BIOGRAPHICAL INFORMATIONSenior Scientist, Infectious Diseases Division and Head, Immunology Lab, icddr,b

Dr Qadri is the Senior Scientist, Infectious Diseases Division at icddr,b in the planning and implementation and setting up of research priorities in the field of vaccines and infectious diseases in this Centre. Also heads a large Immunology Unit composed of scientists, researchers, postgraduate students working in the laboratory, clinical and field sites for carrying out immunological, molecular biological and high throughput genomic and proteomic studies on natural infections and vaccine related responses. As leader in the Immunology Unit, research is carried out with scientists working in the laboratory, clinical and epidemiologal aspects of infectious diseases. The work is led by 8 upcoming scientists as well as 10 clinicians, 15 junior researchers, 10 MSc and 6 PhD level students as well as postdoctoral level fellows working in sandwich programs with international and national institutions. In addition over 150 staff are directly involved in supporting laboratory, clinical and field related activities in projects ongoing in the Unit.

Major interests and original scientific activities

Dr Qadri has made significant contributions to international scientific research, particularly in the field of infectious diseases with emphasis on infections which are very common in children in developing countries. Her work includes basic and applied immunology of infectious diseases but also clinical and large field based studies on enteric vaccines. Major contribution over the years has been made in microbiology and immunology of infectious diseases. Special interests are infections caused by V. cholerae, ETEC, Salmonella spp and Helicobacter pylori. The results have produced an impact in the field of enteric diseases specifically in the area of immunological and genetics and genomic mechanisms, diagnostics and vaccine development.

Over the last 5 years she has been leading the largest field studies in over 400,000 participants of an oral cholera vaccine in an urban slum in Bangladesh so as to pave the way forward for introducing cholera vaccine in high risk people in the country and in the near future in the region and globally. This has been a very important achievement in her career for providing evidence of the success of affordable vaccines that work effectively in the real life situation. The multi-directional work with the Government of Bangladesh and International experts has resulted in providing evidence and also supporting the transfer of technology of cholera vaccine to Bangladesh and its production locally which will hopefully be available at a low and affordable cost. Based on dialogues and intensive reviews, the Bill and Melinda Gates Foundation has funded the project for a period of five years with plans for expansion in Bangladesh and with hopes that the vaccine will be available regionally and in cholera prone areas.

Innovative ideas and projects have evolved on host and bacterial genetics from these vaccine studies in collaboration with the University of Gothenburg and the Wellcome Trust Sanger Institute. These studies will hopefully lead to better understanding of factors related to ‘take rate’ or failure to protection by oral vaccination.

Studies on enteric diseases have been designed based on laboratory and field based activities which together have yielded results that are world class in quality and evolved with active

collaboration with laboratories in the USA, UK and Europe. The focus has been on basic studies on the understanding of the mucosal immunology of diseases with the thrust on natural infections and its application and extension to better determining vaccine efficacy. Coupled to this is the attempt to understand the role of environmental and nutritional factors on the immune responses to natural infection and vaccines. Emphasis has been on studies of cholera, ETEC and typhoid vaccines and determining optimal methods for determining surrogate markers of protection. The hypo-responsiveness of developing country children to oral vaccines is also an area of interest.Study of genetics and genomics of host and pathogens has been important in the attempt to understand infectious diseases, population based predisposition, metagenomics and changing virulence attributes of pathogens.

Working with Centres of Excellence in the UK and USA in the field of proteomics, genomics and metagenomics, key features have been identified that can be related to causes of propensity of infections. Understanding the natural course of immune responses involving innate immunity, adaptive immunity, T and B cell responses, genomic, proteomic and high throughput studies are included within her area of interest.

Another important interest is the development of diagnostic tests for detection of enteric pathogens. This has included development of setting up of a hybridoma laboratory for production of mouse monoclonal antibodies and development of diagnostic assays for detection of V. cholerae O1/O139 and other bacterial pathogens. These monoclonal antibodies are used in Bangladesh and in other countries for diagnosis and research purposes. In addition to this, the development and testing of assays for detection of S.Typhi, S. Paratyphi, V. cholerae O1/O139, ETEC and H. pylori have been developed. One of the key achievements is the development of an immunological assay for the detection of typhoid and paratyphoid fever in febrile patients. The test has been found to be extremely important for the early detection and diagnosis of typhoid fever, a disease which has increased in incidence in Bangladesh and in the region over the last few years. This test known as the TPTest (Typhoid Paratyphoid Test) is used as a diagnostic test in the routine diagnostic laboratories as well as in nationwide typhoid surveillance being carried out with the government of Bangladesh. The conversion of this test to rapid diagnostics.

In addition to the research carried out at the icddr,b, Dr Qadri has set up the “Institute for developing science and health initiatives” (ideSHi; www.ideshi.org) in Bangladesh where national and international experts from Bangladesh and globally form the Board and advisory group. This initiative was set up by utilizing the Christophe & Rodolphe Merieux Grand Prize (€500,000) which she received from the French Academy of Sciences in 2012. The prize was given to her in 2012 as recognition of her contribution to infectious disease research. The primary focus at ideSHi, instituted at the Institute of Public Health within the Centre for Medical Biotechnology of the government of Bangladesh is on the development of science and technology in Bangladesh and encouraging scientists to work in the field of biological sciences by using the facilities offered by ideSHi and icddr,b laboratories and through networking with other institutions in the government and private sector. The focus of work relates to the detection of genetic disorders in young children but also other age groups in the country. For the first time efforts are being made in Bangladesh through ideSHi to diagnose inborn errors of metabolism and other genetic disorders to help in treatment so as to prevent irreversible damage, morbidity and mortality. was also used to further enrich the ideSHi laboratory with equipment. This laboratory has excellent scientists, some of whom have recently relocated to the country to join the institution. International interns and scientists work to contribute and make ideSHi’s mission successful for further enhancing biomedical science. A large part of Dr. Qadri’s career has been focused in developing leaders in the field of infectious disease research from different disciplines and institutions. She has inspired many young scientists through her teaching and research activities. Her penchant for mentoring can be seen in her laboratory and field sites where aspiring fellows from both local and international universities join her team. To date, she has trained and mentored over 200 postgraduate students from Bangladesh and the region. In addition she has been involved in mentoring PhD students from the University of Gothenburg and Japan and Bangladesh. As a result of training programs and training grants received from the NIH (USA), Sida (Sweden) and European Union (Pasteur Institute, France), a network of international and national researchers have been developed. From the NIH funded D43 Fogarty and related grants, 18 MSc and 8 postdocs have been successfully completed training to attain higher levels of prominence in their field including faculty positions globally.

In summary, understanding the genetic and immunological basis of infectious diseases, genetic disorders as well as by technology transfer of research tools, vaccines and diagnostics is her aim and aspiration so as to benefit developing countries. As member of the High Level Panel of the UN Technology Bank which has been set up recently, this role is actively being pursued.
