neurociência, que engloba o estudo da estrutura e de todas as
funções normais ou patológicas, do sistema nervoso. Os avanços que ocorrem nos dias de
hoje permitem que compreendamos cada vez mais as circuitarias neurais associadas às
diferentes funções mentais, porém questionamentos fundamentais da natureza da mente
humana permanecem em aberto, como por exemplo qual a origem da mente humana, a real
natureza dos processos mentais, são algumas das questões que permanecem sem resposta.

OMEGA-3/DHA NUTRITIONAL RESTRICTION DELAYS DEVELOPMENT OF

In the present work, we studied the impact of a dietary restriction of omega-3 fatty acids on
the development of eye specific segregation and topographical specificity of retinofugal
pathways. We also studied the impact of low omega-3 diets on the differentiation of retinal
layers and cholinergic markers in the retina. Female Lister Hooded rats and their litters were
fed with either control (soy oil) or restricted (coconut oil) omega-3 diets. At various postnatal
ages, rat pups received eye injections of neuronal tracers to visualize retinal axons at their
brain targets. Fatty acids profile analysis indicated that the experimental diet led to a selective
reduction in DHA content in the visual system. Omega-3 restriction induced an increase in
the density of retinal axons in the superficial layers of the SC. This effect was observed
throughout the stratum griseum superficiale (SGS), including the ventral and intermediate
SGS layers at post-natal day (PND) 13, PND28 and PND42. The same pattern of expanded
terminal fields was observed in the retinogeniculate pathways. The supplementation with fish
oil (omega-3 fatty acids) for two weeks was able to reverse the abnormal expansion of the
retinocollicular projection. We also studied possible mechanisms involved in the loss of
synaptic stabilization. Restricted omega-3 groups showed, in the visual layers of the SC
decreased levels of GAP-43 phosphorylated form (pGAP-43) consistent with a reduction in
synaptic stabilization. Omega-3 restriction increased the content of AMPAR subunits (GluR1
and GluR2) and decreased NMDAR (NR1, NR2A and NR2B) subunits. Omega-3 restricted
groups showed a decrease in RXRα content and CREB phosphorylation (pCREB) in the
visual layers of the superior colliculus. Retinal differentiation was also affected by omega-
3/DHA deprivation. We describe a reduction in rhodopsin immunolabeling and also a
reduced labeling of cholinergic markers such as VAchT in the inner nuclear layer and β2-
nicotinic receptors in the ganglion cell layer and inner nuclear layer. The data indicate,
therefore, that the chronic dietary restriction of omega-3 fatty acids delays axonal
elimination, interfering with the maintenance of synaptic connections in the visual system.
Also, the results show that the development of the retina is also delayed, specially the
development of retinal cholinergic transmission.

TRATAMENTO COM IL-1 BETA É CAPAZ DE MODULAR OS NÍVEIS DE TNF-
ALFA EM CULTURAS MISTAS DE CÉLULAS DA RETINA

Sabendo do envolvimento desta citocina na sobrevida de CGR, resolvemos analisar se a
própria IL-1 beta (5,0ng/mL) seria capaz de modular os níveis de TNF-alfa em culturas
mistas de células da retina por diferentes intervalos de tempo. Os procedimentos
experimentais com animais foram aprovados pelo Comitê de Ética no Uso de Animais
(CEUA) da UFF (projeto 00124/09). Ratos neonatos (P0-P2) da linhagem Lister Hooded
foram sacrificados e suas retinas dissecadas em solução salina sem cálcio e magnésio (CMF).
Em seguida, as retinas foram incubadas com 0,1% de tripsina. Posteriormente ocorreu
dissociação mecânica com pipeta Pasteur de ponta afilada. As células foram plaqueadas na
densidade de 100000 cels/cm2 em placas de Petri. As culturas foram mantidas em meio
completo (meio 199, 5% de soro fetal bovino, glutamina e antibióticos) na presença ou não
de IL-1 beta. As culturas foram incubadas em atmosfera controlada com 5% de CO2 e 95%
de ar, mantidos a 37ºC por 15min, 24 e 48h. Todos os experimentos foram realizados em
triplicata. Os níveis de TNF-alfa foram determinados pela análise de Western Blot. Ao
tratamos nossas culturas com IL-1 beta a 5,0ng/mL, observamos uma modulação dos níveis
de TNF-alfa em relação ao controle logo no início do tratamento, em 15 minutos (CT100%
IL-1 beta 25,7±4,525). No período de 24 horas de tratamento com IL-1 beta, observamos uma
discreta diminuição nos níveis do TNF-alfa (CT 100% IL-1 beta 14,59±3,167). Ao
investigarmos os níveis de TNF-alfa em nossas culturas no período de 48 horas, observamos
que o tratamento com IL-1 beta é capaz de aumentar os níveis desta citocina (CT 100% IL-1
beta 34,80±9.970). Nossos dados sugerem que haja uma importante modulação entre as
citocinas TNF-alfa e IL-1 beta em células da retina de ratos, à semelhança do que acontece
em outros sistemas. Baseados nesses resultados podemos sugerir que esta modulação possa
contribuir para o efeito trófico da IL-1 beta na população de CGR.

Fator ativador de plaquetas resgata a pluripotência de células-tronco embrionárias
murinas diferenciadas espontâneamente

As células tronco embrionárias murinas (mES), da linhagem USP-1, são células pluripotentes
e por isso apresentam um potencial ilimitado de auto renovação além da habilidade de se
diferenciar em tipos celulares dos três folhetos embrionários. A capacidade de auto renovação
das mES é mantida in vitro pela ação do fator de inibição de leucemia (LIF) conhecido por
ser secretado por uma série de tipos de células. Dados da literatura demonstram que o
fosfolipídio bioativo Fator de Ativação de Plaquetas (PAF) participa da fisiopatologia de
diferentes processos patológicos e regula várias atividades biológicas, incluindo a
proliferação e diferenciação celular.
Alguns resultados prévios do grupo mostraram também que o PAF estaria resgatando a
expressão de marcadores de pluripotência e diminuindo a expressão de marcadores de
diferenciação após a diferenciação espontânea destas células. Para verificar tal fenômeno,
fizemos um tratamento por 24 horas com PAF após 7 dias de diferenciação espontânea sem
adição de LIF. Analisamos a expressão, através de imunocitoquímica, de marcadores de
pluripotência, como Oct-4 e SSEA-1 e marcadores de diferenciação, como o PAX-6 e
Nestina e a atividade da fosfatase alcalina. Com o objetivo de caracterizar e analisar o efeito
do PAF sobre a pluripotencialidade das células-tronco embrionárias murinas após sua
diferenciação espontânea e visando o entendimento dos mecanismos celulares e moleculares
da manutenção e o possível resgate, através do tratamento com PAF, do estado pluripotente
destas células-tronco. Os resultados demonstraram que quando tratadas com PAF durante
24h, após a diferenciação espontânea, a expressão de marcadores de diferenciação (Pax-6 e
Nestina) diminuem. Em contrapartida, os marcadores de pluripotência (Oct-4, SSEA-1 e
atividade da fosfatase alcalina) aumenta em relação ao controle não tratado com PAF e
diferenciado espontaneamente.

ATP AND GLUTAMATE: MODULATION OF GLIOTRANSMITTERS RELEASE
IN CHICK MÜLLER CELLS IN CULTURE

Objectives: Herein, we investigated the effect of purinergic receptors on glutamate release
and of glutamate receptors on ATP release in addition to their effects on calcium mobilization
in cultured retinal cells.
Methods: Müller glia cultures were obtained from 8-day-old chick embryos and cultivated for
2-3 weeks. To measure glutamate release, cultures were incubated with [3H]-glutamate.
Then, cells were incubated in Hanks´ solution for 15 minutes in the presence or absence of
nucleotides. The supernatant was collected and the radioactivity measured by scintillation
spectroscopy. Exocytosis of ATP-filled vesicles was estimated by examining quinacrine
staining before and after treatments. The release of ATP by the cells was evaluated by
luminescence, using an ATP determination kit of Invitrogen. Calcium imaging was carried
out on confocal microscopy using the Fluo-3 AM calcium indicator.
Results: The activation of purinergic receptors induced transient increases in intracellular
calcium concentration in cultured Mülller glia cells. Peak values of the increases represented
224 ± 8%***, 241 ± 15%*** and 213 ± 10%*** (n>12 cells; ***p<0,001) of the resting
levels when cells were treated with 1 mM ATP, 100 µM Bz-ATP and 100 µM ATP,
respectively. We analyzed labeled glutamate release in cultured glia using the same
purinergic agonists and found an increase in the extracellular content of the amino-acid after
15 min. stimulation (3 mM ATP = 145 ± 10%***, 100 µM Bz-ATP = 149 ± 13%** and 100
µM ATP = 139 ± 6%***; n>5, **p<0,01). On the other hand, 1 mM glutamate induced
calcium increases corresponding to 150 ± 5%*** (n=8) of the resting level. While no change
was detected in quinacrine stained, ATP-containing vesicles for at least 10 min in control
cultures, treatment of the cells with 1 mM glutamate resulted in reduced intracellular levels of
quinacrine fluorescence at the same time interval. Incubation of the cells for 5 min with 1mM
glutamate was also able to increase the levels of ATP in extracellular medium by 179 ± 15 %
(p<0.001, n=19), a response that was blocked by the calcium chelator BAPTA-AM.
Conclusion: These results suggest that cultured glial cells from chick embryo retina can
release glutamate following activation of the calcium-inducing purinergic receptors.
Furthermore, cultured Müller cells have ATP-containing vesicles that can be released in a
calcium-dependent way by activation of glutamate receptors.

MOUSE EXPERIMENTAL MODEL OF PARKINSON'S DISEASE INDUCED BY 6-
OHDA PRESENTS TEMPORAL MODULATION OF STRIATAL CHOLINERGIC
SYSTEM

Parkinson's disease (PD) is a neurodegenerative disease characterized by a severe and
progressive motor impairment due to progressive and specific degeneration of A9
dopaminergic neurons of the substantia nigra compacta. Acetylcholine (ACh) is present in the
striatum via cholinergic interneurons and several studies have shown that cholinergic activity
in the striatum regulates the release of dopamine (DA) through the action of nicotinic
receptors (nAChRs). The nicotinic ACh receptors are expressed in dopaminergic nerve
terminals and the normal activity of striatal neurons depends on the balance between DA and
ACh. Disruption of this balance may contribute to the development of PD. Our group has
shown that the lesion by 6-OHDA induces modulation of the mechanisms of ACh synthesis
and degradation, what suggests the existence of a biochemical machinery that may be
involved in the response to tissue damage within a time window.Therefore, we aimed to
investigate the biochemical changes in the cholinergic system during different survival
periods (1, 2 and 4 weeks after injection of 6-OHDA) in a mouse model of PD, in order to
study the possible striatal cholinergic modulation of these animals. Adult C57Bl6 mice were
anesthetized and submitted to unilateral injection of 6-OHDA (2 μl at a rate of 0.5 µl/5 min)
into the left medial forebrain bundle using stereotaxic procedures. The behavioral test was
performed 1-4 weeks later, when animals were injected intraperitonealy with apomorphine
(0,5mg/kg) and visualized through an open field. Then, the mice were euthanized and
striatum was dissectioned to prepare samples for western blot analysis (we used antibodies
against nicotinic ACh receptors α7, β2 and α6). We have observed a reduction in α6 subunit
content, which was expected since this subunit is exclusively localized in nigrostriatal
dopaminergic terminals. Conversely, α7 and β2 subunits content had a progressive increase
that was accompanied by improvements in motor behavior of the animal. Several studies
have shown that α7 subunit, in dopaminergic neurons is involved in neuroprotection
pathways and that β2 subunit is constitutively expressed. These data suggest that the
availability of ACh may be involved in the remodeling of nAChRs as a result of the
dopaminergic loss, what might contribute to partial improvement in motor behavior observed
in animals submitted to 6-OHDA lesion.

Interleucina 6 induz autofagia em Células da Retina in vitro

Avaliar o papel da autofagia no efeito trófico da IL-6 sobre as Células Ganglionares da
Retina. Utilizamos ratos neo-natos Lister Hooded. Na sobrevida, a peroxidase foi injetada nos
colículos superiores antes da cultura, que foi tratada com IL-6 (50ng/mL), mantida na estufa
por 48hr, fixada, revelada e as CGR contadas em microscópio de campo claro. Para as
amostras, células mistas da retina foram tratadas com IL-6 por 15, 45 min e 48 hr e utilizadas
no Westernblot para avaliação dos níveis dos componentes da via autofágica (Beclina 1 e
LC3) e também do modulador da via NF-kB. O marcador Laranja de Acridina (LA)
identificou organelas autofágicas por citoquímica. Na análise estatística, a intensidade das
bandas foi analisada pelo programa ImageJ e expressas em percentual do controle. Os
resultados foram expressos na forma de histograma e na apresentação das bandas reveladas.
Os experimentos foram representados graficamente utilizando o programa GraphPad Prism.
Os valores representam a média ± erro padrão, obtidos pelo teste-t Student. Foi significativo
quando p<0,05. Não há diferença nos níveis da Caspase 3 ativada entre as células da retina
tratadas com IL-6 e o CT tanto em 45 min (CT=100%; Casp3=114%±3,79 n=3), quanto em
48hr (CT=100%; Casp3=100%±9,849 n=3) excluindo um possível papel anti-apoptótico.
Inibindo a autofagia nas culturas mistas de células da retina, o efeito trófico da IL-6 sobre as
CGR desapareceu (CT4hr=100%; CT48hr=46,5±1,2; IL-6=100%±3,5;
Inib.Autofagia=43,6±3,2; IL-6+Inib.Autofagia=48,7±2,4 n=3) . O LA demonstrou de forma
qualitativa que a presença de organelas autofágicas é maior nas culturas tratadas com IL-6
comparadas ao CT. A IL-6 se mostrou capaz de alterar os níveis do NF-kB com um aumento
em 45 min (CT=100%; NF-kB= 127%±29,48 n=3) e uma redução em 48 hr (CT=100%; NF-
kB=56%±10,73 n=3). Ela provocou uma tendência de aumento, em 15 min, nos níveis de
Beclina 1 (CT=100%; BECN1=48,5±25,5 n=2). Além de aumentar os níveis de LC3 em 45
min (CT=100%; LC3=95%±9,018 n=3) e reduzi-los em 48 hr (CT=100%;LC3=36%±8,083
n=3). Baseado em nossos resultados observamos que o tratamento das células da retina com
IL-6 foi capaz de alterar os níveis da BECN1 e a LC3, e do fator de transcrição NF-kB. Além
disso, vimos que a inibição da via autofágica é capaz de abolir a sobrevida das células
estudadas. Portanto,os dados obtidos no nosso estudo, nos permitem sugerir que a autofagia
parece mediar o efeito trófico da IL-6 sobre as CGR.

IL-13 MODULA OS NÍVEIS DE BDNF E pTrkB EM CULTURA DE CÉLULAS
MISTAS DA RETINA DE RATO

A interleucina 13(IL-13) é uma citocina de 10 kDa, cujos efeitos são bem conhecidos no
sistema imune. Trabalhos anteriores de nosso laboratório mostraram que essa citocina, na
concentração de 5ng/mL, aumenta a sobrevida das células da retina de rato in vitro. A IL-13 é
conhecida por compartilhar funções semelhantes aos da IL-4, já que seus receptores formam
um heterodímero. Dados prévios de nosso grupo, mostram que a IL-4 (50U/mL) aumenta
significativamente os níveis intracelulares de BDNF e, que a neutralização extracelular do
BDNF inibe o efeito trófico da IL-4 na população de células ganglionares. No presente
trabalho, avaliamos o efeito da IL-13 sobre os níveis de BDNF e de seu receptor fosforilado
(p-TrkB) em cultura de células da retina de rato após diferentes períodos de tempo.
Ratos neonatos da linhagem Lister Hooded foram sacrificados e suas retinas dissecadas em
solução salina. Em seguida, foram incubadas em solução de 0.1% de tripsina e, após 20 min,
dissociadas com pipeta Pasteur. As células foram plaqueadas (105 cel/cm2 ) em placas de
petri tratadas com poli-L-ornitina (25μg/mL). As culturas foram mantidas em meio 199(CT)
ou na presença de meio contendo IL-13 (5 ng/ml) e incubadas em atmosfera controlada com
5% de CO2 e 95% de ar, a 37º C por diferentes intervalos de tempo. Os níveis de BDNF e
pTrkB foram determinados pela técnica de Western Blot. Os procedimentos experimentais
com animais foram aprovados pela Comissão de Ética no Uso de Animais da UFF(00294/12).
Nossos resultados demonstram que o tratamento com IL-13 por 45min não altera os níveis de
BDNF , porém induz um , os níveis intracelulares de BDNF não foram alterados
significativamente em relação ao controle (CT: 100% ; IL-13: 105,8 ± 6,9% n=3).
Entretanto, houve um aumento de aproximadamente 30% os níveis de pTrkB (CT: 100% ;
pTrkB: 129,3±5,1 % n=3). Em 24h os níveis de BDNF apresentaram um decréscimo de
aproximadamente 20% e os níveis de pTrkB permaneceram aumentados (CT: 100% ;
pTrkB: 121,87±9,65 % n=3). Em 48 horas, observamos um aumento dos níveis
intracelulares de BDNF (CT: 100% ; BDNF: 152,5±7,78% n=3) e uma diminuição nos
níveis de pTrkB (CT: 100% ; pTrkB: 88,58±1,72 % n=3).
Baseados nesses resultados, sugerimos que o tratamento com IL-13 na concentração de 5
ng/ml induz uma modulação tanto dos níveis de BDNF intracelular como do pTrkB,
possivelmente pela liberação de BDNF para o meio extracelular o que levaria a fosforilação
de seu receptor.

IL-13 MODULA O EFEITO PROLIFERATIVO DO EGF EM CÉLULAS DA
RETINA DE RATOS: ENVOLVIMENTO DA SRC E DO NFkB

O Fator de Crescimento Epidermal (EGF) pertencente a família do EGF. A interleucina 13
(IL-13) pleotropica e seus efeitos bem conhecidos no sistema imune. Trabalhos do nosso
laboratório mostraram que essas citocinas isoladamente aumentam a proliferação das células
da retina de rato in vitro, porém o tratamento concomitante de EGF e IL-13 induz uma
diminuição do efeito proliferativo do EGF. Objetivos: No presente trabalho avaliamos o
envolvimento da via da Src e do fator de transcrição NFkB no efeito da IL-13 sobre a
proliferação induzida pelo EGF. Métodos: Ratos neonatos da linhagem Lister Hooded foram
sacrificados e suas retinas dissecadas em solução salina. Em seguida, foram incubadas em
solução de tripsina e após triturada com pipeta Pasteur. As células foram plaqueadas na
densidade de 105 cel/cm2 em placas de Petri. As culturas foram mantidas em meio 199 (CT)
ou na presença de meio contendo IL-13 e ou EGF e incubadas em atmosfera controlada com
5% de CO2 e 95% de ar, mantidos a 37ºC. Os níveis de p-Src e p-NFkB foram determinados
pela técnica de Western blot. Todos os procedimentos foram aprovados pela Comissão de
Ética no Uso de Animais da UFF (00294/12). Resultados: Dados prévios já demonstraram
que o tratamento com IL-13 (1ng/mL) induz aumento de 30% na proliferação e que o
tratamento com EGF (0,1ng/mL) aumenta em até 100% a proliferação de células da retina em
48h. Quando tratamos as culturas com EGF, na presença de IL-13, houve uma diminuição no
efeito proliferativo do EGF em 48, 72 e 96h. Neste trabalho mostramos que o tratamento com
IL-13 (1ng/mL) induz aumento em 40% nos níveis do p-NFkB (CT 100% p-NFkB 140,4±6,8
n=3) em 48h em relação ao controle e na cultura tratada com EGF altera os níveis do NFkB
em 50% (CT 100% p-NFkB 166,5±12,2 n=3), porém quando tratamos as culturas com EGF,
na presença de IL-13, houve uma pequena alteração de 20% do p-NFkB (CT 100% p-NFkB
119,8±2,3 n=3). Avaliamos também o envolvimento da p-Src, quando tratamos as culturas
com IL-13 ocorreu um aumento nos níveis da p-Src em 40% (CT 100% p-Src 149,5±4,9 n=3)
e na cultura tratada com EGF não houve alteração nos níveis da p-Src (CT 100% p-Src
121,4±7,0 n=3), em contrapartida o tratamento em conjunto dessas duas citocinas aumentou
em 90% os níveis da p-Src (CT 100% p-Src 202,1±3,4 n=3). Conclusão: Nossos dados
sugerem que o tratamento concomitante com IL-13 e EGF induza uma diminuição da
proliferação celular possivelmente através da ativação da via da Src e da via do NFkB.

Nitric oxide (NO) regulates death and survival events in the CNS. Our group has shown that
NO is capable to activate AKT by Ser 473 phosphorylation in retinal nerve cells in vitro. We
have also demonstrated that protein synthesis inhibition increases intracellular free L-Arg and
can also promote NO synthesis via nNOS. Inhibition of protein synthesis has been described
as a classical paradigm to prevent developmentally-regulated neuronal loss since it abrogates
the expression of cell death-associated factors. Therefore, we try to investigate if protein
synthesis inhibition could be involved in NO/AKT activated cell protection.
Initially we sought to observe if classical protein synthesis inhibitors (CHX and anisomycin)
could increase intracellular NO levels and if said increase was due to increase of L-Arg
availability. Using Griess method, a clear raise in intracellular NO concentration was
observed in cultures treated with CHX or anisomicyn. Furthermore, this raise was completely
inhibited by NOS inhibitor L-NAME. In order to directly evaluate NO accumulation induced
by CHX we used DAF-FM-DA, a NO-sensitive fluorescent probe and the same effect was
observed.
In order to evaluate protein synthesis inhibition effects on cell survival we maintained our
cultures in serum-deprived medium containing or not CHX. Indeed, serum-deprivation
strongly promoted cell death, an effect completely abolished when cultures were also treated
with CHX. In addition, when these tratments were associated with nNOS or AKT inhibitors
(7-NI and trisciribine, respectively),CHX loses its neuroprotective effects. In a more
aggressive death approach, we induce neurotoxicity death using NMDA for 20 hours
(100µM). Once again, CHX treatment proved effective in preventing cell death, and this
protection was lost in the presence of 7-NI.
We hypothesized that protein synthesis inhibition might also increase intracellular L-Arg
availability to induce a NO-dependent AKT activation, leading to neuronal survival in retinal
cultures.

Human multiform glioblastomas (GBM) are very aggressive central nervous system tumors
and resistant to chemo/radiotherapies, leading patients do decease in 12 months. It has been
demonstrated that inhibition of xC- system, a glutamate (relase)/cystine (uptake) exchanger,
represents the rate-limiting step of glutathione (GSH) synthesis, an essential component for
protection against oxidative stress. The xC- system is composed by a catalytic (xCT) and
regulatory (4F2hc) subunits and it’s described that sulphasalazine (SAS) is a potent inhibitor
of the system, diminishing GSH synthesis. Valproic acid (VPA) presents a histone
deacetylase family (HDAC) inhibitory activity and the effect of enhancing reactive oxygen
species (ROS). These effects have been associated to death induction on glioma cells. The
aim of this work is to verify how SAS and VPA act solely or in a combined way related to
xC- system and the generation of oxidative stress in GBM cells in culture. Methods: U87MG
human glioblastoma cell lines, GBM02 and GBM95 human glioblastoma cells (CONEP nº
2340), rat cortex, rat astrocyte and human astrocyte cells were harvested for 24h and treated
with SAS and/or VPA for 48h with various concentrations. After, cells were submitted to cell
count; immunocytochemistry for KI67 proliferation marker; ethidium homodimer (EtD-1)
death marker incorporation; western blot assay for xCT protein; L[³H]glutamate uptake and
relative intracellular ROS determination by fluorescence microscopy. We were able to
observe the treatment with SAS and/or VPA was able to diminish glioma cell number in vitro
and the combination of both drugs resulted in a most robust effect compared to drugs alone
after 48h treatment. The same effect was observed for EtD-1 incorporation and KI67
immunoreactivity in glioma cells, but rat cortex or astrocytes cultures, as well as human
astrocytes’, maintained the same parameters unaltered with combined or alone treatments.
We verified that treatment with SAS and SAS+VPA did not alter xCT protein levels after 48h
treatment, although a robust inhibition of total intracellular content GSH (next to 50%) and
xC- system uptake was observed on SAS or SAS+VPA treated U87MG and GBM02 cells.
We observed that SAS treatment is able to diminish intracellular antioxidant defenses while
and enhance ROS intracellular content, as well as VPA. Thus, combined treatment with SAS
and VPA get both effects together, leading glioblastoma to augment cell death.

Phenolic acids display several antioxidant properties and have been associated with
decreasing inflammation in different cellular
paradigms. Chlorogenic acid (CGA) is a phenolic acid that has been shown to regulate
activation of microglial cells at high doses.
Microglia represents a class of resident immune cells of the central nervous system which
release neuromodulators and can regulate
synaptic transmission. However, microglia may be involved in neuronal loss during
neuroinflammatory conditions mainly by the release of cytotoxic doses of glutamate. The
activity of cytoplasmic tyrosine kinases, such as those from the Src family, is enhanced
during neuroinflammation but the role of Src in microgliamediated glutamate extracellular
accumulation or microgliainduced neuronal damage is elusive. We used the microglial cell
line N9 and purified cultures of 8dayold chick embryo retinal neurons as experimental
models. To evaluate protein expression, we used the western blot technique and
immunocytochemistry with specific antibodies to measure protein expression and
morphological parameters. We used ethidium homodimer1 as a probe to evaluate cell death
on both cell culture models. Extracellular glutamate was measured in N9 cultures by the
formation of alphaketoglutarate and NADH in a reaction catalyzed by glutamate
dehydrogenase. We show that CGA is a potent inhibitor of basal or LPSevoked microglial
activity evaluating the CD11b expression by immunocytochemistry, CGA also decreased Src
function by activating its endogenous repressor Csk. Blocking Src expression with
lentiviruses containing specific shRNAs for this proteins decreased microglial extracellular
glutamate accumulation (73.6% ± 6.4, N=3). On the other hand, shCsk promoted and increase
of extracellular glutamate (149.6% ± 10.3, N=4). Furthermore, microgliadependent glutamate
extracellular accumulation activates NMDA receptors to trigger retinal neuronal cell death.
Microglia treatment with CGA prevented this glutamate excitotoxicity in retinal neurons. In
conclusion, we found that 1) increased Src activity in microglia promotes an accumulation of
extracellular glutamate and triggers retinal neuronal cell death, and 2)CGA is strongly
efficient in counteracting this microgliainduced excitotoxicity by inhibiting Src. The results
also support the hypothesis that Src inhibition by CGA is mediated through activation of the
PKA/Csk pathway.

INIBIÇÃO MEDIADA POR AMP CÍCLICO DO CRESCIMENTO DE CÉLULAS
GLIAIS APÓS LESÃO MECÂNICA DE CULTURAS DE RETINA DE EMBRIÃO DE
GALINHA

INTRODUÇÃO: O AMP cíclico (AMPc) é um sinalizador intracelular produzido a partir da
ativação da adenilil ciclase por receptores acoplados à proteína Gs. Neste trabalho
investigamos o papel deste nucleotídeo cíclico no crescimento de células gliais de retina em
culturas submetidas à lesão mecânica.
MÉTODOS:
Culturas de retinas de embrião de galinha com oito dias (E8) foram cultivadas por sete dias
(E8C7) e lesadas mecanicamente para gerar uma área livre de células. A área da lesão das
culturas foi fotografada sob iluminação de contraste de fase em microscópio Nikon Eclipse
(objetiva de 10x) e a área livre de células determinada pelo programa Image J.
RESULTADOS E DISCUSSÃO:
O tratamento prolongado das culturas lesadas com 0,5 mM de RO 20-1724, um inibidor da
fosfodiesterase dependente de AMPc foi capaz de inibir o crescimento das células gliais para
a área de lesão. Enquanto a área livre de células nas culturas controle diminuiu de 19,4 ± 0,9
em E8C7 para 4,7 ± 1,3 mm2 x 10-2 em E8C10, esta área diminuiu de 19,1 ± 1,8 para 14,8 ±
2,1 mm2 x 10-2 (N = 5) nas culturas tratadas com RO 20-1724. Já o tratamento das culturas
com 100 µM de IBMX, outro inibidor da fosfodiesterase, também foi capaz de inibir o
crescimento das células gliais para a área da lesão após 3 dias de tratamento (E8C10: controle
= 9,8 ± 0,9 mm2 x 10-2; IBMX = 16,2 ± 1,5 mm2 x 10-2; N = 3). Para confirmar um efeito
inibitório do AMPc sobre o crescimento de células gliais, culturas submetidas à lesão foram
tratadas com forskolina 10 μM, um potente ativador da adenilil ciclase. Este tratamento foi
capaz de inibir o crescimento das células após 3 dias de tratamento (E8C10: controle = 6,3 ±
0,8 mm2 x 10-2; forskolina = 16,4 ± 1,8 mm2 x 10-2, N = 4). Resultados semelhantes foram
obtidos após 3 dias de tratamento com 8-CPT-AMPc 200 µM, um análogo permeável do
AMPc (E8C10: controle = 6,3 ± 0,8 mm2 x 10-2; 8-CPT-AMPc = 19,9 ± 1,5 mm2 x 10-2,
n=4).
CONCLUSÕES: Estes resultados em conjunto sugerem que o aumento intracelular de AMP
cíclico seja capaz de inibir o crescimento glial em culturas de células de retina de embrião de
galinha submetidas à lesão mecânica.

Macroalgas marinhas são ricas em compostos bioativos com atividades anti-inflamatória,
antimicrobiana, antitumoral, antiviral e antioxidante (Mayer et al. 2011). Desta forma, o
estudo de produtos extraídos de algas marinhas vermelhas tem despertado interesse, como
novas estratégias de tratamentos de várias patologias. No entanto, no sistema nervoso central
(SNC) até o momento, não há relatos das potencialidades terapêuticas de tais produtos. O
objetivo do presente estudo foi avaliar os efeitos dos extratos alcoólicos das algas vermelhas
Amansia multifida e Meristiella echinocarpa em modelos experimentais de convulsão
química induzida por Pentilenotetrazol (PTZ) e Estricnina (EST). As algas vermelhas
Amansia multifida e Meristiella echinocarpa foram coletadas durante a maré baixa, em
Paracuru (São Gonçalo do Amarante, Ceará, Brasil). Em seguida foram lavadas com água
destilada, para remover epífitas e outros organismos do solo, secadas, maceradas e
liofilizadas. A amostra liofilizada foi submetida à extração duas vezes com etanol a 70% em
1:20 (m/v). Os extratos etanólicos de A. multifida (EEAm) e M. echinocarpa (EEMe) foram
concentrados por destilação sob pressão reduzida antes da sua utilização nos ensaios
biológicos. Camundongos Swiss fêmeas (25 a 35 g) foram mantidos em ciclo claro/escuro de
12 h, com livre acesso a água e ração. EEAm (2.5, 5, 10 mg/kg) e EEMe (10, 20, 40 mg/kg)
foram dissolvidas em salina estéril (NaCl 0,9%) e administradas por via intraperitoneal (i.p.)
30 min. antes da administração de PTZ (100 mg/kg; i.p.) ou EST (20 mg/kg; i.p.). O grupo
controle recebeu salina. Imediatamente após a administração do PTZ ou EST, os animais
foram colocados em caixas individuais e observados por 20 min. Os experimentos foram
aprovados pelo Comitê de Ética e para o uso de animais de experimentação da UECE (CEUA
n°3319515129/14). O EEAm não inibiu as atividades convulsivantes promovidas por EST e
PTZ. O EEMe também não inibiu a atividade convulsivante promovida pela EST, mas
aumentou a latência de morte no modelo de convulsão induzida pelo PTZ na dose de 20
mg/kg (446,6 ± 112,3 vs controle: 37,67 ± 6,433). Podemos concluir que a EEMe, mas não a
EEAm apresenta efeito neuroprotetor no modelo de convulsão induzida por PTZ.

ACTIONS OF CAFFEINE ON GABAERGIC SYSTEM OF DEVELOPING CHICK
EMBRYO RETINA

Objectives: To evaluate the effects of 2-day in vivo exposure to caffeine on GABAergic
system of chick embryo retina.
Materials and Methods: This project was approved by the animal research ethics committee
of the Federal Fluminense University (no 112/09). Eggs of Gallus domesticus with 14
embryonic days (E14) were injected with caffeine (30mg/mL), or vehicle (control). At E16,
retinas were collected and submitted to western blot, immunohistochemistry, or [³H]-GABA
uptake and release techniques. The results obtained from western blot and GABA
immunoreaction were quantified through optical densitometry mediated by ImageLab and
ImageJ softwares and analyzed by teste T. Number of GABA+ cells were counted in 5 fields
in each triplicate and analyzed (test T). Results of [³H]-GABA uptake and [³H]-GABA-
release induced by glutamate were obtained as CPM through a liquid scintillation technique
and statistically analyzed.
Results: Caffeine induced an increase in the immunoreactivity of GABA in amacrine cells
(vehicle=100% n=4; caffeine=124.8±9.21 n=4 P<0.05), horizontal cells (121.6±6.8% n=4
p<0.05) and cells in the ganglion cell layer (145.9±19.9% n=4 p<0.01). Although, caffeine
reduced the levels of GAT-1, a GABA transporter subtype (vehicle= 100% n=4
caffeine=60.71±11.48% p<0.05 n=7) we were not able to find changes in [³H]-GABA uptake
during 1 hour (vehicle=100%, n=6; Caffeine=103±11.71%, n=6), or 40 minutes of incubation
(vehicle=100% n=8; caffeine=101.6±7.44% n=8). Glutamate-induced [³H]-GABA release
was also unaffected by caffeine treatment (vehicle=6.54±1.31% n=5; caffeine=6.84±1.11%
n=6)
Even though GAD protein levels did not suffer any effect upon caffeine treatment,
(vehicle=100% n=10, caffeine=104.2 ± 8.944% n=10), we noticed a decrease in the number
of GAD+ amacrine cells (vehicle=100% n=4; caffeine=60.7917% p<0.01 n=4).
Conclusions: This data suggests that caffeine can modulate some aspects of GABAergic
system in the developing chick embryo retina. However, the mechanism responsible for the
increase in the GABA cell content remains to be elucidated.

INTRODUÇÃO: Atualmente, as doenças crônicas como o diabetes e a hipertensão arterial
sistêmica representam um importante problema de Saúde Pública para o Brasil1,2. Dado ao
seu potencial de provocar o acidente vascular encefálico (AVE), que cursa com a perda
repentina da função neurológica, decorrente à interrupção do fluxo sanguíneo para o encéfalo,
privando-o de oxigênio e de nutrientes essenciais3. O AVE isquêmico é o tipo de agravo
comum entre pessoas idosas, impingindo-lhes imobilidade física. MÉTODOS: Revisão de
literatura, utilizando livros didáticos e os recursos da biblioteca virtual de saúde (BVS), entre
2008-2014. OBJETIVO: Demonstrar a capacidade cerebral, de reconstituição dos
movimentos musculares perdidos em pacientes idosos, acometidos por AVE.
RESULTADOS: Foram 05 os textos disponíveis on line em língua vernácula e 01 livro
didático, que fundamentaram a DISCUSSÂO: Quando o cérebro apresenta uma área com
funcionamento precário, o mesmo utiliza-se de áreas adjacentes, para suprir tal necessidade4.
Ainda que os movimentos adquiridos pós-precariedade cerebral, não correspondam aos
antecessores, os mesmos demonstram que a Plasticidade – um processo adaptativo cerebral4,
com propriedades de atuar na estrutura e função do sistema nervoso afetado, a partir da
interação com o meio externo, levando informações ao meio interno5. O Brain-derived
neurotrophic factor (BDNF), uma proteína, importante ao desenvolvimento do sistema
nervoso adulto, exerce ações neuroprotetoras em doenças neurodegenerativas, como a
depressão, a esquizofrenia, o transtorno obsessivo-compulsivo, a doença de Alzheimer e a
esclerose múltipla ou após lesão cerebral5. Apresentando influência na plasticidade cerebral,
assim como no aprendizado e na memória. Tal proteína é também responsável pela
neuroplasticidade relacionada ao exercício físico, sendo sintetizada e liberada pelo músculo
esquelético, exercendo uma função trófica nos neurônios motores5. CONCLUSÂO: Sendo o
corpo dotado de uma interioridade, não explicável somente pela imanência da consciência e
nem pela sua exterioridade6, pode-se conceber que a essência entre o campo externo e o
interno, seja a multiplicidade que envolve a integralidade, a subjetividade e singularidade das
pessoas, que a enfermeira busca estimular, quando administra cuidados técnicos focados no
corpo e, espirituais focados nas estruturas mentais.

INTRODUÇÃO: A Política Nacional de Educação Permanente em Saúde (PNEPS1, 2004)
visa fortalecer o SUS, através da educação dos profissionais da saúde2. MÉTODO: Revisão
integrativa de literatura, utilizando os recursos virtuais no espaço 2004-2013, mediante
utilização dos descritores: Enfermagem; Educação continuada; Saúde, para atender ao
OBJETIVO: Conhecer como a PNEPS, pode contribuir com a ergonomia mental dos
trabalhadores da saúde? RESULTADOS: Selecionou-se 05 publicações disponíveis na
biblioteca virtual de saúde. DISCUSSÂO: A ergonomia coloca-se como aliada da PNEPS,
uma vez que o prefixo Ergon e o sufixo Nomos, provenientes do grego indicam “trabalhar” e
“legislação”3. A ergonomia tem como finalidades, promover a adequação ou modificação
dos sistemas de trabalho e das atividades, às características, habilidades e limitações das
pessoas3; já a PNEPS - também com caráter humanitário, busca além de educar para a saúde
no trabalho, atender a saúde física e mental, integralizando outras dimensões humanas, na
busca do bem-estar físico, mental e social4, dignificado o trabalhador, com a promoção do
equilíbrio entre a satisfação profissional e a vida dentro e fora do ambiente de trabalho5.
CONCLUSÃO: A PNEPS como um instrumento voltado à construção de um saber-fazer e
refazer as ações profissionais age como um processo ergonômico, auxiliando o trabalhador,
na criação de novas habilidades individuais e coletivas, permeadas por mudanças que
somente a educação pode promover, na transformação dos comportamentos cognitivos,
afetivos e psicomotores das pessoas, permitindo a construção ou a reconfiguração de atitudes
seguras e humanizadas, durante a prestação da assistência na área da saúde.

involved with nervous system development. Previous studies from our group demonstrated
that exogenous ATP was able to regulate the proliferation of retinal progenitor cells in vitro
via P2Y1 receptor, a G protein coupled receptor. Based on these results, the objective of this
study was to evaluate the function of adenine nucleotides in vivo during the development of
the retina of newborn rats. The intravitreal injection of PPADS, a P2 receptors inhibitor,
decreased the number of proliferating cells labeled with BrdU in approximately 23% at the
concentration of 1000 µM in the P2 animals. Furthermore, we identified the presence of
P2Y1 receptor in all layers of the retina at P2 animals, including proliferating Ki-67 positive
cells in neuroblast layer, suggesting that it is a possible candidate receptor for the action of
adenine nucleotides on the neuroblasts proliferation. Indeed, intravitreal injection of
MRS2179, a selective antagonist of P2Y1 receptor, at the concentration of 100 µM reduced
approximately 21% of the neuroblasts proliferation at P2 animals. No profile of cell death
was observed during the treatment with this drug. There was no change in expression of the
p27 protein, a cyclin-dependent kinase inhibitor, and no alteration in the number of phospho-
histone H3 cells after the treatment with MRS2179. Intravitreal injection ADPβ-S and ATPɣ-
S, a non-hydrolysable analog of ADP and ATP respectively, at the concentration of 100 µM
did not affect the proliferation rate. However, in the presence of apyrase, an enzyme that
hydrolyzes nucleotides, both ADPβ-S and ATPɣ-S, at the same concentrations described
above, increased the number of proliferating cells in about 30%. Increasing endogenous ATP
at P4 animals by the treatment with ARL67156, an ectonucleotidase inhibitor at the
concentration de 200 µM, stimulated the proliferation of retinal cells in approximately 50%.
Therefore, we concluded that adenine nucleotides modulated proliferation of neuroblasts
during early stages of retinal development through P2Y1 receptor and probably changes in
this modulation could affect the histogenesis of retina and consequently it function.

OUABAÍNA REGULA A SINTESE E LIBERAÇÃO DE IL1-β EM CULTURAS DE
CELULAS DA RETINA

Ouabaína (OUA) é um hormônio esteróide que se liga à enzima Na+, K+ -ATPase e estimula
diferentes vias de sinalização intracelulares. Dados previos demonstraram a participação da
IL1-β, mediando o efeito trófico da ouabaína (3nM), sobre a população de células
ganglionares da retina (CGR) em cultura. Baseados nisso objetivamos avaliar o papel da
OUA na modulação da resposta inflamatória desta citocina, visando compreender os
mecanismos pelos quais a IL-1β contribui para a sobrevida das CGR.
Ratos da linhagem Lister Hooded (P0-P2) foram mortos, suas retinas dissecadas e as células
dissociadas e plaqueadas na densidade de 105 cels/cm2 em placas de Petri. As culturas foram
mantidas em meio 199 (CT) ou na presença de meio e OUA numa atmosfera de 5% CO2
/95% ar à 37ºC. Os níveis de IL1-β, IL1-RI (receptor de IL-1 do tipo 1) foram determinados
pela técnica de Western Blot e sua liberação através da técnica de ELISA (kit R&D systems
RatIL-1β/IL-1F2). A marcação da IL1- β for determinada pela técnica de imunohistoquímica
utilizando anticorpos biotinilados. Os procedimentos experimentais com animais foram
aprovados pela Comissão de Ética no Uso de Animais da UFF (00294/12).
Nossos resultados mostram que o tratamento com a OUA modula a produção de IL1-β:
15min (CT 100% OUA 168,46 ±2,7 n=4 p=0,008), 45mim (CT 100% OUA 101,16±2,18 n=3
p=0,7), 24h (CT 100% OUA 40,87±1,37 n=4 p=0,0001), 48h (CT 100% OUA 99,81±2,7 n=3
p=0,9); bem como os níveis de seu receptor IL1-R1: 15min (CT 100% OUA 124,56±6,08
n=3 p=0,0003), 45min (CT 100% OUA 112±2,3 n=4 p=0,02), 24h (CT 100% OUA
84,03±5,4 n=3 p=0,04) e 48h (CT 100% OUA128,56±7,1 n=4 p=0,03). A liberação desta
citocina também é modulada pela OUA: 15min (CT 6,31 pg/mL ±2,04 p=0,01 OUA 7,41
pg/mL ±1,93 p=0,006 n=8), 45 min (CT 13,7pg/mL ±2,35 p=0,0011 OUA 7,2pg/mL ±1,36
p=0,002 n=7), 24 horas (CT 6,57pg/mL ±2,2 p=0,02 OUA 19,52pg/mL ±3,7 p=0,002 n=7),
48 horas (CT 15,6pg/mL ±4,8 p=0,04 OUA 22,2pg/mL ±6,4 p=0,04 n=4). A
imunohistoquímica revelou a marcação para IL-1β na camada de células ganglionares.
Nossos dados demonstram que a OUA é capaz de modular a produção e a liberação desta
citocina, bem como os níveis do seu receptor, indicando que a presença deste esteróide em
baixas concentrações, regula a resposta inflamatória nas culturas de células da retina o que
pode estar relacionado ao seu efeito na sobrevida das CGR.

ESTUDO COMPORTAMENTAL EM MODELO 6-OHDA DA DOENÇA DE
PARKINSON

A doença de Parkinson é caracterizada pela degeneração progressiva e específica de
neurônios dopaminérgicos A9 da substância nigra compacta (SNc) que enviam aferências
para o corpo estriado, levando a sintomas motores característicos como bradicinesia, tremor
de repouso, rigidez e instabilidade postural. Além destes, sintomas não-motores também
podem ser observados, como depressão, alterações do sono, déficits cognitivos e alterações
gastrointestinais. Existem vários modelos para o estudo deste distúrbio, sendo o da lesão
unilateral com a 6-hidroxidopamina um dos mais utilizados. Nos últimos anos, nosso grupo
vem estudando o curso temporal das alterações bioquímicas que ocorrem após a lesão
dopaminérgica característica deste modelo. Neste trabalho, nosso objetivo foi avaliar o
comportamento rotacional no teste da apomorfina e a capacidade de memória de
reconhecimento de camundongos adultos C57Bl16 submetidos à lesão por 6-OHDA. Os
animais foram anestesiados e submetidos à injeção unilateral de veículo ou 6-OHDA (2µl a
uma taxa de 0,5 µl/5 min). 1, 2 e 4 semanas após a cirurgia, realizamos diferentes testes
comportamentais como o do campo aberto, análise do número de rotações para ao lado
contralateral da lesão após a injeção intraperitoneal de apomorfina (0,5 mg/kg) e o teste de
reconhecimento de objetos para avaliar a capacidade de memorização. Nossos resultados
mostraram que os camundongos lesionados apresentam um padrão rotacional para o lado
contralateral da lesão e os animais com 4 semanas de sobrevida deste grupo têm o menor
número de rotações. No teste de reconhecimento de objetos, os animais exploraram os
objetos de maneira igual na fase treino, mostrando que não há nenhum prejuízo motor que
pudesse afetar os resultados. Na fase teste, os animais lesionados obtiveram um índice de
discriminação negativo, sugerindo um déficit no reconhecimento de objetos.

DEVELOPMENT OF RETINOGENICULATE AND RETINOTECTAL
CONNECTIONS AT MICE WITH RETINAL DEGENERATION.

The retina is responsible for capturing light through the photoreceptor cells, and transmit
visual signals through the ganglion cells to subcortical targets as dorsal lateral geniculate
nucleus (NGLd) and superior colliculus (SC). Initially, the formation of topographic maps of
the retina at the targets depends on attractive/repulsive cues that guide the growth of axons to
specific areas on target. Subsequently, the pattern of spontaneous and evoked electrical
activity induced by opening of the eyes, is responsible for refining the initial projections,
generating a high specificity pattern of connections. The administration of drugs that
interfere with neuronal electrical activity compromises the formation of sub-cortical maps
and primary visual cortex. Retinal diseases affecting the electrical activity can influence the
formation of retinotopic maps. Retinitis pigmentosa (RP) is a group of inherited diseases that
cause degeneration of retinal photoreceptors leading to gradual loss of vision. RDS mice
(slow retinal degeneration) have a progressive loss of photoreceptor by apoptosis during the
first two postnatal weeks, with peak degeneration at postnatal day 18 (DPN18). The
electroretinogram of these animals are always reduced and becomes absent within 4 weeks.
The aim of this study was to evaluate the retinofugal projections in RDS animals comparing
them to control animals at different stages of development. Our results showed that the
distribution of retinal fibers in RDS animals of the second postnatal week is normal. It is
possible to note the visual clusters at ventral layer of the superior colliculus and axons from
ipso and contrallateral retina slightly segregated in the thalamus. Animals of the third week
(DPN21) have presented some fibers dispersed in the visual ventral layer of the SC and also
in the thalamus, but no quantitative difference was observed. Adult animals of two months
have a pattern still compatible with normal animals with the presence of retinal afferents in
the subcortical targets analyzed, even in the absence of retinal electrical activity. Taken
together, these results show that retinofugal projections are formed even during normal
degeneration of photoreceptor. Moreover, the retinal ganglion cells maintained their synaptic
contacts the target even long after the complete loss of photoreceptors, suggesting that the
prolonged maintenance of these pathways may contribute to the maintenance of cortical
activation.

CARACTERIZAÇÃO DO CONTEÚDO E ATIVIDADE DE ENZIMAS
COLINÉRGICAS ESTRIATAIS EM UM MODELO ANIMAL DE DOENÇA DE
PARKINSON INDUZIDA POR 6-OHDA

Objectives: Acetylcholine (ACh) is present in the striatum via cholinergic interneurons. ACh
is synthesized by choline acetyltransferase (ChAT) and it can be hydrolyzed by
acetylcholinesterase (AChE), which ensures a rapid decrease of ACh after its release from pre
synaptic terminals in the synaptic cleft. Studies have shown that the normal activity of striatal
neurons depends on the balance between dopamine (DA) and ACh and those disruptions in
this balance may contribute to the development of Parkinson's disease (PD), a
neurodegenerative disease characterized by a severe and progressive motor impairment due
to progressive and specific degeneration of A9 dopaminergic neurons of the substantia nigra
compacta. Therefore, we aimed to investigate, in a mouse experimental model of PD,
biochemical changes in the cholinergic system during different survival periods (1, 2 and 4
weeks) after PD induction, in order to study the possible striatal cholinergic modulation in
these animals.
Methods: Adult C57Bl6 mice were anesthetized and submitted to unilateral injection of 6-
OHDA (2μl at a rate of 0.5 µl/5 min) into the left medial forebrain bundle using stereotaxic
procedures. Mice were then allowed to survive for either 1, 2 or 4 weeks thereafter. The mice
were euthanized and striatum was dissectioned to prepare samples for western blot (WB)
analysis (we used anti-AChE and anti-ChAT) and determination of AChE and ChAT activity
according to the colorimetric method of Ellman et al. (1961) and Fonnum (1975)
respectively.
Results: Our data show that the injured side presented a higher activity of ChAT and lower
AChE activity in the early stage after injection of the drug. Increased ChAT activity can be
explained at least in part by the increased expression at this time. The expression of AChE
did not change during the time period studied, suggesting the existence of another regulatory
mechanism that does not involve changes in transcription processes. The modulation of the
cholinergic system which occurs in response to the injection of 6-OHDA in this model
indicates a biochemical reorganization in response to the striatal dopaminergic loss.

O TRATAMENTO COM PMA MODULA A LIBERAÇÃO DAS CITOCINAS
INFLAMATÓRIAS TNF ALFA E IL-1 BETA EM CÉLULAS DA RETINA DE
RATOS NEONATOS

A proteína cinase C (PKC) está envolvida em vários eventos no desenvolvimento do sistema
nervoso(SN). O tratamento com forbol 12-miristato 13-acetato (PMA), ativador da PKC,em
células da retina de ratos neonatos, em 48 horas aumenta a sobrevida das células ganglionares
(CGRs). Dados mostraram que o tratamento das culturas da retina com TNFα e IL-1β
aumenta a sobrevida das CGRs.Demonstramos que o PMA modula os níveis intracelulares de
TNFα e IL-1β,causa aumento no início e queda a partir de 24h. O objetivo do presente
trabalho foi analisar o efeito temporal do PMA 50ng/mL na liberação das citocinas IL-1β e
TNFα em culturas de células da retina.
Ratos Lister Hooded (P0-P2) foram decapitados, suas retinas dissecadas, incubadas em
solução de tripsina a 0,1%,dissociadas por pipeta Pasteur. As células foram plaqueadas na
densidade de 105 céls/cm2. As culturas, mantidas em meio 199 (CT), ou na presença do
PMA 50ng/mL, em atmosfera de 5% CO2/95% ar/37ºC por diferentes períodos de tempo. Os
níveis extracelulares foram medidos pela técnica de ELISA com o kit “Quantikine ELISA
ReD systems rat TNFα e rat IL-1β/IL-1F2”. Os valores constitui a média ± erro padrão. As
diferenças foram analisadas por ANOVA one-way e o teste post-hoc Newman-Keuls. Os
dados foram significativos quando p<0,05. Os procedimentos com animais foram aprovados
pela CEUA da UFF (00294/12).
Nossos resultados mostram que o PMA não altera a liberação de IL-1β em relação ao
controle após 15min (CT 67,8±27pg/ml;PMA 147,4±55pg/ml; n=8) e 45min (CT
74,7±18,8pg/ml;PMA 92,4±17,2pg/ml; n=6). O tratamento por 24h induz um aumento, em
relação ao controle 24h (CT 43,4±14,9pg/ml;PMA 86,6±25pg/ml; n=7). Após 48h de
tratamento não há diferença nos níveis em relação ao controle CT 101,5±16,9pg/ml;PMA
161±50pg/ml; n=5). Em relação aos níveis extracelulares de TNFα, não há alterações por
15min (CT 6,3±2pg/ml;PMA 10,3±2,35pg/ml; n=6) e 45min (CT 13,8±2,3 pg/ml;PMA
8±1,35 pg/ml; n=7). Após 24h de tratamento observamos um aumento significativo nestes
níveis (CT 6,5±2,2pg/ml;PMA 21,6±4,66pg/m; n=7), sem diferença em relação ao controle
após 48h (CT 20,6±4,8pg/ml;PMA 15,8±4,08pg/ml; n=4).Nossos dados mostram que a
ativação crônica da PKC modula a liberação de TNFα e IL-1β. Há um aumento nos níveis
intracelulares no início, sugerindo regular a produção, aumentando a liberação em relação ao
controle em 24h. A regulação sobre a produção e liberação das citocinas pode ser notável
para o efeito do PMA na sobrevida das CGRs

THE BLOCKADE OF P2Y1 RECEPTORS ALTER HORIZONTAL CELL IN VIVO
OF RETINA RAT

The retina is a tissue member of the central nervous system, it consists of six different types
of neurons (cone and rods photoreceptors, horizontal, amacrine, bipolar and ganglion) and a
type of glial cell (Müller glia), divide in nuclear and plexiform layers in the adult. Changes
that occur during development can cause alterations in histology and consequently in its
functioning. Adenine nucleotides are known to modulate neuroblasts cell proliferation in the
early stages of retinal development. Previous results of our group reported that intravitreal
injection of MRS2179, a selective antagonist for the P2Y1 receptor, at concentration of 50
μM and 100 μM reduced cell proliferation approximately 20% in rats at 3-postnatal (P3).
Based on this data, the purpose of this study was to evaluated how the reduction in the
number of proliferating cells in the retina, induced by the blockade of P2Y1, could affect the
histogenesis of this tissue. The treatment with MRS 2179 at concentration of 50 μM,
decreased 30% in the number of calbindin positive horizontal cells and apparently there was a
reduce dendritic arborization in these cells, after 20 hours of treatment. To exclude the
possibility of death induction by this drug, the assessment of apoptotic profiles was
performed during the treatment period, but did not identify any patterns of death. Amacrine
cells and photoreceptors were evaluated after 20, 48 and 72 hours of treatment, and did not
change in cell number and morphology was observed in these populations. We conclude that
adenine nucleotides acts during early stages of retinal development, in P2 apparently
modulated the horizontal cells differentiation. But it is necessary to assay other population
after MRS 2179 treatment, so as to obtain overview of P2Y1 blockade in the differentiation
of retinal cells.

CUIDADO MATERNO NEONATAL E EXPRESSÃO DE FATORES
NEUROTRÓFICOS NO SISTEMA NERVOSO CENTRAL

Objetivos:
A separação materna tem sido empregada como modelo experimental para avaliar os efeitos
do estresse neonatal ao longo do desenvolvimento. Ratos neonatos privados da mãe
apresentam alterações na citoarquitetura do hipocampo e na expressão de fatores tróficos
nesta região, destacando-se o fator trófico derivado do cérebro (BDNF). Algumas dessas
alterações vêm sendo correlacionadas com a propensão ao desenvolvimento de
psicopatologias como, por exemplo, a depressão maior. O objetivo de nosso trabalho foi
analisar o efeito da separação materna, por 30 minutos e por 6 horas, durante a primeira
semana pós-natal, na expressão de BDNF e IGF-1 no hipocampo e na retina.
Métodos e Resultados:
Animais da linhagem Lister Hooded na idade de P2 (72h pós nascimento) foram separados
das mães, durante sete dias, por trinta minutos a cada dia. Sempre foi obedecido o mesmo
horário para a separação dos animais (15:30 -16:30 para privação por 30 minutos e 17:00 -
23:00 para animais privados por 6 horas). No oitavo dia os animais foram sacrificados as
retinas e o hipocampo foram retirados e processados para a extração de proteínas. Os níveis
de BDNF e IGF-1 foram avaliados através da técnica de western blot. Todo o procedimento
com animais foi feito de acordo com os princípios éticos de experimentação animal da
SBCAL cuja aprovação nos fora previamente dada pelo comitê de ética e pesquisa animal
(CEPA) da Universidade Federal Fluminense (projeto # 00196/10). Nossos resultados
indicam que a privação materna por 30m dia, induz um aumento significativo nos níveis de
BDNF e IGF-1 na retina e no hipocampo em p9. O animais privados por 6 horas dia (p2 até
p8) foram avaliados em P30. Interessantemente observamos que estes animais apresentam os
níveis de BDNF aumentados.
Conclusão:
Nossos dados indicam que a privação materna, por curto espaço de tempo, aumentou os
níveis de BDNF sugerindo uma possível resposta adaptativa. Além disso, nossos resultados
sugerem que as alterações na retina possivelmente reflitam os efeitos sistêmicos do estresse.
Esse resultado nos leva a sugerir também que o sistema visual como mais uma região a ser
analisada visando a compreensão dos efeitos sistêmicos do estresse.

DETERMINATION OF CALPAINS AND METALLOPROTEASES ACTIVITIES IN
MDX MOUSE SKELETAL MUSCLE ON THE PHASE OF MYONECROSIS AND
REGENERATION

Objetivos:
Context: Duchene muscular dystrophy (DMD) in humans and in the mdx mouse is a genetic
inflammatory myopathy with mutations/deletions in locus Xp21 coding for dystrophin, a
protein of the sarcolemma which associates actin filaments and transmembrane glycoprotein
complex. Lack of dystrophin compromises the integrity of the sarcolemma resulting in
greater intracellular influx of calcium ions by activating the autoproteolysis cascade and cell
death by necrosis with further activation of inflammation. Calpains are calcium dependent
enzymes with major role on cytoskeletal remodeling. Metalloproteases (MMPs) are Ca+2 and
Zn-dependent endopeptidases, with two isoforms: MMP-2 and MMP-9. MMP-9 is secreted
mainly by inflammatory leukocytes, and MMP-2 in constitutive form by myoblasts is
associated with proliferation, migration and fusion of myogenic cells for efficient muscle
regeneration. Objective: Relate the activity of Calpains and MMPs with myonecrosis in the
mdx mouse. Methods: We used 4 and 12 weeks-old male mdx mice at the height of
myonecrosis and muscle inflammation, and C57BL10 as control nondystrophic.
Gastrocnemius and brachial muscle of both lineages was processed in appropriate buffer for
gel zymography 7.5% with gelatin for identification of MMPs, as described by Heussen e
Dowdle (1980). And gel 10% with casein for µ and m Calpains activity according to protocol
described by Arthur e Mykles (2000). The results analysis was done by densitometry in L-Pix
Image (Loccus, Brazil). Results: It was observed an increase in the activity of MMP-9 and
Calpains in gastrocnemius muscle of mdx mice. The data indicate the presence of elevated
levels of intracellular Ca+2 activation in mdx dystrophic muscles thus suggesting intense
activation of Calpains proteolytic cascade associated with myonecrosis and also
inflammatory infiltrate due to an increase in the MMP-9 activity. Discussion: At the peak of
mdx myonecrosis the release of inflammatory cytokines by activated leukocytes contribute to
muscle degeneration due to necrosis and apoptosis of myofiber, and further development of
peripheral neuropathy due to loss of innervations. Conclusion: By using this methodology we
were able to demonstrate that mdx skeletal muscle presents at the height of myonecrosis
increased activity of Calpains related to muscle degeneration and increased inflammatory
activity (MMPs) mediated by activated leukocytes.

A proteína cinase C (PKC) é uma serina-treonina cinase mediadora de diversos eventos
intracelulares tais como proliferação, diferenciação, apoptose e sobrevida. O tratamento das
culturas de células da retina de ratos com forbol 12- miristato 13-acetato (PMA), um ativador
da PKC, por 48h promove um aumento na sobrevida das células ganglionares da retina
(CGR). Dados do nosso grupo demonstraram que o tratamento das culturas da retina com IL-
1beta, uma citocina pró-inflamatória, em uma faixa estreita de concentração, aumenta a
sobrevida das CGRs. O objetivo do presente estudo foi de analisar o efeito do tratamento
com PMA 50ng/mL nas células da retina, sobre os níveis da IL1-beta em diferentes períodos
de tempo.
Ratos Lister Hooded (P0-P2) foram decapitados, suas retinas dissecadas e incubadas em
solução de tripsina a 0,1%, e em seguida dissociadas por pipeta Pasteur. As células foram
semeadas (1,0 X105 céls/cm2) em placas de Petri de 35mm. As culturas foram mantidas em
meio 199 (CT), ou na presença do PMA 50ng/mL, em atmosfera umidificada de 5% CO2
/95% ar/37ºC) por diferentes períodos de tempo. Os níveis de IL1-beta foram determinados
pela técnica de Western blot. Os procedimentos experimentais com animais foram aprovados
pela Comissão de Ética no Uso de Animais da UFF (00294/12).
Nossos resultados demonstram que o tratamento com o PMA aumenta os níveis intracelulares
de IL1-beta em culturas tratadas por 15 minutos (CT 100% PMA 143,3% ±0,56 n=3) e por 45
minutos (CT 100% PMA 167,25% ±10 n=3). Já as culturas tratadas com PMA por 24 horas
exibem uma diminuição significativa nos níveis de IL1-beta (CT 100% PMA 52,1% ±9,3
n=3) e essa queda persiste após 48 horas em cultura (CT 100% PMA 79% ±11 n=3).
Nossos dados demonstram que a ativação da PKC produz um aumento inicial nos níveis
intracelulares de IL1-beta, seguido por uma queda após 24 horas em cultura. Essa modulação
dos níveis de IL1-beta pode estar relacionada ao aumento na sobrevida das células
ganglionares da retina mediado pelo tratamento com PMA.

TRATAMENTO COM OUABAÍNA REGULA PROTEÍNAS DA FAMILIA DA IL-1
EM CULTURAS DE CÉLULAS DA RETINA

Ouabaína (OUA) liga à enzima Na+, K+ -ATPase e estimula diferentes vias de sinalização
intracelulares.Nossos estudos demonstraram que o tratamento com OUA é capaz de modular
a produção e liberação de IL1-β e que esta citocina atua mediando o efeito trófico da
OUA(3nM), nas células ganglionares da retina (CGR) em cultura.Objetivamos avaliar se o
tratamento com a OUA é capaz de modular também a produção de IL-1Ra, molécula com
perfil anti-inflamatório, membro da família da IL-1. O objetivo é compreender de que forma
a modulação das moléculas da família da IL-1 contribui para o efeito de sobrevida das
CGR.Ratos da linhagem Lister Hooded (P0-P2) foram mortos, suas retinas dissecadas e as
células dissociadas e plaqueadas na densidade de 105 cels/cm2 em placas de Petri.Culturas
mantidas em meio 199 (CT) ou na presença de meio e OUA numa atmosfera de 5% CO2
/95% ar à 37ºC. Os níveis de IL-1Ra e de IL-1α determinados pela técnica de Western
Blot.Os procedimentos experimentais com animais foram aprovados pela Comissão de Ética
no Uso de Animais da UFF (00294/12).Resultados preliminares mostraram que o tratamento
com a OUA modula a produção de IL1-β: 15min (CT 100% OUA 168,46 ±2,7 n=4 p=0,008),
45mim (CT 100% OUA 101,16±2,18 n=3 p=0,7), 24h (CT 100% OUA 40,87±1,37 n=4
p=0,0001), 48h (CT 100% OUA 99,81±2,7 n=3 p=0,9); bem como os níveis de seu receptor
IL1-R1: 15min (CT 100% OUA 124,56±6,08 n=3 p=0,0003), 45min (CT 100% OUA
112±2,3 n=4 p=0,02), 24h (CT 100% OUA 84,03±5,4 n=3 p=0,04) e 48h (CT 100%
OUA128,56±7,1 n=4 p=0,03). A liberação desta citocina também é modulada pela OUA:
15min (CT 6,31 pg/mL ±2,04 p=0,01 OUA 7,41 pg/mL ±1,93 p=0,006 n=8), 45 min (CT
13,7pg/mL ±2,35 p=0,0011 OUA 7,2pg/mL ±1,36 p=0,002 n=7), 24 horas (CT 6,57pg/mL
±2,2 p=0,02 OUA 19,52pg/mL ±3,7 p=0,002 n=7), 48 horas (CT 15,6pg/mL ±4,8 p=0,04
OUA 22,2pg/mL ±6,4 p=0,04 n=4). Diante destes dados avaliamos se nosso tratamento
modulava a produção de IL-1Ra e IL-1alfa e verificamos que os niveis de IL-1Ra foram
modulados pela ouabaína: 45mim (CT 100% OUA 129,2±2,5,87 n=3 p=0,03), 48 horas (CT
100% OUA 142,2±3,39 n=3 p=0,006). No entanto a ouabaína não alterou os níveis de IL-
1alfa: 15mim (CT 100% OUA 103,4±10,3 n=3 p=0,6), 24 horas (CT 100% OUA 95,62±5,7
n=3 p=0,52).Nossos resultados mostram que o tratamento com OUA além de ser capaz de
modular ao longo do tempo a produção e a liberação da IL-1β, bem como os níveis do seu
receptor, também modula os níveis da citocina anti-inflamatoria IL-1Ra.

INIBIÇÃO DA ENZIMA ADAM10 INTERFERE NA PLASTICIDADE DAS
PROJEÇÕES RETINOTECTAIS DE ROEDORES

A Doença de Alzheimer é um mal que acomete milhões de pessoas no mundo atualmente.
Sua fisiopatologia está associada a um desbalanço proteolítico no processamento da Proteína
Precursora do Amilóide (APP), resultando em um aumento na produção do peptídeo beta
amiloide e na redução dos níveis da forma solúvel α (sAPPα). Sua função fisiológica, assim
como de seus metabólitos, ainda não é completamente compreendida, apesar de diversos
grupos sugerirem funções como participação nos processos de sinaptogênese, sobrevivência,
crescimento celular e plasticidade sináptica. Nosso grupo, nos últimos anos, vem buscando
decifrar o papel da APP na plasticidade das conexões retinocoliculares utilizando o sistema
visual de roedores como modelo de estudo. Nossos dados prévios mostram que a expressão
da APP no CS de ratos é alta até a terceira semana pós-natal e que, no modelo de plasticidade
, enucleação monocular (EM) esta sofre redução transitória nos seus níveis, assim como um
favorecimento da via não amiloidogênica, levando a um aumento dos níveis de sAPPα. Neste
contexto, este trabalho visa investigar os efeitos da inibição da principal α-secretase,
ADAM10, sobre a distribuição das projeções retinocoliculares ipsolaterais de animais
submetidos à EM. Ratos pigmentados (Lister Hooded) foram submetidos ao implante
intracraniano de ELVAX contendo GI204523X (10 e 20mM) ou veículo (DMSO) em DPN7.
Em DPN10, os animais, implantados ou não, foram anestesiados e submetidos à EM. A
distribuição dos terminais ipsolaterais foi examinada em cortes cerebrais coronais após a
injeção intraocular de HRP. Nossos resultados mostraram que os animais implantados com o
inibidor na concentração de 10mM apresentaram uma ausência da expansão dos terminais
ipsolaterais, com clusters bem delimitados e poucas fibras nas regiões mais dorsais, perfil
semelhante ao visto em um animal não enucleado. Estes dados foram confirmados pela
análise quantitativa baseada na densidade óptica da marcação da camada visual total, assim
como da camada de clusters. Neste contexto, nosso trabalho sugere que a ADAM10 promova
o aumento de sAPPα e que esta proteína tenha um papel crucial na plasticidade das fibras
retinocoliculares induzida por EM.

MODELO DA DOENÇA DE PARKINSON INDUZIDO POR 6-OHDA PROMOVE
INFLAMAÇÃO E ALTERAÇÕES MORFOLÓGICAS NO SISTEMA NERVOSO
ENTÉRICO

Objetivos e Métodos
O objetivo deste trabalho é investigar se o modelo animal da Doença de Parkinson (DP)
induzido por 6-OHDA promove alterações funcionais, morfológicas e quanto a presença de
marcadores inflamatórios no Sistema Nervoso Entérico (SNE) colônico. Para a indução do
modelo, camundongos C57Bl6 adultos foram submetidos à administração unilateral de 6-
OHDA na região do estriado esquerdo por procedimentos estereotáxicos. Um segundo grupo
de animais operados não lesionados (sham) foi utilizado como controle. Ambos os grupos
tiveram tempos de sobrevida de 1, 2 e 4 semanas. O ensaio de constipação foi realizado a fim
de acessar a proporção relativa de água nas fezes dos animais e analisar o estado funcional do
SNE. Após este ensaio os animais foram eutanasiados e o cólon removido para realização de
procedimentos de histoquímica e imunofluorescência. Utilizamos anticorpos anti-IBA1
(proteína presente em macrófagos) e anti-GFAP (proteína marcadora de células gliais).
Resultados
Os resultados de histoquímica demonstram a partir de 1semana pós-lesão a camada muscular
intestinal comprometida nos animais tratados com 6-OHDA. Também neste grupo, a partir do
tempo de 2 semanas de sobrevida, foi possível notar descaracterização da estrutura da mucosa
intestinal, nas porções oral e anal do cólon e o contínuo comprometimento da camada
muscular. O plexo mioentérico da porção oral nas sobrevidas 1 e 2 semanas mostrou-se
hipertrofiado. Especificamente nos animais tratados com 6-OHDA com 1 semana pós-lesão,
notou-se a presença de infiltrado inflamatório na porção oral. Com 4 semanas pós-lesão o
quadro alterado da citoarquitetura continuou sendo observado e células hipertrofiadas no
plexo mioentérico anal puderam ser observadas. Quanto à imunofluorescência, houve um
aumento da expressão da proteína IBA1 na mucosa intestinal dos animais tratados com 6-
OHDA 1, 2 e 4 semanas comparados aos animais sham. Além disso, também ocorreu
aumento da expressão de GFAP nos animais tratados com 6-OHDA nos tempos de 1 e 2
semanas. Estas alterações ocorreram em ambas porções oral e anal do cólon.
Conclusão
As alterações apresentadas pelos animais tratados com 6-OHDA demonstram que o SNE e o
trato gastrintestinal encontram-se comprometidos neste modelo animal da DP. O aumento da
expressão de IBA1 na mucosa intestinal demonstra a ocorrência de inflamação local enquanto
o aumento de GFAP sinaliza a ocorrência de reatividade glial entérica no cólon de animais
modelo da DP.

GLUTAMATERGIC SIGNALING AND PSD-95 PROTEIN MODULATES NITRIC
OXIDE LEVELS IN A MICE MODEL OF PARKINSON´S DISEASE

INTRODUCTION: Parkinson’s disease (PD) is a progressive motor disorder characterized by
dopaminergic neuronal degeneration in the substantia nigra and severe motor symptoms. In
the postsynaptic density, dopamine receptors physically associates with scaffolding protein
PSD-95 that organizes the molecular architecture of postsynaptic sites and regulates
interactions between NMDA glutamate receptors and downstream-signaling molecules.
Alterations in PSD-95 levels and glutamatergic signalling were found in several
neuropsychiatric and neurological disorders such PD. This effect may influence dopamine
and NMDA receptors interactions and also nitric oxide (NOx) production. Synapsins are
phosphoproteins localized within presynaptic terminals and are thought to participate in the
regulation of the synaptic vesicle life cycle and neurotransmitter release. According to their
important roles in synapses formation, maintenance and plasticity, we focusing in evaluate
the levels of these pre- and post-synaptic proteins in a 6-OHDA model of PD in different
periods post-lesion.
METHODS: We used male C57BL/6 animals that underwent to unilateral striatal lesions
with intracranial injection of 6-hydroxydopamine neurotoxin, a dopamine analogue that
induce the selective death of dopaminergic neurons. After 1, 2 or 4 weeks post-injury, the
animals were subjected to behavioral analysis to assess motor behavior in an open field after
apomorphin ip injection. Then we proceed with Griess reaction to analyze NO production,
western blot and immunofluorescence techniques for PSD-95, synapsin and NMDAr subunits
(GluN1, GluN2a, GluN2b) in striatal samples.
RESULTS: In behavioral test, mice exhibited contra-lateral rotational activity and remarkable
difficulties in coordinating movements. 6-OHDA lesion into striatum induced an up-
regulation in PSD-95 levels and modification in GluN1, GluN2a, GluN2b subunits of NMDA
receptor composition. Synapsin expression tends to decrease two weeks post-lesion in the
injured side. Comparing control x lesioned side, NO production was diminished in lesioned
striatum 1,2 or 4 weeks post-lesion, wich correlates with PSD-95 increase and GluN2a
and/GluN2b shift.
CONCLUSION: In this study 6-OHDA induced pre and postsynaptic terminals
modifications. Indeed, NMDAR changes composition may reflect an adaptive system
response to dopamine depletion and rearrangement of theirs subunits together with PSD-95
increasing may influence NOx levels in striatum

Plasticidade neuroglial no modelo experimental da Doença de Parkinson

Objetivos: O objetivo do trabalho é investigar se o modelo da DP induzido por 6-OHDA
apresenta alterações na rede neuroglial em porções proximais e distais do intestino grosso 1,
2 ou 4 semanas pós-cirurgia.
Métodos: Camundongos C57Bl6 foram submetidos à administração unilateral de 6-OHDA no
estriado esquerdo. Outro grupo de animais operados não lesionados (sham) foi utilizado
como controle. Após a eutanásia, o intestino grosso foi coletado e dividido em duas porções
(oral e anal) para realização de imunofluorescência utilizando anticorpos anti-GFAP
(marcador de glia) e anti-α-sinucleína (marcador de terminais pré-sinápticos).Este trabalho
faz parte do projeto de pesquisa aprovado pelo comitê de ética em pesquisa animal local sob
o número 00129/09.
Resultados e Conclusões:
Podemos observar que o grupo 6-OHDA apresenta um aumento na marcação para GFAP na
mucosa da porção oral do intestino grosso com até 2 semanas de sobrevida, enquanto neste
mesmo grupo notamos uma tendência à diminuição na presença da célula glial na camada
muscular oral, que se acentua com 4 semanas sobrevida em relação ao controle. O cólon
distal parece ser mais sensível à administração de 6-OHDA, apresentando diminuição da
presença da célula glial a partir da 1ª semana pós-cirurgia, sendo mais expressivo na 4ª
semana. O conteúdo de α-sinucleína encontrou-se reduzido em todas as camadas avaliadas
tanto da porção oral, quanto na anal do intestino grosso de animais tratados com 6-OHDA.
Esses resultados indicam que reatividade glial encontra-se aumentada na mucosa e diminuída
na camada muscular dos animais submetidos ao modelo da DP. Há ainda uma evidente perda
de marcação neuronal no modelo da DP em todo intestino grosso. Diante deste cenário,
podemos sugerir que o modelo da DP induzido por 6-OHDA é capaz de causar uma ruptura
na circuitaria neuroglial entérica.

A COMPUTATIONAL MODEL OF LGN-V1 DYNAMICS AND THE EMERGENCE

Gamma oscillations have been hypothesized to play a central role in active vision,
contributing to inter-areal information selection and routing. Furthermore, emerging evidence
suggests that gamma oscillations play a fundamental role in feedforward neuronal
communication. In order to constrain these hypotheses it would be useful to know under what
conditions gamma is prominent during active vision. We recently showed (Bastos et al.,
2014) that gamma oscillations are an emergent property of the cortex, and are not present in
the LGN of alert behaving monkeys. Here, we set out to explain these results in terms of an
underlying biophysically realistic network model composed of Hodgkin-Huxley neurons. The
prime candidate mechanism to explain our experimental results was recurrent inhibition: the
anatomy of the cortex is full of recurrence, allowing excited pyramidal cells to activate
inhibitory cells, which can then inhibit a large number of pyramidal cells and set the pace of
the gamma oscillation (the classical "PING" mechanism - Pyramidal Interneuron Network
Gamma rhythm). However, this recurrence of excitation back onto local inhibitory cells,
appears to be lacking within the primate LGN (but perhaps not in the feline LGN) at an
anatomical level. In our model of the LGN lacking local recurrence, the circuit did not
engage in gamma oscillations even when the Thalamic Reticular Nucleus (TRN), a structure
that provides inhibition onto relay cells and is excited by geniculocortical collaterals, was
included. However, the cortex, which did contain recurrent inhibition, entered a rhythmic
mode despite receiving arrhythmic LGN input. Intriguingly, even when the cortex projected
back onto the LGN in a gamma rhythmic mode, this did not rhythmically entrain the LGN.
The primary reason for this appears to be the synaptic time constants of corticothalamic
synapses, which due to their slow time constants act as a low-pass filter. However, when we
did include local recurrent inhibition in the LGN (as a putative model of feline LGN), gamma
oscillations did emerge - which may explain interspecies differences between cats and
primates. We conclude that the circuitry of the primate LGN is well-suited to ignore cortical
gamma rhythms, to focus on a sparse and temporally precise neuronal code, and to maximize
the feedforward throughput of information from the retina to the cortex.

INHIBITION OF PHOSPHATASES ALTERS TOPOGRAPHIC ORGANIZATION
AND ENHANCES SYNAPSE MATURATION IN THE SUPERIOR COLLICULUS
OF THE RAT

Molecular processes of synaptic plasticity such as long-term depression and long term
potentiation (LTD and LTP) have been correlated with the establishment of topographical
maps, essential to the proper sensory system processing. Protein phosphatase 1 (PP1) and
calcineurin are phosphatases highly related to synaptic plasticity, acting as inductors of LTD
and synapse elimination. In this context, the objective of this work is to evaluate the roles of
PP1 and calcineurin and some of their main cellular targets in the development and
maintenance of retinocollicular topography in Lister Hooded rats. All experimental
procedures were approved by the local Animal Care Committee. Analysis of the ipsilateral
topography, using the anterograde labeling of horseheadish peroxidase (HRP), showed that
local inhibiton of PP1 using ELVAX inplants increased the density in terminal labeling of the
ipsilateral pathway when compared with the control group. This increase was consistent with
a sprouting of the ipsilateral axonal projections during the second postnatal week. We also
showed that pCREB content was increased six hours after either PP1, PP2B or PP1+PP2B
local inhibition. PSD95 content was also analyzed and we observed an increase in this protein
content following either PP1 and/or calcineurin inhibition in the SC during the second
postnatal week. Other markers of synapse maturation, such as a decrease in NR2B content
and an increase in NR2A glutamate receptors were also observed. Our results suggest,
therefore, that PP1 and PP2B may act together in the refinement of topographical maps in the
SC thought the control of mechanisms involved in the dynamics of synapse maturation and
elimination in the developing visual system.

EARLY ALCOHOL EXPOSURE CAUSES ABNORMAL TOPOGRAPHY IN
DEVELOPING VISUAL PATHWAYS, REDUCES PLASTICITY AND INDUCES

One the remarkable features of sensory systems is the precise targeting of connections
between sets of neurons conveying information along sensory pathways. In the visual system,
retinal ganglion cells axons form organized connections with the lateral geniculate nucleus
(LGN) and superior colliculus. The topographical fine-tuning is acquired by a combination of
activity-dependent and molecular cues-based processes that sculpts sensory pathways during
a critical period of brain development correspondent to the human last gestational
trimester/early years and to the first three postnatal weeks in rodents. Fetal alcohol spectrum
disorders are a major cause of mental retardation in western countries and children present
many alterations in sensory processing and activity-dependent neuronal plasticity that may
result from deficits in the correct wiring pattern of cortical afferents from subcortical nuclei.
We studied the impact of an early ethanol exposure on the development of retinofugal
topographical fine-tuning, eye-specific segregation and plasticity using the anterograde
transport of neuronal tracers in pigmented rats. We found that even low doses of ethanol
(1mg/kg ip) at postnatal days 4, 6 and 8 were able to induce alterations in the organization of
subcortical visual pathways. Retinocollicular axons/terminals displayed a shift towards the
lateral aspect of the superior colliculus with decreased density of terminals in the medial
aspect. This shift became more pronounced two weeks after the end of the treatment (P21),
suggesting that ethanol-treated rats presented a progressive distortion of topographical
representation in the SC. The data also showed that eye-specific segregation in the LGN was
altered by ethanol treatment with overlapping ipsi and contralateral terminal fields. Unilateral
retinal lesion at P14 showed that ethanol exposure significantly reduced the plasticity of
intact axons from the non-lesioned eye, a result that is consistent with a premature closure of
the critical period. Behavioral tests were performed in P90 to assess cognitive and motor
changes. In the Open Field test the ethanol-treated animal had a lower overall locomotion. In
turn, the Elevated Plus Maze showed that ethanol group remained less time in enclosed-arm.
The data suggest that ethanol induced a severe impact on organization of subcortical visual
pathways with an impairment of activity-dependent plasticity with negatives consequences in
sensory and motor systems.

Objectives: In the present work, we investigated if nucleotides regulate the growth of Müller
cells in scratched chick embryo retinal monolayer cultures.
Methods and results: When retinal cell cultures cultivated for 7 days were mechanically
scratched, a significant growth of glial cells at the empty area was detected between the 1st
and 3rd days after scratch and the area devoid of cells decreased by ~70%. Incubation of
cultures with 2.5 U/ml apyrase (APY) significantly attenuated the growth of glial cells and
while in control cultures the area free of cells represented 31.4 % of the original area after 3
days, in apyrase-treated cultures, it represented 74.8 % of the original area. Incubation of
cultures with the P2 antagonists, 100 µM Suramin and 40 µM Reactive Blue 2 (RB-2) and
also the P2X7 receptor antagonists, 10 µM BBG and 200 µM ATPo, significantly attenuated
the growth of glial cells, the area free of cells representing 68.2, 52.3, 76.2 and 88.4 % of the
original area, respectively, suggesting that nucleotide receptors are involved in the growth of
glial cells. UTPγS (10 µM) but not ADPβS (50 µM) antagonized apyrase-induced growth
inhibition in scratched cultures, suggesting the participation of UTP-sensitive receptors. In
apyrase-treated cultures, glial cytoplasm protrusions were smaller and unstable. Actin
filaments were less organized and alfa-tubulin-labeled microtubules were mainly parallel to
scratch. In contrast to control cultures, very few vinculin-labeled adhesion sites could be
noticed in these cultures. Treatment with 0,2 µM Src inhibitor-1, 10 µM LY294002, a
PI3K/Akt inhibitor and 10 µM PF573228, a FAK inhibitor, also decreased glial growth over
the scratch, the area free of cells representing 74.5, 51.4 and 64.5 % of the original area,
respectively. RB-2 decreased dissociated glial cell attachment to fibronectin-coated dishes
(50 µM RB-2 = 37.5 % inhibition and 100 µM RB-2 = 58.9 % inhibition of the control
values). Treatment with 100 µM RB-2 also decreased by 62.9 % the migration of purified
glial cells through transwell membranes. Treatment with the P2X7 receptor agonist, 100 µM
Bz-ATP also inhibited the adhesion of glial cells by 34.6 %. These data suggest that
nucleotide receptors regulated adhesion and migration of glial cells.
Conclusion: Mechanical scratch of retinal cell cultures induces growth of glial cells over the
empty area through a mechanism that is dependent on activation of nucleotide receptors,
SRC, PI3K and FAK.

CONSEQUÊNCIAS COMPORTAMENTAIS DA SEPARAÇÃO MATERNA EM UM
MODELO ANIMAL DO TRANSTORNO DE DÉFICIT DE ATENÇÃO E
HIPERATIVIDADE.

Estudos em animais indicam que aplicar situações de estresse a neonatos, como a separação
maternal, pode influenciar na plasticidade do sistema neural, provocando efeitos abrangentes
na atividade neuroendócrina, função imunológica e processo de neurotransmissão. Diversos
trabalhos mostram que a separação materna em ratos pode causar prejuízos cognitivos e
alterações locomotoras em animais adultos, características também observadas no transtorno
de déficit de atenção e hiperatividade (TDAH). Porém, existem poucos trabalhos que avaliam
os efeitos comportamentais da separação maternal no período da adolescência. O objetivo do
presente estudo foi investigar as consequências comportamentais da separação materna em
um modelo animal de TDAH.
Métodos: Foram utilizados 17 ratos machos das linhagens WKY e SHR (modelo animal do
TDAH), provenientes do biotério da Universidade Federal Fluminense (UFF). Cada linhagem
foi dividida em 2 grupos: controle e separação materna (n=3 a 6). Diariamente durante 3h,
animais entre P02 e P10 foram retirados das gaiolas e isolados em recipientes plásticos, com
temperatura de 37ºC. Durante o período da adolescência, P22 e P34, os animais foram
submetidos a 5 dias consecutivos de testes comportamentais. No primeiro dia, o animal foi
posicionado no centro do labirinto em cruz elevado (LCE) e filmado por 5 minutos. Nos
segundo e terceiro dias, cada animal foi colocado no campo aberto (CA) por 10 minutos. No
quarto, foram realizadas 2 sessões de 5 minutos, com intervalo de 30 minutos, para avaliar a
memória discriminativa no reconhecimento de objeto. No último dia, os animais foram
submetidos a 2 sessões de 5 minutos, com intervalo de 1 hora, ao labirinto em Y. Análise dos
vídeos realizada com Any-Maze e estatística através do StatSoft Statistica 8.
Resultados: No LCE, a separação maternal diminuiu a atividade motora seletivamente em
animais SHR. No CA, a separação materna aumentou a locomoção na linhagem WKY e
diminui na linhagem SHR. No reconhecimento de objetos, a separação materna causou
prejuízo na discriminação dos objetos. Os demais parâmetros não foram alterados pela
separação materna.
Conclusão: O estudo, ainda em fase preliminar, sugere que a separação materna piora o
desempenho cognitivo e locomotor em um modelo animal do TDAH durante a adolescência.

ALTERAÇÕES COMPORTAMENTAIS NA PROLE DE RATAS PREVIAMENTE
EXPOSTAS A IL-6: UM MODELO DE AUTISMO

O autismo é um distúrbio do neurodesenvolvimento que afeta menos de 2% da população,
ocorrendo mais em homens. É uma doença que afeta a interação social, a comunicação e os
padrões de comportamento, eventualmente ocasionando estereotipia. Recentemente, foi
mostrado que a interleucina 6 (IL-6) está relacionada com as causas desse transtorno. Ratos
com níveis de IL-6 elevados no cérebro apresentam habilidades cognitivas prejudicadas,
déficits de aprendizagem, traços de ansiedade anormal e diminuição da interação social.
Essas são consequências do prejuízo à plasticidade neural. O objetivo deste trabalho foi
observar as alterações comportamentais na prole de fêmeas tratadas com IL-6.
Ratas fêmeas, adultas, da linhagem Lister Hooded receberam uma única injeção de IL-6
recombinante (5μg/kg) no dia gestacional 12,5 (E12,5). O grupo controle recebeu o veículo
utilizado para dissolver a IL-6 (PBS). A prole obtida a partir dessas fêmeas foi submetida aos
testes comportamentais de campo aberto (CA) e labirinto em cruz elevado (LCE), para
avaliar alterações de ansiedade, cognição e motricidade.
No LCE, administração de IL-6 diminuiu o comportamento relacionado à ansiedade como
observado pelo aumento da porcentagem do número de entradas nos braços abertos. Em
relação ao percentual de tempo gasto nos braços abertos, estatisticamente não se observou
diferenças, assim como o número de entradas nos braços fechados. Em relação ao CA, no
primeiro dia observou-se uma diferença relacionada à locomoção total entre o grupo IL-6 e o
PBS, de forma que o grupo IL-6 apresentou menor locomoção. No segundo dia, a mesma
diferença foi observada. Não houve diferença de locomoção entre os dois dias. Já em relação
à locomoção central, não houve diferença entre os dois grupos.
A partir dos resultados preliminares obtidos é possível concluir que o modelo animal de
autismo utilizado no presente estudo apresenta mudança no perfil motor e no comportamento
relacionado à ansiedade. Esse modelo pode ser uma ferramenta útil para estudar as
características comportamentais e neuroquímicas observadas no autismo.

Efeitos da acupuntura em um modelo animal do Transtorno de Déficit de Atenção e
Hiperatividade

O Transtorno de Déficit de Atenção e Hiperatividade (TDAH) caracteriza-se pelos sintomas
de desatenção, hiperatividade e impulsividade, os quais parecem resultar de desequilíbrios
dopaminérgicos nos circuitos do estriado e córtex pré-frontal. Trabalhos anteriores
demonstraram que a acupuntura pode modular os níveis de dopamina extracelular no núcleo
accumbens. Contudo, há uma carência de estudos avaliando os efeitos da acupuntura em
modelos animais do TDAH. Ratos da linhagem SHR, quando comparados aos WKY, são
amplamente utilizados como um modelo animal do TDAH.
O objetivo deste trabalho foi investigar a atividade motora e o comportamento relacionado à
ansiedade em um modelo animal do TDAH. Para isso foram utilizados ratos adultos das
linhagens SHR e WKY. Ambas as linhagens foram separadas em três grupos: controle
(naïve), acuponto (HT7), não-acuponto (cauda). Os animais foram manipulados por um
minuto durante três dias consecutivos. No quarto dia, os animais foram contidos
manualmente e as agulhas foram inseridas nos grupos acuponto e não acuponto, utilizando
como estímulo o movimento manual de puxar/introduzir durante um minuto seguido de um
minuto sem estímulo. Este procedimento foi repetido por três vezes consecutivas. O grupo
controle foi contido durante o mesmo período de tempo e sem a inserção de agulha. Após 30
minutos do procedimento de acupuntura, os animais foram submetidos os testes
comportamentais. A atividade motora foi avaliada no teste do campo aberto durante 10
minutos. O comportamento relacionado à ansiedade foi avaliado no labirinto em cruz elevado
durante 5 minutos. O tratamento de acupuntura no HT7 diminuiu a locomoção seletivamente
em animais da linhagem WKY. Por outro lado, a estimulação do não-acuponto causou
aumento da locomoção apenas em animais WKY. A atividade motora dos animais SHR não
foi afetada por nenhum dos tratamentos. Além disso, o tratamento no acuponto HT7 diminuiu
o comportamento relacionado à ansiedade seletivamente em animais SHR. E o tratamento no
não-acuponto (cauda) aumentou o comportamento relacionado à ansiedade. Os resultados são
preliminares e indicam que a acupuntura deve ser realizada em pontos específicos para,
eventualmente, ser promissora para melhorar algumas características observadas no TDAH.

Avaliação antidepressiva da metformina em roedores

A depressão não possui etiologia definida, sendo o transtorno psiquiátrico mais prevalente em
todo o mundo. A atrofia hipocampal tem sido relatada em casos de depressão maior. O
tratamento crônico com antidepressivos clássicos pode induzir neurogênese e restaurar o
volume hipocampal (Nestler et al, 2012). Além disso, algumas evidências apontam para
alterações nos níveis glicêmicos em indivíduos com depressão (Park et al., 2012). Estudos
recentes mostram que o hipoglicemiante metformina pode induzir neurogênese no hipocampo
de roedores (Stein SC et al., 2005; Kahn BB et al.,2000). O objetivo deste trabalho foi
investigar os efeitos da metformina no comportamento relacionado à depressão em roedores.
Para isso, ratos da linhagem Lister Hooded e camundongos Swiss, ambos durante a idade
adulta, foram utilizados. Após 30 minutos da administração de metformina (300 mg/kg),
fluoxetina (20 mg/kg) ou veículo (NaCl 0,9%), pela via intraperitoneal (ip), os animais foram
submetidos aos testes comportamentais. O comportamento relacionado à depressão foi
avaliado no teste do nado forçado (TNF). Para ratos, o TNF foi realizado em dois dias. No
primeiro dia, os animais foram expostos ao TNF durante 15 minutos. No segundo dia, os
animais foram expostos ao TNF durante 5 minutos e o tempo de imobilidade foi registrado.
Para camundongos, o TNF foi realizado em apenas uma sessão de 6 minutos e o tempo de
imobilidade foi registrado. A temperatura da água, para ambas as espécies, foi ajustada a 25 ±
1 °C. O teste do campo aberto foi realizado em uma arena (60 cm x 60 cm x 60 cm) e foi
utilizado para avaliar a atividade motora dos animais. O tratamento com metformina ou
fluoxetina não alterou o tempo de imobilidade em ambas as espécies. Por outro lado, a
metformina diminuiu a locomoção seletivamente em camundongos. Os resultados mostram
que a metformina, na dose testada, não altera o comportamento relacionado à depressão. Mais
estudos, com diferentes doses e em um esquema de tratamento crônico, precisam ser
realizados para descartar o possível efeito antidepressivo do hipoglicemiante.

MICROGLIAL ACTIVATION IS NECESSARY FOR RETINOTECTAL
PLASTICITY INDUCED BY UNILATERAL MONOCULAR ENUCLEATION

Lesions in the central nervous system (CNS) can often induce structural reorganization within
intact circuits of the brain. In literature, several recent studies show advances in the
understanding of mechanisms of brain plasticity and the role of the immune system
activation. In the present study, we evaluated role of microglial activation in the sprouting of
intact axons following lesions of converging visual pathways (Serfaty et al 2005). Microglia
is a cell population with immune functions colonizing the CNS during early phases of
development. Their activation is dependent of calcineurin (CaN), a phosphatase also
associated with the process of synaptic stabilization/elimination (Trindade et al 2011).
Therefore, we evaluated the temporal course of microglial activation in the superior
colliculus following a contralateral monocular enucleation (ME) and the possible
involvement of microglial cells in the plastic reorganization of the uncrossed retinotectal
pathway from the intact eye. Lister Hooded rats were enucleated at PND 10 and submitted to
a chronic and/or acute systemic treatment with inhibitors of microglial activation: cyclosporin
A, (10 mg/kg, SC - chronic treatment/ 50mg/kg, SC - acute treatment), and minocycline (125
mg/kg, SC - acute treatment), a specific inhibitor of microglial activation. The use of
neuroanatomical tracers allowed us to evaluate the structural plasticity and
immunofluorescence together with western blot techniques allowed us to observe the
expression and morphology of microglial cells and its main transcription factor, NFAT.
Following a ME, Iba-1 immunoreactivity showed a progressive increase of microglial
activation in the contralateral SC after 24h, peaking at 72h after the lesion and its activation
correlated with CaN expression. Treatment with inhibitors of microglial activation reduced
the structural plasticity of intact uncrossed retinotectal axons and microglial activation as
seen by the decrease of Iba-1 immunoreactivity, 24h after the lesion. The data support the
hypothesis that microglial activation is a necessary step for the regulation of neuroplasticity
induced by lesions during early brain development. This study was approved by CEUA/UFF
