    Summary

    This report describes l'esults of studies to determinethe intravenous dose of A ent 926 necessary to produce nauseaand vomiting inman. The experiments followed the methoprescribed by the Medical Research Laboratories.

    The agent was administered to adult male inmate volunteersat Holmesburg Prison, Philadelphia. Dosage started at 0.05microgram per kilogram (ug/kg) and was raised by 40% incrementsto a dose of 1.0 ug/kg. Nausea occurred at a dose of 0.74 ug/kgand emesis at 1. 0 ug/kg. These were the dominant symptoms.Mild sedation also occurred. There was no significant peripheraleffect or toxicity.

    Minimal Effcc ive Dose Determination for Agent 926

    INTRODUCTION

    Agent 926 was tested at Ho1mesburg Prison. "Philadel~determine the dose that would produce naqsea and vomiting. The_e>..-perimems started 5 fay 1969 and ended 23 Jul 1969. The study-was can uc e according to he general protocol draft 0(9 July 1964prepared by the Medical Research Laboratories (!v!RL) as modifiedoy :vIRL Research Plan 17,109 of 27 February 1969.

    METHOD

    Subjects were adult male inmate volunteers, 21to 34 years ofage. All subjects had at least 8 years of formal education, the abilityto perform at least 20 simple addition problems in 3 minutes, noevidence of psychosis or Significant neurosis according to the MinnesotaMultiphasic Personality Inventory (University of Minnesota, 1943), nohistory of arrest for violent acts and good health by clinical and laboratoryexamination. Twenty subjects were used for the 22 trials in the study.An interval longer than the prescribed 30-day minimum separated thetrials for the two subjects who partrcipated twice. SUbjects entered thetesting area the afternoon preceding the test day and remained thereuntil the fnllowing morning. Subjects were tested in individual rooms.Before the agent was administered, series of control measures wereobtained for pupil size; vital signs, including blood pressure and pulsesupine and after standing; and performance on the Numerical FacilityTest (NF) of the Hogg Foundation for Mental Health, 1956.

    The agent was injected in ravenously on the morning of the testday in a concentration of 5, 10or 20 mg. per milliliter. Doses startedat O . 05 microgram per kilogram (ug/kg) and were raised by 40%increments, according 0 the protocol, up to doses of 1 . 0 ug/kg.Inaccord ...ith safety precautions specified by MRL, no morefuan twosubjects were tested initially at each dose level. Increments were madeonly after laboratory studies at specified intervals indicated absence oftoxicity.

    Naus a was first reported between two and three minutes afteriljccnon for most of the affected subjects, and one"subject, after 1. 0 ug/kg,.~~ote -:8 ..,ymptom as early as one minute forty-five seconds. In thissmall series, however , the average onset time did not occur earlier forthe 1 lgher dos ge level. The period of nausea for subjects who did notvom"t ended bout 20 minutes after injection. Among the three suchsubjects the symptom persisted longer for the one man at 1. 0 ug/kg thanfor the two at O . ug/kg. Thus, the duration of nausea, at least whenpresent wi ho t vomiting, may be greater with incr eastng dosage.Vorni "ng se led to relieve the nausea and in such subjects the symptomended earlier, at approximately 10 minutes after injection.

He had originally been kept recumbent,but he clid vomit almost immediately after standing. Theoth.er two subjects,wl 0were not recumbent, vomited about one minute after reporting nausea.Sta ing alw ys aggravated nausea or precipitated vomiting. The onesubject who id not vomit after 1. 0 ug/kg had been allowed to remainrecumbent throughou the experiment. Vomiting or wretching usuallylasted two or three minutes.

    effective antidote, :lIlditdid pl"oduce some drowsiness. the prompt i~rove-~ ~ment of nausea o1ter vomi 'ng among untreated subjects makes it difficult toassess the effect of Tigan in this xperiment.

    Sedation was the other consistent effect, but it was mild and muchless Significant than the nausea and vomiting. This symptom was notedearlier when subjects were recumbent and, in these instances, occurredt approxim tely the same time as the nausea. The two subjects who werenot recumbent did not report sedation until after they had vomited and nauseawas deer-cast g. It is probable that these strong and unpleasant symptomsobscured the milder sedative effect.

    The speech of most of the affected s bjects was softened in_intenstty but remamed clear. This may have resulted from a combinati.onof seda 'on and disinterest m speaking during unpleasant symptoms.orne subjec s appeared mildly unsteady on Walking, and this also may havebe n a mani estation of sedation or marked nausea. A few subjectsEepolted a sensation of !!highness,. If as if they were mildly intoxicated orad received a narcotic. One subJect mentioned this atter fifteen seconds:'.:..:.:he val"d"ty or his repor is questionable. Generally this symptomace 'red C oser in time to the sedation.
