Alles fing damit an, dass ihre Großmutter ihr, als sie 12 Jahre alt war, zehn Pfund gab damit sich Amy ein paar Süßigkeiten kaufen konnte doch anstatt sich Süßigkeiten zu kaufen, kaufte sie sich die CD „The Man Who“ von Travis. Durch diese CD wollte sie selbst Musik spielen und zum Glück lagen in ihrem Elternhaus mehrere Gitarren herum, die ihr Vater aus seiner Jugendzeit verwahrte.
Sie nahm keinen Unterricht sondern lernte das Gitarre spielen dadurch das sie die Lieder die im Radio gespielt wurden einfach nachspielte und wenn sie Akkorde nicht kannte recherchierte sie diese im Internet. So machen das zwar auch Hobbymusiker aber bei ihr hatte schon fast ihre Karriere damit begonnen.
Sie kaufte sich nun noch viele weitere CDs um die Einzelheiten der Musik kennen zu lernen. Nachdem ihre Freunde nach einem Auftritt von Pete Doherty die Gitarre herumgehen ließen und jeder Lieder spielte die ihm gerade einfielen wusste sie dass sie Musikerin werden möchte.

Im Juli 2007 erreichte Amy Macdonald mit dem Titel Mr. Rock & Roll Platz 12 der UK top 40.
Im Herbst 2007 begleitete sie Paul Weller auf seiner Deutschland-Tournee.
Mit ihrem Album „This Is the Life“ erreichte sie Anfang 2008 Platz 1 der UK-Chats und erhielt Zweifach-Platin-Status.
Die gleichnamige Single verkaufte sich in ganz Europa mehr als 1,2 Mio. mal.
Die Aufnahmen für das zweite Album A Curious Thing fanden im Black Barn Studio von Paul Weller statt. Das Album erschien am 12. März 2010, das Lied „Don’t Tell Me That It’s Over“ wurde bereits Ende Februar veröffentlicht. Dieses Album erreichte Platz 1 der deutschen, österreichischen, schweizerischen und europäischen Album Charts. Dazu erreichte es noch Chart eintritt Gold-Status. In Deutschland und der Schweiz erhielt das Album Platin.
