NIH/NIGMS GM069338 Lipid Maps Consortium (Ed Dennis PI) 9/01/03-8/31/2014 (NCE) Sphingolipid core (ca $300,000 per year) This “glue” grant will analyze all the lipids of macrophage cell lines and mouse macrophages under different states of activation. NIH 1R01GM076217 (Merrill, P.I.) 5/01/07-3/31/2016 Characterization of mammalian ceramide synthases Approx. direct costs requested: ($250,000 per year) The goal of this grant is to characterize the family of enzymes that are responsible for adding the N-acyl linked fatty acids of ceramide. This grant has identified the genes that code for ceramide synthases (CerS), studied regulation of expression of the mRNA/protein for the CerS, determined their biochemical properties, and is evaluating their roles in cell regulation and disease

Completed in the past 10 years: NIH R03 CA137812 (Merrill, P.I.) 7/01/09-6/30/2012 Chemoprevention via modulation of $100,000 total autophagy by sphingolipids The goal of this grant is to understand the regulation of this switch between “protective” and “lethal” autophagy in breast cancer cells via the induction of autophagy by dihydroceramide and accumulation of sphinganine. An underlying premise of this grant is that diet might affect dihydroceramide biosynthesis and induction of autophagy; therefore, the principles could be relevant to understanding the relationship between obesity and cancer. Memorial Sloan-Kettering Institute (subcontract under NCI grant to Gary Schwartz) 1/2005-07 Analysis of safingol pharmacokinetics in a phase I clinical [~$100,000] trial of safingol in solid tumors Dr. Schwartz is conducting one of the first human clinical trails of the sphingolipid analog safingol, and Dr. Merrill has developed an LC MS/MS assay to determine the blood concentrations of safingol and safingol metabolites in these patients NCI-U19-CA87525 (Merrill, P.I.) 7/01/00–6/30/07 Naturally occurring sphingolipids and sphingolipid analogs as mechanism-based anti-cancer agents [Approx $3,900,000]* The goal of this National Cooperative Drug Discovery Grant (NCDDG) is to develop new sphingolipid analogs for screening in in vitro and in vivo systems as potential cancer chemoprentive and chemotherapeutic agents. EmTech Biotechnology Development, Inc. 9/1/03-8/31/06 Cancer chemoprevention by sphingolipid-based pharmaceuticals $100,000 This grant tested the anti-cancer agent “Enigmol” for synergy or antagonism with other anti-cancer agents using human cancer cells in culture.
