The following safety notes will help to avoid possible dangers when using the pHpO Module and give advice regarding the correct behavior to avoid dangerous situations. Special dangers when working with the pHpO Module are commented on in particular in the corresponding chapters of this manual.
Operation only by qualified personnel! The pHpO Module has been developed for the use in a laboratory. We assume, that the user is familiar with the necessary safety instructions as well as the op-erating of electrical equipment and will act accordingly.
Not accredited for the use in medical diagnostics or therapy! The pHpO Module was not developed for the use in medical diagnostics or ther-apy and is not to be used in such applications. It is not a medical device.
Do not open device! Danger of electric shock! The housing of the device is not to be opened by the user. In the inside of the housing there are no parts that have to be exchanged or adjusted by the user.
Do not use the device with damaged power cable or power plug!Danger of electrical shock! Replace damaged parts only with original parts of the manufacturer.
Service and Repair Work Service and repair work, beyond the maintenance work described in the user manual are only to be performed by DASGIP’s service technicians or technicians authorized by DASGIP.

Take note of user manuals and documentations of other manufactur-ers! Please heed to the instructions and safety notes of all devices included in the deliverable, especially the sensor manufacturer’s instructions.
Ambient Conditions During set-up and operation of the pHpO Module the necessary ambience condi-tions have always to be met (compare Chapter 4.3). If the device is to be operated at high ambience temperature and/or high humid-ity, the suitability of the device has to be discussed with DASGIP first.
The device is not protected against spray water!
Electromagnetic Radiation Strong electromagnetic radiation, e.g. from cell phones, might affect the meas-urement signals.
Reconsignment for Repair or Return You have to notify us of a reconsignment. If you fail to do so we will refuse acceptance. Furthermore, you have to declare, that the consignment is free from any decontaminations. Please read and heed to the information regarding the Decontamination Declaration at the end of this manual and complete the Decontamination Declaration carefully. Please insure, that the Decontamination Declaration reaches DASGIP 10 days prior to the delivery of the consignment.
For the reconsignment the original packaging should be used. Please heed to the instruction sheet How to Pack DASGIP-Modules at the end of the user manual!
If the original packaging is not available, the client is responsible for proper and suitable packaging. Please do not hesitate to inquire at DASGIP to have original packaging material sent to you.

2 Introduction
This manual is for all the product variants of the parallel sensor module pHpO. The variants only differ in the number and combination of the measurement channels (e.g. pH4pO4, pH8pO8, etc.). The manual describes the installation and appropriation of the module as a stand-alone device or as part of a DASGIP par-allel cultivation system cellferm-pro® or fedbatch-pro®. Furthermore, it contains important information and Tipps regarding the work with the sensor module.
The pHpO Module support standard pH and pO2 sensors. The temperature meas-urement is done using the NTC senors integrated in the pO2 sensors. Separate inlets for Pt100 handheld sensors facilitate an automatic temperature compensa-tion during measurement and calibration. The individual measurement channels of the device are separated galvanicly from each other; they cannot affect or damage each other.
With the sensor module pHpO you can monitor online e.g. the pH and pO2 val-ues in several parallel reactors or culture vessels. Different combinations of pH ,pO2 and temperature measurement offer various possibilities of the parallel monitoring of process values.
All sensor module variants can be operated as stand-alone devices or connected with a PC. For the control and monitoring of DASGIP devices the software DAS-GIP EasyAccess is availabe. In addition, the devices can be integrated into a DASGIP parallel cultivation system (e.g. cellferm-pro® or fedbatch-pro®).

Using the Software DASGIP EasyAccess on the CD included in the delivery, the DASGIP pHpO Sensor Modules can be controlled as stand-alone devices via a PC.
DASGIP EasyAccess is user interface based on MS-Internet Explorer, facilitating the control and monitoring of all important functions of the DASGIP devices. Via a simple web-page the set point values and process values can be accessed. DASGIP EasyAccess also offers effective support regarding process data logging.

Operating the web-pages is very easy. Extended training is not needed. DAS-GIP EasyAccess offers effective support in everyday laboratory tasks.
Detailed instructions regarding the work with DASGIP EasyAccess can be found in the DASGIP EasyAccess User Manual. The integration of the module into other applications using the DASGIP EasyAccess Driver Software is also described and examples are given.

At the back of the device, the power supply connector unit, communication con-nections, serial number and various venting slots can be found. Further informa-tion regarding the individual elements are listed in the Table further down.

4 Serial Number The serial number of the device should be stated in every enquiry with the manufacturer.
5 Communication Connections
RS232 and RS485 communication connections are explained in Chapter 3.2.1.
6 Venting Slots The venting slots are needed for heat dissipation and are not to be covered (Chapter 4.3).

4 Commissioning
Unpacking
The sensor Module pHpO device is delivered completely wrapped and should be taken out of the box following the Packaging Instructions for DASGIP Modules at the end of this manual. Whenever possible, the device should be unpacked at the location of installation.
Onword Transport
For the transport to another location or shipping for servicing, the original pack-aging should be used. This is the only way to insure the device being sufficiently protected. Check the Re-Consignment Information and the Packaging Instruc-tions for DASGIP Modules for additional advice (at the end of this manual).

To insure undisrupted operation of the pHpO Module you should respect the limits regarding the ambient conditions listed in the Table further down and the following notes regarding the set-up location.
The set-up location should be level and appropriate for the weight of the device (compare Chapter 6.1).
DASGIP Modules can be stacked. A maximum number of three DASGIP Modules are to be stacked, otherwise the stack becomes unstable. Please consider the resulting total weight.
The ambience of the pHpO Module has to be clean, dry and largely dust free. The limits for ambience conditions have to be complied with during transport, storage and operation.
When setting up the device it has to be insured, that sufficient convection can develop both on the front and back of the device, otherwise the developing heat cannot escape.
Do not cover the venting slots!

The sensors have to be connected to the corresponding sockets via the sensor cables and screwed tight.
Please heed to the input connection values regarding the connection of the sen-sors (Chapter 3.1).
The pO2 sensors have to be polarized for 2 – 6h prior to operation.
In the case of the device being operated in a DTP network, connect the port “DTP in” either with a serial interface of the process control computer (usually COM1) or with the port “DTP out” of another communication participant.

If two devices of the same type are to be operated in a DTP network, the ad-dresses have to be adapted e.g. according to the following table.
Changing the DTP address can be accomplished either using the configuration menu of the device or the configuration software DASGIP DTP Insight.
The corresponding device addresses for the operation of the sensor module as a part of a fedbatch-pro® or cellferm-pro® system can be taken from the following Table:

Please insure, that the calibrated sensors are connected to their corresponding sockets. If the sockets are interchanged, the calibration has to be run again for the changed slot.
For autoclaving the sensors can be disconnected from the sensor module. Please insure the correct allocation of the sensors to their slots.
Polarisation
Prior to operation, the pO2 sensors need a polarization time of 2 – 6h.
Connect the sensor to the corresponding socket and switch on the sensor mod-ule. Polarized sensors may be disconnected briefly from the sensor module. After a disconnection time an equally long polarization time should be kept.
Please heed to the sensor manufacturer’s notes.

Inspection of the pO2 Sensors
Prior to every calibration and using the pO2 sensors, you have to thoroughly in-spect the sensors:
1. Inspect the sensor membrane for visible signs of damages or soiling.
2. Clean the soiled membranes by wiping them with a soft, damp tissue.
3. Optional:Renew the electrolyte in regular intervals (compare instructions below).
4. Additionally, check, whether all connections are free of crystallizations, andcheck cables for bends, embrittlement and damages.
Please heed also to the instructions given in the pO2 sensor’s operating manual.
Electrolyte Change
Depending on mode and duration of use, the electrolyte has to be exchanged in regular intervals. At least every 6 months. In the following paragraph the electro-lyte exchange is described for standard Mettler Toledo sensors.
Attention! The 02 electrolyte is strongly alkaline. Please wear appropriate protective clothing – especially gloves. The liquid will certainly escape the membrane body and come into contact with your hands when opening the membrane body. Avoid contact with skin and mucosas, especially the eyes. Rinse contaminated areas thoroughly with water.
1. Unscrew cap sleeve from sensor body.
2. Carefully pull sleeve off sensor.
3. Remove membrane body from cap sleeve.
4. If body is tight fitted, eject by pushing it with flat finger tip.
5. Rinse interior body with DI water and carefully dap it dry with a paper tis-sue.
6. Examine O-rings and trapezoidal seal visually for mechanical defects.
7. Half-fill membrane body with O2 electrolyte.
8. Remove air bubbles by carefully tapping membrane body.
9. Slip membrane body over interior body while holding sensor in vertical posi-tion tip down.
10. Remove excess electrolyte with a paper tissue.
11. Carefully slip the clean and dry cap sleeve over fitted membrane body andscrew tight.

After each exchange of electrolyte or membrane body, sensor has to be re-polarized and recalibrated due to change of setpoint and slope.
The sensor has to completely polarize (6-8h). Afterwards, carry out a calibration!
Please heed also to the manufacturer’s notes in the corresponding documenta-tion for the pO2 sensors.

5 Maintenance and Trouble-shooting
Maintenance Intervals
The devices are tested by the manufacturer and are for the most part mainte-nance-free. To insure a fault-free operation we recommend to send the Module to us for a calibration service every one to two years.
In regular intervals, the internal buffer battery of the device should be renewed. Depending on the number of times the module was used this should be done every one to three years.
Please do not hesitate to contact the DASGIP service if you experience any errors or faults you cannot repair using the information given in the following Chapter or the user manual.
Cleaning
Superficial soiling can be removed using a damp cloth. Depending on the type of soiling, a mild detergent can be used. Remove residual acid or base remnants with clear water.
Please do not use scoring agents or corrosive detergents!
The device is not protected against spray!

8 How can the device be integrated into other systems?
Answer
Via the digital communication interface, all parameters, proc-ess values and set point values can be accessed. All DASGIP devices use the communication protocol DTP. For the easy integration of the device into Windows-PC appli-cations, a library is provided by DASGIP EasyAccess. The cor-responding driver can be found on the CD that was part of the delivery. An integration into other control or operating systems can be implementing using the protocol. Please inquire with DASGIP for the protocol specifications. An integration into other control or system software can be done by implementing the protocol. Please contact DASGIP for the protocol specifications. Due to the multitude of parame-ters, set point and process values in the device, no analogue interfaces are provided. The AUX port provides floating relay contacts for the indication of cumulate alarms.

The lines RXD, TXD,GND, RTS, CTS of the ports DTP-in and DTP-out are directly interconnected.
The maximal permissible load of the relay contacts is 30 VDC/1A.
In undisturbed state, the pins 1 and 6 are interconnected (break contact/ nor-mally closed). In case of an error, the pins 1 and 9 are connecting (working con-tact/ normally open).

The RS 485 communication ports are isolated galvanically from the remaining device electronics.
The connector pin assignment is according to the specifications "Modbus Over Serial Line Specification and Implementation Guide Version 1.0", Chapter 3.5.1. The lines B/D1, A/DO, Common of the two RJ45 port are directly interconnected.
The ends of the RS485 network have to be terminated using an appropriate re-sistance / condenser serial connection. Typical values are 1nF/16V and 120 Ohm.
The +5V emergency power at Pin 7 can be used for the polarization of a RS485 network. For polarization, B/D1 is connected to +5V and A/D0 to Common using in each case a resistance of 560 Ohm. The inner resistance of the voltage source +5V is 100 Ohm.

In order to provide maximum protection for our employees, associates, and the environment, please pay attention to the consecutively described arrangements for sending equipment to the DASGIP as well as for the on-site service:
DASGIP requires complete disclosure of biohazard and/or toxicity information on materials contacting or processed in our equipment. In the case of rental equipment, DASGIP will require this information before the rental equipment is shipped to the customer site.
Equipment returned to DASGIP must be free of substances that are bio-hazardous or any substances that are not bio-hazardous but may require special handling precautions. The cleaning and decontamination procedure must be completely documented (Page 1 of this form).
For equipment returned to DASGIP for service or any other reason please fill out the form (Page 1) completely and pre-send it via fax or email to DASGIP and attach the original to the outside of the package. Equipment which was sent to DASGIP without the completed form will not be handled. If the form is missing or contaminations are visible and no declaration is subsequently handed in, the goods will be returned at the sender’s costs.
This form must also be completed before any on-site service, whether covered under warranty or not!
Based on the information provided, each material should be assigned to one of the four Biosafety Levels as defined in the European Directive 2000/54/EG. The use of rental equipment is restricted to materials assigned to Biosafety Level 1. The use of materials assigned to a higher Biosafety Level needs prior written agreement by DASGIP.
Substances that are not bio-hazardous but may require special handling precautions must also be listed. Information should include name of chemical, chemical formula and hazard rating. Examples of these substances include, but are not limited to, acids, bases, radioactives and flammables.
Material Safety Data Sheets (MSDS) for any of the above items must be included with this form.
Cleaning and Decontamination
The cleaning and decontamination protocol described below is understood as a minimal procedure for cleaning and decontamination of equipment in contact with material appending to Biosafety Level 1.
Any substances that are bio-hazardous or any substances that are not bio-hazardous but may require special handling precautions must be removed, neutralized or inactivated in an appropriate manner. Check your in-house standard operating procedures concerning cleaning and decontamination to minimize any risk.
Please be aware, the undersigned is solely responsible and liable that all send parts, components and modules are safe for shipping and handling.
In the case that you are using alternative cleaning agents, please check chemical compatibility charts for all surface materials, in particular, but are not limited to, with components made out of acrylic glass. If in doubt, please check with DASGIP for advise.

Protocol (Applicable for Biosafety Level 1 only):
A. Control System Components, Modules, Cables, Gas Tubing, Incubators, Racks and Waterbath:
• Control system components include, but are not limited to, process computer, monitor, mouse, keyboard, uninterruptiblepower supply (UPS), balances, magnetic stirrer control box or immersion thermostats.
• Modules include, but are not limited to, sensor module, multi pump module, gas mixture module, one or multi positionmagnetic stir plates.
• Cables include, but are not limited to, power cords, extension cords, sensor cables (pH and pO2), DTP control cables,sensor module control cables or multi position power strips.
• Incubators include, but are not limited to, dry incubator, UNITRON-pro incubator shaker or shaking tables.
• Gas tubing include, but are not limited to, gas tubing, push-in connectors or stainless steel inline filter.
• Racks and waterbath include, but are not limited to, acrylic racks incl. shelves, acrylic stands or acrylic or stainless steelwaterbath.
Wipe down all surfaces, if applicable inside and outside, with 80% EtOH.
B. Glass Ware, Stainless Steel Parts, PTFE Parts, Caps and Fittings, Pump and Feed Tubing:
• Glass ware includes, but is not limited to, glass body of culture vessels, glass body of reservoir bottles, glass body ofharvesting bottles, glass ball agitators or spargers with porous glass frits.
• Stainless steel and PTFE parts include, but are not limited to, stainless steel flanges, stainless steel pipes, stainless steelspargers, heating and cooling coils of immersion thermostat, PTFE fittings or PTFE head gears, paddle type agitator.
• Caps and fittings include, but are not limited to, screw caps, screw caps with port, stainless steel caps, stainless steelcaps with port, tube fittings, Luer and Luer-lock fittings or compression fittings.
• Pump and feed tubing include, but are not limited to, PTFE feed lines, Pharmed tubing or Maprene tubing.
Soak in 0.2M NaOH for 1 hour followed by a 3x deionized water rinse (if applicable inside and outside). Autoclave at 121 °C for 30 min. in sterilization pouches indicating the exposure to sterilization conditions and ship sealed pouches back.
C. Sensors:
• Sensors include, but are not limited to, pH sensors, pO2 sensors or temperature sensors.
Soak sensor tip in 0.2M NaOH for 1 hour followed by a 3x deionized water rinse (if applicable inside and outside). Autoclave sensor, if applicable (e.g. an exception would be a hand held temperature sensor with permanent attached cable), at 121 °C for 30 min. in sterilization pouches indicating the exposure to sterilization conditions and ship back sealed pouches.
D. Not Returnable Parts: Parts not to be returned unless unused or otherwise advised by DASGIP include, but are not limited to, venting filters, gas in- and outlet filter (with the exception of the stainless steel inline gas filters), silicone tubing, silicone washers, silicone septa, silicone gaskets, rubber seals, sampling valves, or one way valves or cannulas.
