Lighting techniquesThe success of an image analysis measurement system is 70%dependent on the lighting. The microscopes are often equippedwith several lighting techniques.
Bright Field
Bright Field lighting is the simplest observation technique. The light is transmitted directly viathe preparation or reflected by the sample.
Dark field
It is an oblique illumination technique used afterpart of the light has been stopped by an opaque fil-ter (e.g. Membrane between two slides).
Polarization
Polarization uses two polarizers on bothsides of the sample that orientates the lightaccording to the observed structures.
Interferential Contrast (D.I.C)
Phase contrast
The light passing through 2 rings situated in thelens and the condenser illuminates the object’scontour.
The interferential contrast, also calledNormarski, is obtained via a polarized lightguided by a prism.
Fluorescence
The ultraviolet rays stimulate certain molecules in the samples that emit energy resulting in a visiblewavelength passing through a filter. This low-level light technique requires signal intensification viaimage accumulation.
