macrophages• all these noted metabolic changes were counteracted by culture of tissue

samples with trametinib

Conclusions

• Dynamic 3D culture in bioreactor is suitable for pathogenic studies and fordrug testing in ECD

• The technology allowed us to define outcomes down-stream oncogenicmutations, and specifically to identify rewired metabolism as a peculiarfeature of ECD histiocytes.

• Our model can be further exploited to design new therapeutic strategies forECD and conceivably for other forms of histiocytosis.
