I hope that all of you are enjoying your summer spending some quality time with your family and friends. Taking time to enjoy some memorable times going to the beach or camping in the mountains will be ones to last a lifetime. It just doesn’t seem possible but August has approached us and before you know it the 2017 National APHON Conference will be here. I am hoping that many of you will be able to capitalize on the opportunity to attend the conference being held at the Palm Spring’s Convention Center in Palm Springs, California on August 17-19, 2017. The theme for this year’s conference is “Expanding Horizons: Innovative Approaches in Care” and the education opportunities and networking are going to be something that you do not want to miss! Our chapter will be a co-sponsor of the Pool Party Networking Event on Friday, August 18th which will be a nice way to spend the evening meeting up with friends and colleagues. We have also donated a membership renewal for the prize drawing that takes place in the conference exhibit hall. I hope to see many of our SCAPHON members at the conference!
The exciting news to share with all of you is that our newsletter-The SCOOP will now become electronic and be available on our website for viewing. According to our recent conference evaluations, 68% of the participants preferred to receive it electronically. You will be able to download and print the document if you prefer but we will no longer be mailing the document. You will also be able to register and pay on line for the quarterly education meetings by accessing our website. If you want to print the registration form and mail your check you are welcome to do so. Another concern expressed on our conference evaluations-not knowing about the quarterly education dinners in a timely manner to help with work schedules. So please pencil in the following dates for our upcoming meetings: September 19, 2017, December 12, 2017 and March 13, 2018. They will be at the Claim Jumper in Fountain Valley.
We had a very successful conference at the Hyatt Regency, Mission Bay in San Diego on May 11-12, 2017. The speakers were well received and truly represented our conference theme Mission: Care, Cure, Connect. It was so wonderful to see so many nurses in attendance and I hope you were able to learn new things to bring back to your institutions as well as enjoyed visiting with exhibitors and networking with your colleagues. The good news is that we were able to secure the Hyatt Regency, Mission Bay for next year! So please mark your calendars for next year’s conference that will take place May 10-11, 2018.
I hope you will also join us for our September Quarterly Education meeting at the Claim Jumper on September 19, 2017. Shire will be sponsoring the education program and I am sure you will have a great time networking as well as enjoying the planned presentation. You should be receiving an email blast to remind you soon!
So I hope I will see you in Palm Springs at the 41st Annual APHON Conference as well
as our next Quarterly Education Event in September! I encourage all of you to get involved in
SCAPHON! Remember: Today is Your Day! Your Mountain is Waiting, So ……Get on Your

Targeted Immunotherapy: Testing a Novel Small Molecule Checkpoint
Kinase Inhibitor, Prexasertib, in Patients with Recurrent or Refractory Solid
Tumors, including CNS Tumors
Prexasertib (LY2606369) is a LY2606368 is a novel, small molecule checkpoint kinase
inhibitor, dually active against Checkpoint Kinase Proteins 1 and 2. Checkpoint kinases
are protein kinases that are involved in cell cycle control. Two checkpoint kinase
subtypes have been identified-CHK1 and CHK2.
CHK1 and CHK2 are key proteins to elicit cell-cycle blocks. In response to DNA damage
or replication stress, cells initiate a DNA damage response (DDR) that activates cell cycle
checkpoints to halt progress through the cell cycle to allow DNA repair. By inhibiting
CHK1, cancer cells lose the ability to repair damaged DNA which allows
chemotherapeutic agents to work more effectively.
This drug has been tested in adult phase I trials in women with breast or ovarian cancer as
well as patients with advanced solid tumors and has been well tolerated. The antitumor
activity of LY2606368 has been evaluated in xenograft models which indicated an
objective response in neuroblastoma, rhabdomyosarcoma, Ewing sarcoma, and
desmoplastic small round cell tumors. There have been no previous pediatric studies with
LY2606368. The agent is being provided for this study by Eli Lilly Company for use in
the Children’s Oncology Group Study, ADVL1515.
How is the Drug Administered to the Patient?
Prexasertib is given as an intravenous (IV) infusion over 60 minutes on Days 1 and 15
of a 28 day cycle. The starting dose for this pediatric phase 1 trial, ADVL1515, will be
80mg/m2. Patients will continue to receive up to 13 cycles of therapy unless the patient
has disease progression or unacceptable dose limiting toxicity. This agent is
incompatible with normal saline or lactated Ringer’s. Infuse the agent using 0.22
micron protein sparing filter and flush the line with Dextrose after the infusion is
complete.

Common Side Effects of Drug:
Neutropenia/Febrile Neutropenia
Anemia, thrombocytopenia
Fatigue
Diarrhea, N & V
Anorexia
Headache
QTc interval prolongation
Possible infusion reactions (fever, tachycardia, rash, hypotension)
Special Considerations and Monitoring Required:
▪ Obtain baseline vital signs immediately prior to initiation of infusion and at end of
infusion. Important to monitor for possible signs and symptoms of infusion
reactions (fever, tachycardia, rash, and hypotension) during the administration
and for 30 minutes post infusion. No pre-meds are required with the first
infusion. If patient should experience an infusion reaction, subsequent infusions
should include the use of Benadryl and/or other pre-medications at the discretion of
the physician. The infusion rate may also be decreased by 50%.
▪ Monitor CBC with diff and platelet counts for neutropenia and thrombocytopenia.
▪ Monitor electrolytes for abnormal levels.
▪ Patients should avoid concomitant medication known or suspected to prolong QTc
interval.
▪ EKG’s are done at baseline, then prior to and one hour after dose of LY2606368
for the first 2 cycles of therapy only and then ordered by the physician if clinically
indicated.
▪ Monitor patient for GI symptoms such as diarrhea. If patient has nausea and
vomiting, Zofran prior to infusion may also be helpful.
▪ Based on the light absorbing properties of LY2606368, patients should be advised to
avoid the use of tanning beds and if long exposure to sunlight is expected, to use
sunscreen and wear sunglasses.
