It’s been nearly 20 years since my late parents, Jay and Betty Van Andel, undertook the ambitious mission to create a world-class biomedical research and science education organization in Grand Rapids. It not only succeeded, but it is flourishing, thanks in large part to you – a community of people committed to improving the health and enhancing the lives of current and future generations.
There is no time to wait. Cancer alone kills some 570,000 Americans annually – the equivalent of a mid-sized city. The good news is that with your help, we can stop diseases like cancer and Parkinson’s.

Time, Treasure anD TalenT
George Sharpe Sr. is a self-made man. Growing up in Detroit, George understood the value of hard work and the importance of community. Starting as a car porter, he washed cars for a local dealership. His determination and sales ability served him well, and in 1982 he moved to West Michigan with his wife Linda, daughter Nicole and sons, Blair and George Jr., and purchased his first dealership. George’s two sons soon began working alongside their father, learning the business and the tools of the trade.
Blair and George Jr. now run a number of successful luxury car dealerships. “We learned early about the business and grew to love it,” said George Jr. “My dad made sure we knew what it meant to earn a dollar and the value of hard work,” said Blair.
When George Sr. was diagnosed with Parkinson’s disease, the brothers decided it was their responsibility, as businessmen and community leaders, to support research that would improve the lives of those living with the disease. Giving back and leveraging time, treasure and talent in the service of community is the Sharpe family credo.
“We believe in what we’re doing, and one of our corporate philosophies is that you are only as strong as your community,” said Blair. “Community and business are a symbiotic relationship.”
The Sharpe family has partnered with Van Andel Institute, collecting funds and chairing special events to benefit Parkinson’s disease research. The family was chosen by Van Andel Institute Board of Governors member Tony Asselta to chair the popular
Winterfest event that he led for more than eight years. “The Sharpes are dedicated individuals in the community and everything they do comes from the heart,” said Asselta. “Like the Sharpes, my father also had Parkinson’s, so I know how important it is to support Van Andel Institute’s research into this disease.” The Sharpes had the opportunity to meet researchers like Dr. Patrik Brundin, Head of the Laboratory of Translational Parkinson’s Disease Research at Van Andel Institute, who share their passion for what research can do to benefit the greater good. Blair believes that their family partnership with the Institute can have a lasting impact on the future of Parkinson’s disease.
“My father says, ‘We might not find a cure for Parkinson’s disease in my time, but hopefully my children and grandchildren won’t have to deal with this disease.’”

Parkinson’s disease affects more than 1 million people in the United States. The neurodegenerative disease impacts an individual’s motor functions, cognitive abilities and quality of life and often goes undetected until the physical symptoms become apparent in the patient. Currently, there is no cure for Parkinson’s disease.
“I saw my father battle Parkinson’s disease for 22 years, and I know how the disease can impact patients,” said Brundin. “I also know that Van Andel Institute is completely devoted to researching and solving the many issues presented by Parkinson’s disease, and we’re going to stay with it until we’re done.”

Trials initiative designed by The Cure Parkinson’s Trust

first time, the team showed that a misfolded protein
associated with Parkinson’s disease can travel from
neurons in the olfactory bulb – one of the areas first
impacted by the disease – to other regions of the brain.

an unlikely Team
Dolly Konwinski and Dr. Jeff MacKeigan are an unlikely team. Dolly is retired and in her early 80s, and MacKeigan is Associate Professor and Head of the Laboratory of Systems Biology at Van Andel Institute, but together they are a powerful force in the fight against cancer.
In grade school, Dolly played sandlot baseball pick-up games with the boys in her West Side Chicago neighborhood, and at the age of 15, Dolly went on to play for the post-war Women’s League in the 1940s and 1950s. Being one of the first women in professional sports was difficult, but the experience strengthened her character and resolve.
This resolve was tested years later when her husband Bob was diagnosed with cancer. After her husband’s battle with cancer, surgery and recovery, Dolly became increasingly aware of the prevalence of cancer. One day while watching her beloved Detroit Tigers, Dolly noticed something that would set her on an unforeseen path.
“I sat down at the beginning of a game and one player has a pink bat, and another guy in the on-deck circle has a pink bat… and I thought I’d get some pink baseball bats and get champion athletes to sign them, then I’d sell them and give the money to cancer research,” said Konwinski.
Dolly decided to leverage her recognition as a ballplayer to collect more than 40 professional athletes’ signatures on four bright pink bats. Sports luminaries such as Jack Nicklaus, Gale Sayers and Pete Rose all signed Dolly’s bats. Dolly reached out to Dr. Jeff MacKeigan, Associate Professor and Head, Laboratory of Systems Biology, and told him of her plans. MacKeigan invited Dolly to the Institute for a tour
and was struck by her passionate support of the work he was doing. “She had that personality where she wanted to see directly where her funds were going,” MacKeigan said.
“She got to visit the laboratory and wanted to help us buy a piece of equipment or support an experiment… she emphatically wanted to help.”
After Dolly’s visit with MacKeigan, she committed to donate all of the funds from her pink bats to the Institute in support of cancer research. In Dolly’s words, her efforts were a “labor of love.”
small DonaTions, Big impacT
It has been said that there is no such thing as a small donation, and that is especially true at Van Andel Institute. Because of the generous
endowment left by founders Jay and Betty Van Andel, 100% of every donation goes directly into the labs. At the Institute, there is a direct, personal connection between donors and Institute researchers.
Gifts of any size have a significant role to play in forwarding Van Andel Institute’s mission. MacKeigan and his team began the TSC (tuberous sclerosis complex) Pathway of Hope initiative in 2013 that hopes to develop improved treatments for the rare tumor disorder. This initiative was funded by private donors as well as the Michigan Strategic Fund through the Michigan Economic Development Corporation.
“All funds, no matter how large or small, are going to an important experiment or piece of equipment that helps push our research forward,” MacKeigan said.

key Discoveries
Breast Cancer Van Andel Institute scientists have shown the
interaction between MET and HER2 (ERBB2+) receptors
(two important genes related to cancer) is linked to
tumors resistant to targeted therapies. The discovery and
understanding of this relationship not only shed light
on some of the mechanisms behind breast cancer but
also may aid in the development of novel treatments.
Anxiety, Depression and Migraines Van Andel Institute scientists, along with colleagues in
China, discovered the structural blueprint that will aid
in the design of more selective activators of serotonin
receptors. Serotonin is a neurotransmitter, and its
receptors are important drug targets for conditions
such as anxiety, depression and migraines.
OsteoarthritisResearch initiated in Van Andel Institute’s Center for
Cancer and Cell Biology and continued with colleagues
in the Center for Skeletal Disease and Tumor Metastasis
has linked a deficiency of Mig-6, a gene associated with
joint stability, to osteoarthritis development and the
regulation of cartilage cells. This work could lead to
disease-modifying treatments for osteoarthritis, which
is the most common form of joint disease in humans.

Nathaniel is a spirited five-year-old who was referred to Dr. Matt Steensma, an Assistant Professor at Van Andel Institute’s Center for Skeletal Disease Research, who over-sees the Institute’s Laboratory of Musculoskeletal Oncology. Steensma is also an orthopedic oncological surgeon with Spectrum Health Medical Group who practices at Helen DeVos Children’s Hospital and maintains a position at Michigan State University College of Human Medicine. Following an MRI and biopsy, Steensma discovered Nathaniel had an aneurysmal bone cyst – an aggressive, bone-eating tumor. Although not cancerous, the tumor was damaging bone, causing pain and making it difficult for Nathaniel to walk. Because of the tumor’s location, surgery was not an option.
“This was one of the most severe cases of an aneurysmal bone cyst in a child,” Steensma said. “When these tumors occur on the spine, they can be very difficult to treat because of the anatomy that’s around them.”
Steensma scoured literature trying to find an effective treatment and discovered an FDA approved treatment for a related tumor. This therapy targets a protein called RANK ligand (RANKL), and he found significant evidence that aneurysmal bone cysts also contain the protein RANKL. He decided repurposing this therapy might benefit his patient.
With oversight by Dr. Deanna Mitchell, a pediatric oncologist at Helen DeVos Children’s Hospital, Nathaniel was placed on a monthly regimen that entailed a simple injection beneath the skin. In two months, his pain was almost completely gone, and he was able to walk without difficulty.
“Nathaniel has had a dramatic response with this drug, and it’s been sustained more than a year with minimal side effects,” Steensma said. “We are delighted. It was a much better treatment than surgery.”
In order to determine if this therapy would benefit other patients with similar tumors, Steensma had to return to his lab at Van Andel Institute.
“I was able to look at VAI’s banked samples of aneurysmal bone cysts and demonstrate that RANKL is expressed highly in these specific tumors, which suggests that other bone cysts like Nathaniel’s can be treated similarly,” Steensma said.
Steensma is proud that his clinical team at Helen DeVos Children’s Hospital and the research team at Van Andel Institute played a significant role in changing Nathaniel’s life, with the potential to benefit others around the world.

Prostate Cancer Van Andel Institute scientists are working to further
understand a key signaling pathway that may provide
more effective treatments for patients whose prostate
cancer has spread to their bones. Currently, there is
no curative treatment and few dependable palliative
options once the cancer moves to the skeleton.
Osteoarthritis Scientists in Van Andel Institute’s Center for Skeletal
Disease and Tumor Metastasis have shown that
a mutation of the Lrp6 gene can lead to severe
osteoarthritis and further clarifies the roles of the Lrp6
and Lrp5 genes in bone stability. By using a genetic
model, Van Andel Institute scientists were able to
ascertain a clear link between the genes and the cells
responsible for the formation of bone.

rewarDing research
Focusing on the patient is an everyday reality for Dr. Lena Brundin, Associate Professor in Van Andel Research Institute’s Center for Translational Medicine and Associate Professor at Michigan State University College of Human Medicine. Dr. Brundin was a practicing clinical psychiatrist in Sweden for almost 10 years, and this direct clinical work with patients inspired her to search for answers regarding mental health in her laboratory at Van Andel Institute.
“It’s an extremely rewarding way of working,” said Brundin. “I think it’s really fundamental to start with patients and build on that in the laboratory in order to have progress in science and develop good treatments.”
Brundin, who focuses on depression and suicidality, uses basic research and clinical practice to develop new therapies for psychiatric patients with a focus on depression and suicidality. She collects data from patients and uses that information to conduct tests in her Van Andel Institute laboratory in collaboration with Spectrum Health and Pine Rest Christian Mental Health Services.
Her team made exciting discoveries in 2013 that may lead to more effective treatments for patients living with severe mental health issues. Brundin also engaged in a study measuring inflammatory markers in cerebrospinal fluid, the fluid that surrounds the brain and spinal cord, of patients with Parkinson’s disease. The study found a significant association between inflammation and non-motor symptoms of Parkinson’s disease such as fatigue, depression and anxiety, which dramatically affect a patient’s quality of life.
“If inflammation is really the cause of some severe psychiatric symptoms, there are plenty of anti-inflammatory medications that are already approved for clinical use. We’re a couple steps ahead compared to inventing something completely new,” said Brundin.
Repurposing drugs that have already been approved by the FDA is a practice common in translational medicine that gives patients access to new, more effective treatments faster than developing new therapies. More than 14.8 million adults are affected by depression in the United States,
and the need for improved treatments has never been greater. Brundin’s collaborative research is at the forefront of translational medicine, and her focus remains on how discoveries can benefit patients.
“We’re trying hard to find new treatments against
depression,” said Brundin. “We are excited about the progress we are making so far.”

Exciting discoveries in translational medicine, made possible through institutional collaborations, may provide more effective treatments for millions of mental health patients.
Malignant Melanoma Using melanoma-based avatar models that mirror
human tumor progression, Van Andel Institute scientists
identified several acquired drug resistance mechanisms
for targeted cancer therapies. The group also identified
a combination drug approach that led to sustained
remissions in a validated pre-clinical system.
Pancreatic Cancer Van Andel Institute scientists have developed a new
method that will aid in the diagnosis of precancerous
cysts in the pancreas, which can develop into an
aggressive form of cancer. In two studies, the new
method demonstrated 93% and 91% accuracy, making
the test more reliable than current diagnostic methods.
Pancreatic cancer is the fourth leading cause of cancer
death in the U.S. and can spread quickly, making early
diagnosis vital for treatment.

When Jason Dunn, a fifth-grade teacher, first became involved with Van Andel Education Institute five years ago, he had no idea just how much it would influence his career and his classroom. Dunn, along with other West Michigan teachers, was invited to the Science Academy to hear about the program’s mission and learn how educators can play an instrumental role in inquiry-based science education, a model that encourages critical thinking, problem solving and collaboration.
“The Science Academy’s goal is to get students to think and act like scientists, and through this process you actually see them applying those skills, as opposed to other students who skim a textbook in search of an answer,” said Dunn.
Having successfully completed the three-year training program, Dunn now implements the model on a daily basis. Much like the work conducted by biomedical researchers at Van Andel Research Institute, the students in Dunn’s class address questions through a number of investigations that allow them to ask scientific questions, hypothesize answers and make their own discoveries.

a scienTisT of Tomorrow in The making
When Eric Nollet attended the annual West Michigan Regional Undergraduate Science Research Conference at Van Andel Institute, he was delighted to find a number of college students who shared his recently discovered passion for medical research. He listened to presentations and discussed findings with a number of peers throughout the day, but for Nollet, the most important presentation didn’t come until the end of the conference. It was a presentation that would fundamentally change the course of Nollet’s life.
Dr. Nick Duesbery, Associate Professor, Center for Cancer and Cell Biology, Head, Laboratory of Cancer and Developmental Cell Biology, spoke about using basic research to develop and test a type of protein-based drug that could kill blood vessels in a specific type of tumor. Although Duesbery did not think his presentation would alter someone’s life path, what he said that day had a profound impact on Nollet’s future.
“It was really inspiring to see someone who was doing basic research suddenly translate that to medicine,” said Nollet. “This presentation helped me realize how important basic research really was to discovering new types of treatments or therapies.” Nollet was overjoyed in 2010 when he was accepted into the exclusive Van Andel Institute Graduate School. “I loved the small class size,” says Nollet. “It’s more hands-on, and the lessons are driven by what you need to know.”
Van Andel Institute Graduate School teaches students how to conduct themselves in research laboratories with a focus on discussion and hands-on training led by world-renowned researchers in classes containing a maximum of four students. When students aren’t studying, they can be found in the research labs working alongside their mentors and peers.
Nollet has spent his time working in the lab run by Dr. Cindy Miranti, Associate Professor, Center for Cancer and Cell Biology, Head, Laboratory of Integrin Signaling and Tumorigenesis, for nearly four years, working on a single project revolving around prostate cancer that is resistant to standard treatment.
“I’m more of a critical thinker now because of the way classes are conducted, and we are held accountable for our work just like the researchers are. Although I look forward to continuing my career, it will be difficult to leave. I like it here,” said Nollet, reflecting on the past three years.

When a family member of an eighth-grade student at Duncan Lake Middle School was diagnosed with cancer in 2009, she presented the idea for a cancer walk – and the entire school
rallied behind her. The school decided to support cancer research at Van Andel Institute because of its 100% Hope® promise.
“We know that 100% of what we raise goes to research,” said Ryan Graham, Principal at Duncan Lake Middle School. “For our students, seeing items in the lab that Duncan Lake helped purchase and hearing
a researcher talk about his work is powerful.”
Duncan Lake’s Cancer Walk was planned by a leadership team of six students and implemented by an army of 60 student council volunteers. The entire school participated in a fundraising competition for six weeks leading up to the event, and the students managed all the fundraising activities. The students held bake sales, raffled off five-pound gummy bears and sold tickets to throw wet sponges at teachers.
The school currently has three students battling cancer, and most students have been touched by the disease through a friend or family member. “My grandpa had cancer,” explained eighth-grade organizer Jason Blunt. “Knowing that there are people who are working hard to try to cure cancer makes me feel good.”
For these students, fighting cancer is an expression of their support for cancer research and the joyful feeling of school spirit. “We take joy in fighting cancer; celebrate with us!” said Graham.
The power of purple
Purple Community, Van Andel Institute’s grassroots fundraising program, connects individuals who want to make a difference with the resources needed for action. Working together, individuals, businesses, schools and communities have the ability to influence the future of human health.
Duncan Lake Middle School students have changed the course of research at Van Andel Institute by raising more than $40,000, supporting the research of Dr. Nate Lanning, Postdoctoral Fellow in the Lab of Systems Biology.
One focus of Lanning’s research is to understand the metabolism of glioblastoma multiforme (GBM), the most common and aggressive malignant primary brain tumor found in humans. Thanks to the donations from Duncan Lake Middle School, a new piece of equipment was purchased in 2013. This equipment allows him to take more accurate measurements in his experiments and provides critical insights into the metabolism of GBM.
“I cannot tell you how fast the equipment has accelerated my research,” said Lanning. “It has supported not only my research but also the work of others researching breast cancer, melanoma and more at Van Andel Institute.”

a Tale of Two ciTies
More than 1.6 million people are diagnosed with cancer in the United States each year, but for those living with cancer, hope knows no geographical boundaries. The need for improved treatments and diagnostics is shared by everyone affected by this devastating illness. The goal of any cancer awareness campaign is to give a feeling of connectivity and community to patients, caregivers and their families. The campaign can also highlight those who are working to better understand cancer and develop improved therapies.
When Van Andel Institute partnered with the Detroit Red Wings to bring about Hockey Fights Cancer Night, a powerful message of hope was sent across the state. The event involved more than 20,000 people in support of cancer and disease research at Van Andel Institute and earned more than four million impressions. The famed Joe Louis Arena was enveloped with purple rally towels and Purple Community signage. Fans in attendance also had the opportunity to participate in awareness initiatives in remembrance of friends or loved ones who had been affected by cancer.
This night gave hope not only to the thousands of people at the game but also to the millions watching in the far-flung neighborhoods of Motor City, towns in Michigan and regions across the United States. This night let people know that when it comes to fighting cancer, Van Andel Institute and the Red Wings believe in the power of community, no matter where you call home. Tom Wilson, President and CEO of Olympia Entertainment, the organization in charge of all Red Wings operations, feels this connection to community is inherent in the Red Wings’ philosophy.
“Connecting with the community has always been a high priority for the Red Wings, especially when it comes to battling cancer and disease,” said Wilson. “We are honored to be able to team up with Van Andel Institute, a leader in biomedical research and education, and Purple Community
is a great way to engage our fans and raise funds for an outstanding cause.”
This idea of community connection is woven throughout Van Andel Institute’s philosophy of engagement, and each year organizations like the Detroit Red Wings decide to partner with the Institute and Purple Community to proactively support dynamic biomedical research and empower their communities in the fight against cancer and disease.

In addition to hosting company events, businesses support Purple Community through special products and offers.
Purple Community currently has twelve corporate cause marketing partners and is regularly adding new ones. Restaurants like Brann’s Steakhouse and Grill donate a percentage of sales to Purple Community; Lake Michigan Credit Union sells purple tribute badges in its branch locations; and AOL promotes Van Andel Institute on its homepage to help spread its mission.

Rebecca Wierda is an entrepreneurial fashionista, a woman with strong business acumen, drive and a passion for fine women’s clothing. When she took ownership of Leigh’s, a women’s specialty store and a 30-year-old retail institution in Grand Rapids, she didn’t just fill the racks; she built a new store. Today, Leigh’s is a thriving small business and one of the few locations in West Michigan where women can purchase high-end designer collections.
For eight years, Wierda and her team have been partnering with Van Andel Institute to organize Couture for a Cure, a fundraising event benefiting the Institute’s research and education initiatives. The event, first conceived by Carol Van Andel and Wierda, started small with only a few dozen guests gathering at Leigh’s. The event has since grown to host more than 500 guests at Van Andel Institute and has raised hundreds of thousands of dollars for the Institute’s programs.
“It’s a great showcase for the work being done at Van Andel Institute and a great opportunity for my business to partner with the Institute in support of cancer and disease research as well as science education,” said Wierda.
The dynamic businesswoman is proud that Leigh’s can play a role in supporting life-changing cancer and disease research.
“My husband lost his father to cancer and my mother is a cancer survivor, so I think it’s really special that we have this world-class research institute in our community and I love that we can bring this important work into focus,” said Wierda.
